UV radiation can trigger an allergic reaction leading to skin damage. The purpose of this study was to investigate the healing potential and wound healing. mechanism of Caulerpa racemose extract (CE) and Caulerpa racemose polysaccharides (CP) for wound healing after UV (3.87 mJ/cm2) irradiation. In cell culture, C2C12 cells, a mouse fiber cell line, and RAW264.8 cells, a mouse monocytic cell line, were cultured to evaluate the effect of Caulerpa racemose on wound healing. The experiment were divided into six groups, including control group, Caulerpa racemose extract treatment group, Caulerpa racemose polysaccharides treatment group, UV treatment group, UV plus Caulerpa racemose extract treatment group and UV plus Caulerpa racemose polysaccharide treatment group.
The results showed that the extract of the Caulerpa racemose and the polysaccharides could stimulate the repair of the wound. It was confirmed that the stimulation of the extract could promote the Monocyte chemoattractant protein-1 (MCP-1) expression of monocyte. At the same time extract can stimulate cell stagnation in the G2/M phase and promote cell mitosis. Using HPLC assay, the results indicated that the extract contains gallic acid. Our results suggest that Caulerpa racemose can decrease the damage of cells treated with UV irradiation possibly via stimulating cell migration rate to achieve accelerated wound healing effect.
