Evaluation of biological effects of various Houttuynia cordata extracts / 評估多種魚腥草萃取物的生物效應

碩士 / 元培醫事科技大學 / 生物科技暨製藥技術系碩士班 / 105 / Houttuynia cordata, a Saururaceae herbaceous plant, has been applied as a traditional medicine to improve immune responses and to exert anti-viral, anti-bacterial and anti-inflammatory effects.Phytochemicals in Houttuyniacordata can be divided into essential oils, polyphenols, flavonoids, alkaloids and other substances.It is known that Houttuyniacordatacontains numerous major active ingredients, such asdeCanoy acetaldehyde, lauric aldehyde, α-pinene, linlool, etc. However, different extraction methods would lead to the significant variation inactive ingredients within the product development. Using different extraction methods, the objective of this study was to compare biological activities of various Houttuynia cordata extracts.Stems and leaves of dried plants were finely ground, and then extracted with water at room temperature, 50 ºC or 70 ºC, or with 70% ethanol, ethyl acetate or chloroform at room temperature for 24h.The extracts were filtrated, vacuum-concentrated, lyophilized and stored at -20ºC.Results showed that the amount (1.58 g) of water extract from 10g ofHouttuyniacordata powder was highest, followed by those ofalcohol extract (1.27 g), chloroform extract (0.28 g) and ethyl acetate extract (0.27 g).Furthermore, the weight of the water extract isincreased as the temperature increases.Among various Houttuyniacordata extracts,the ethanol extract contained the highest amount of total polyphenols (1155 uggallic acid equivalent/g),however, the ethyl acetate extract contained the least (358 uggallic acid equivalent/g). The amounts of total flavonoidsextracted from organic solvents(813~1142 ug catechin equivalent/g) were higher than thosefrom three water extracts(~250 ug catechin equivalent/g), and the chloroform extract was the most (1142 ug catechin equivalent/g).When fatty acid compositions of the Houttuyniacordata sample were analyzed, it contained palmitic acid (C16: 0) (30.1%), α-Linolenic acid (C18: 3n-3) (21%), Oleic acid (C18: 1n-9) (15.6%), Linoleic acid (C18: 2n-6) (14.8%), Stearic acid (C18: 0) (9.3%) and others.When examined antioxidant properties, the ethanol extractexerted the strongest DPPH scavenging activity(80%) and the reduction capacity. However, the chloroform extract had the least abilities to scavenge DPPH free radicals (40%) and perform the reduction capacity.No anti-bacterial effect of Houttuyniacordataextracts was observed when using methods of disk diffusion method and minimal inhibition concentration.Since the addition of Houttuyniacordataextract at concentrations above 100 ug/ml resulted incytotoxic effect on murine RAW264.7 macrophages, a fixed concentration (50 μg/ml)of various extract was used to determine nitric oxide (NO), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in following cell culture studies.Among various extracts, the ethanol extract inhibited the production ofNO (by 28 %), TNF-a(by 27 %) and IL-6 (by 72 %).Our results showed that Houttuyniacordata extracts exerted the anti-inflammatory properties by inhibiting the production of NO, TNF-α and IL-6.In summary, active polyphenols, flavonoids and other substancesextracted from Houttuyniacordataexert antioxidant and anti-inflammatory properties, and these active compounds can be used as the development of future biopharmaceutical products.
