Oligo-fucoidan Improved Unbalance The Th1/Th2 and Treg/Th17 Ratios in Asthmatic Patients: an ex vivo study / 小分子褐藻醣膠改善氣喘患者T細胞亞型比例的不平衡—擬體內試驗

碩士 / 弘光科技大學 / 營養醫學研究所 / 106 / Asthma has been a chronic disease with a high incidence. There are many studies that have confirmed that the proportion of people suffering from asthma in recent decades has been rising. In the role of cytology, the imbalance between the type 1 T helper cells and type 2 T helper cells and that of regulatory T cells and type 17 helper T cells has become a pathogenic mechanism for asthma. At present, the treatment of asthma is mainly to reduce the inflammation of the respiratory tract, and some nutritional supplements with anti-inflammation activity seem to have some benefit in the respiratory tract and become an adjunct to the treatment of asthma. The aim of this study was to investigate the effect of the oligo-fucoidan for T cell subtypes and cytokines. Peripheral blood mononuclear cells (PBMCs) of 30 asthmatic patients and 15 non-asthmatic patients were collected and divided into 3 groups: control group, oligo-fucoidan 100 μg/ml group and oligo-fucoidan 100 μg/ml group. The cells were collected at 48 hours for cell surface markers and antibody binding to intracellular cytokines. The proportion of T cells were analyzed by flow cytometry, and cytokine levels were measured by enzyme immunoassay. In the asthmatic patient group, we found that the administration of oligo-fucoidan, either 100 μg/ml or 500 μg/ml, significantly increased the concentration of IFN-γ, and the performance of Treg cells significantly increased. Not only the ratio of IFN-γ to IL-4 was increased after the administration of oligo-fucoidan, but also the ratio of Treg cell to TH17 showed a tendency toward Treg cell. This study expoled that oligo-fucoidan can improve the inflammation of respiratory tract in asthma patients and may be an adjunct to asthma treatment.
