this study, the ayu roe hydrolysate (AH) displayed blood pressure lowering
effect in ACE inhibition (in vitro) and animal model (in vivo) experiments.
The ultrafiltered ayu roe hydrolysate (UAH) showed ACE inhibition of 59.95%. Both of AH and UAH demonstrated inhibitory activities of plasmid DNA oxidative damage induced by H2O2. Fraction A from UAH purified by FPLC with anion exchange column (DEAE) exhibited the highest ACE inhibitory activity among all the fractions. Furthermore, fraction A-IV, its molecular weight is between 1,400 and 204 Da, obtained from A fraction purified by Sephadex G-25 column showed better ACE inhibitory activity and the inhibition (%) of fraction A-IV was 52.37%. In addition, cell assay exhibited that UAHs below 10 mg/mL were not toxic to cells such as OC-2, CAL-27, Ca9-22, OECM-1, HSC-3 and HGF-1. Flow
cytometry analysis of Ca9-22 showed that UAH above 20 mg/mL had a retadation effect on the cell cycle Sub G1. Moreover, it demonstrated dosedependent effects. A-IV-4 fraction, a purified peptide containing 5 amino acids, obtained from A-IV by HPLC with C18 column had been sequenced. Its molecular weight is about 660 Da.
