碩士 / 國立高雄海洋科技大學 / 海洋環境工程研究所 / 106 / This study aimed to establish a key to identify the major families of larval fishes in the coastal waters of Yunlin, Taiwan. A total of 92 families of larval fishes were identified by microscopic examination from specimens collected between 2001 and 2016. Among them, 23 families were more common in terms of abundance and occurrence rate, including Engraulidae, Clupeidae, Sillaginidae, Sciaenidae, Sparidae….etc. Based on their morphological characteristics, an identification key for these dominant families, particularly postflexion larvae was established. This would provide a better basis to the knowledge of relevant larval fish studies in the adjacent waters.
DNA sequencing combined with Multiplex COI haplotype-specific PCR(MHS-PCR) were used to identify the species for the most abundant family, Engraulidae, due to its small body size and extremely similarity in morphology among species especially during larval stages. Following the results of DNA sequence analysis, the Engraulidae species composition and its spatiotemporal distributions were investigated. Random sampling of 576 Engraulidae larvae from different stations and different seasons were rapid DNA screened by MHS-PCR for species identification, and results suggested that there may have 3 genera and 5 species of Engraulidae in Yunlin area. These Engraulidae includes Stolephorus commersonnii(59%), Thryssa kammalensis(32.1%), Encrasicholina heteroloba (3.8%), Thryssa dussumieri (0.7%) and Thryssa hamiltonii (0.2%), respectively.
From the seasonal changes of Engraulidae, S. commersonnii can be found in all seasons. We can speculate that S. commersonnii may spawn throughout the year; while T. kammalensis can be found only from first to third seasons and the rest species appear less frequently. The occurrence difference between stations are less obvious.
