Purification and characterization of Chromobacterium aquaticum produced bacteriocin-like substances / 水生色桿菌生產之類細菌素的純化與特性分析

碩士 / 國立屏東科技大學 / 生物科技系所 / 106 / Bacteriocins is an antibacterial protein or peptide that synthesized by bacterial ribosome. Antibacterial activity of bacteriocins have been demonstrated by their bacteriostatic and bactericidal activity against diverse microbes including bacteria and fungi. The use of microbes-produced bacteriocin as biocontrol agent to prevent disease infection of improve quality of aquatic environment is an emerging method in the aquaculture. In this study, a bacteriocin-like substances producing bacterium was isolated from lake. According to the sequence and analysis of 16s rDNA and biochemistry characterization, the bacterium is belonged to Chromobacterium aquaticum, and named as Chromobacterium aquaticum in our lab. Partial purification of bacteriocin-like substances from C. aquaticum was performed by ammonium sulfate precipitation. The antimicrobial activity of partial purified product against Escherichia coli、Streptococcus agalactiae and Vibro vulnificus are demonstrated by agar well diffusion method. C. aquaticum have catalase, oxidase and motility rod-shaped Gram-negative bacteria, suitable for culture temperature of 28 ℃. Identification of broth from C. aquaticum, it has been found that C. aquaticum has beta-lactamase and than has an antibiotic-resistance. In the future, we expected the bacteriocin-like substances from C. aquaticum can provide as an effective biocontrol agent for disease infection in the aquaculture.
