碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 106 / The performance of Kappaphycus alvarezii in land tank culture has not been well studied. Therefore, the present study sought to determine the performance of K. alvarezii cultivated in a tank culture system. Several algal cultivation biotechnologies were also applied to stimulate the growth and the cold-tolerance of K. alvarezii in tank culture. We found that optimal biomass was obtained when thalli were cut to an initial weight of 50 g at the start of cultivation, and after 55 days of summertime cultivation, the highest total mean biomass was 1038.5±12.5 g. Experiments were also performed to test the growth promotion and cold-temperature tolerance effects of the application of low-dose ionizing radiation. These ionizing radiation experiments found that growth of the F1 generation of K. alvarezii was promoted substantially under 30 Gy of 60Co γ-ray radiation, with an average increase of algal biomass of 61.39%. The radiation treated K. alvarezii had an enhanced total polysaccharide composition of 22.91% (dry weight) compared to the untreated control, which had a polysaccharide composition of 17.87%, this is a 1.28 times greater total polysaccharide quantity in the radiation treated algae compared to the untreated control. We also found that the irradiated algae had improved tolerance to cold temperatures(as low as 13℃) in the F2 generation in the winter. In addition, growth stimulation experiments were conducted using varied concentrations of Sargassum horneri extract solution. We found that the greatest total mean biomass of 997.0±79.5 g, which was 2.25 times that of the untreated controls, was achieved when K. alvarezii was treated with higher concentrations of S. horneri extract solution (500~1000 ppm) and longer dipping times (60~180min).
