Investigating the effect of components of Antrodia camphorata on attenuating Alzheimer's disease pathology in APP/PS1 transgenic mice / 探討牛樟芝成份減輕APP/PS1基因轉殖小鼠之阿茲海默氏症相關症狀的效果

碩士 / 國立陽明大學 / 生物藥學研究所 / 106 / Alzheimer's disease (AD) is a neurodegenerative disease and the most common type of dementia. As the disease progresses, patients may experience memory degradation, difficulty in self-care, abnormal image and spatial judgment. Difficulties in thinking and decision-making, emotional instability and abnormal behavior, and thus loss of motivation. The main cause of AD is the abnormal accumulation of beta-amyloid protein (Aβ) in the brain to form plaque. The aim of this study is to investigate the potential of the components of Antrodia camphorota, including Ergostatrien-3β-ol (EK100) and Antrodin C for the treatment of AD. Besides, we use a structural analogs of EK100, Ergosterol, and nonsteroidal anti-inflammatory drugs (NSAID), Ibuprofen, as control groups. The results showed that the five-month-old APP/PS1 transgenic mice fed with EK100, Antrodin C or Ibuprofen for one month significantly improved the nesting behavior. In neuropathology, it was observed that EK100 and Antrodin C reduced the number of plaques in the cerebral cortex and hippocampus, and also reduced the number of plaque-associated glial clusters. In addition, EK100 and Ibuprofen also inhibited Non-cluster glial activation in Hippocampus and Antrodin C inhibited Non-cluster microglial activation. On the other hand, Ergosterol had no significant effect in the above experimental results. In terms of the antioxidant mechanism of Nrf2, we found that the expression of Nrf2 in the cerebral cortex of APP/PS1 transgenic mice was significantly higher than that of Wile type mice. After feeding EK100, Antrodin C or Ibuprofen, the expression of Nrf2 was significantly decreased. Therefore, we believe that EK100 and Antrodin C may reduce plaque deposition through undetermined mechanisms, thereby inhibiting the expression of Nrf2.
