The Extraction and Separation of Polysaccharides from Brown Algae Saccharina japonica / 海帶中海帶多醣萃取與分離

碩士 / 國立高雄應用科技大學 / 化學工程與材料工程系博碩士班 / 107 / The polysaccharides from Saccharina japonica have many interesting functional properties, such as antioxidant activity, anticoagulant, immunomodulation, antithrombotic, anti-proliferative activities and anticancer. In this study, the crude polysaccharide was extracted by different solvents at 100 ˚C for 120 min from Saccharina japonica. Various conditions of solvents (water, 1% sodium hydroxide, 3% citric acid, 3% sodium hydrogen carbonate, 3% sodium carbonate) were assessed for their extraction efficiency. Alginate was separated by calcium chloride from the crude polysaccharide. The best yield of alginate was obtained from 3% sodium carbonate. Remaining separated liquid hydrolysates were precipitated by different conditions of methanol (80% methanol, 60% methanol, and 30% methanol). The best yield of purified polysaccharide was obtained from 1% sodium hydroxide extraction (and following 80% methanol precipitation). All products were analyzed by FT-IR. The results showed that the extracted and separated polysaccharides individually features as the alginate and fucoidan, and that the hot process using 3% Na2CO3(aq) or 1% NaOH(aq) as the extraction solvents is economical
