碩士 / 國立東華大學 / 海洋生物研究所 / 107 / In the study on the chemical constituents of the cultured soft coral, Briareum violaceum have led to the isolation of twenty briarane-type diterpenoids, including seven new compounds, briaviolides RX (17), as well as thirteen know compounds, 2β-acetoxy-2-(debutyryloxy)stecholid E (8), 9-deacetylbriareolide H (9), briaexcavatolides A (10), P (11), and T (12), briareolide B (13), brianthein W (14), briaviolide O (15), dollfusilin A (16), excavatoid I (17), excavatolides B (18) and E (19), and stecholide C acetate (20). The structures of metabolites 120 were established by the spectroscopic methods and comparison of the spectroscopic data with known related compounds. The absolute configurations of briaranes 14, 18, and 19, were determined by single-crystal X-ray diffraction analysis.
In terms of anti-inflammatory test, the extract of B. violaceum showed inhibitory effects on the generation of superoxide anion and the release of elastase (inhibition rates = 72.24 ± 11.33 % and 59.27 ± 5.30 %, respectively) by human neutrophiles at a concentration of 10 g/mL. Briaviolide S (2) showed a significant inhibitory effect on the generation of superoxide anion (IC50 = 5.37 ±1.10 M).
