碩士 / 國立中山大學 / 海洋生物科技暨資源學系研究所 / 107 / In this study, the effect of the light intensity and magnesium (Mg) on the cell growth, lutein content, production and productivity of a microalga Chlorella sp. 5-3 has been examed. Using the normal BG-11 medium culture,an exposure to 50 E m-2 s-1 showed the highest lutein content (1.44 ± 0.04 mg/g), exposure to 150 E m-2 s-1 had the highest lutein production (2.37 ± 0.11 mg/L) while 350 E m-2 s-1 illumination exhibited the highest lutein productivity (0.53 mg/L/d). In response to low (0.0375 g/L) and normal (0.075 g/L) Mg content, the highest lutein content (2.81 ± 0.11 mg/g) and the highest lutein production (1.35 ± 0.02 mg/L) were obtained under low Mg content and 50 E m-2 s-1 illumination for 3 days. Next , 7-days time course experiment using low Mg content under low and high light intensities was compared.For low Mg ,the highest lutein content (2.19 ± 0.09 mg/g) was obtained at day 2, and the highest lutein production (1.63 ± 0.14 mg/L) and productivity (0.54 ± 0.05 mg/L/d) were appeared at day3. For 350 E m-2 s-1, the highest lutein content (1.07 ± 0.05 mg/g) was obtained at day 1, the highest lutein production (0.97 ± 0.18 mg/L) at day 3 , while the highest lutein productivity(0.44 ± 0.02 mg/L/d) was obtained at day 2. The result showed enhanced light intensity cannot increase lutein content.In conclusion the low light intensity together with low Mg content were the optimal culture conditions of lutein production in a microalga Chlorella sp. 5-3.
