碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 107 / The objective of this study is to investigate the effects of Sarcodia suiae polysaccharides by different extraction conditions, and analyze the compositions and assessment the activity of antioxidant of the extracts by membrane filtration and ethanol precipitation, and then investigate the effects of hemocyte activity and non-specific immunity of Litopenaeus vannamei on Sarcodia suiae.
The higher proportion of carbohydrates was found on Sarcodia suiae algae powder. The optimal sugar extraction rate was obtained with an algae-water ratio of 1:25, the temperature of 100 ° C and an extraction time of 1 hour, but the long time and high temperature extraction condition was found the thermal degradation of polysaccharides.
The five kinds of extracts HE, CPE, MPE, SPE, and EE was obtained from the filtration of Sarcodia suiae extract on hot water extraction by ultrafiltration and ethanol precipitation. In this study, the Sarcodia suiae polysaccharide was found the rich sulfate, and the stability of sulfate was not affected by ethanol precipitation. Among the antioxidant substances, we found that the components in the sample may change after being precipitated by ethanol. These changes are also responsible for the difference in their antioxidant activity.
The white shrimp hemocytes safety test and non-specific immunoassay of HE\CPE\MPE\SPE and EE extracts, the results was found the five extracts are non-toxicity to white shrimp hemocytes, while the EE was inhibition cell activity slightly at higher concentrations. In the non-specific immunization experiment, the best immune performance at the concentration of HE/CPE/MPE/SPE and EE are 20、1、1、0.01~1 and 0.1~5mg/mL respectively.
Based on these results, this study suggested the extracts of Sarcodia suiae polysaccharide has the potential to be a natural antioxidant and immunomodulatory.
