The aim of this study was investigating the growth characteristics of the green macroalgae Caulerpa lentillifera under different spectrum environments. In this study, C. lentillifera were cultured under six light sources : blue (B), red (R), white (W) blue, 1:1 (1B1R), 1:2 (1B2R), 1:5 (1B5R) blue and red light combination light-emitting diodes (LED), three different light intensity of 50, 100, 150 μmol photos•m-2•s-1, and three different photoperiods of 8L/16D, 12L/12D, 16L/8D (light/dark in 24 hours). The weight gain percentage, pigment content (chlorophyll a, chlorophyll b, and β-carotene), antioxidant activity (ABTS antioxidant activity and reducing power), and phytoene desaturase (PDS) gene expression in different light spectrums were investigated. In order to correctly sequence PDS gene of C. lentillifera and to design highly efficient primers for quantitative polymerase chain reaction (qPCR), phylogenetic tree and vector transfer was employed to identify genes belonging to the C. lentillifera. The results showed that weight gain percentage significantly increased under 1B5R light. The β-carotene content, antioxidant activity and PDS gene expression of C. lentillifera benefitted from blue light, whereas it has disadvantageous effects on weight gain percentage. It was demonstrated that blue light was a stress condition to C. lentillifera, and it needed to increase antioxidant accumulation by up-regulated PDS gene to survive under blue light. When light intensity was 100 μmol photos•m-2•s-1 and 12L/12D photoperiod, the weight gain percentage higher to the others. However, the β-carotene content showed significant increase under higher light intensity and longer photoperiod. The weight gain percentage of 1B1R and 1B5R were higher than the 1B2R. The results suggested that C. lentillifera has special react under specific ratio of blue and red light combination.
