Inhibitory effects of Laminaria japonica fermented product on low density lipoprotein oxidation and foam cell formation / 海帶發酵物抑制低密度脂蛋白的氧化和泡沫細胞形成之研究

碩士 / 國立臺灣海洋大學 / 生命科學暨生物科技學系 / 107 / In this experiment, we investigated the difference between Laminaria japonica water extract (JWE) and Laminaria japonica fermented product (JFP) on functional groups, inhibition of low density lipoprotein (LDL) oxidation and inhibition of foam cell formation using the model of oxidized low density lipoprotein (oxLDL)-induced macrophage. Results exhibited that JFP had better inhibition of copper ion-induced LDL oxidation than JWE. In cell viability experiment, it was showed that JFP increased cell viabilities in the concentration range of 10 to 150 μg/ml, while JWE inhibited cell viabilities. Then we used oil red O staining to observe the effect of JFP and JWE on foam cell formation, it was showed that JFP had better inhibition of form cell formation than JWE. From gene expression analysis, we found that JFP can inhibit the expression of oxLDL scavenger CD36 and SR-A, and it can also inhibit the expression of proinflammatory gene TLR4 and TLR6. Then, protein expression analysis showed that JFP and JWE both could significantly inhibit the expression of CD36, SR-A, TLR4 and TLR6 at a concentration of 80 μg/ml, and the ability of JFP to inhibit protein expression was better than that of JWE. Observed in the Fourier transform infrared spectrometer (FTIR) data that JFP has more sulfated groups, carboxylic acids, aldehydes, ethers and other functional groups than JWE. From the results speculated that JFP inhibits the mechanism of atherosclerosis, mainly through reducing LDL oxidation, inhibiting foam cell formation and blocking the transmission of subsequent inflammatory responses.
