Orientador: Maria Denise Lopes / Resumo: Neste estudo foram utilizados 20 cães, sem raça definida, de fertilidade desconhecida, com pesos variando entre 15 e 20 Kg, sorologicamente negativos para leptospirose e brucelose canina. Após um período de condicionamento, foram colhidos seis ejaculados de cada cão, por manipulação digital do pênis, sendo três antes da deferentectomia e três após. O sêmen foi imediatamente analisado após a colheita, considerando os seguintes parâmetros: volume, cor, motilidade progressiva, vigor espermático, porcentagem de espermatozóides morfologicamente normais, pH, integridade de membrana (teste hiposmótico e coloração com sondas fluorescentes). Foram determinadas as concentrações de proteínas totais, cloretos totais, cálcio, potássio e sódio. A SDS-PAGE foi realizada utilizando-se duas concentrações de poliacrilamida, 13% e 22%, no gel de separação. Os géis foram corados com coomassie azul brilhante R-250, suas imagens digitalizadas e avaliadas num programa analisador de imagens, onde foram estimados os pesos moleculares e a densidade óptica integrada de cada banda, para cada amostra. Uma correlação negativa foi verificada entre o potássio e a motilidade progressiva, vigor espermático, resultados do teste hiposmótico e porcentagem de espermatozóides com integridade de membrana na coloração com sondas fluorescentes. Apenas o cálcio mostrou-se significativamente mais elevado após a deferentectomia. Foi verificado um total de 37 bandas antes da deferentectomia e 35 após. Antes da deferentectomia, encontrou-se uma correlação positiva e significativa entre duas bandas protéicas, B4 (67 kDa) e a B5 (58,6 kDa) e a motilidade progressiva, vigor espermático, porcentagem de espermatozóides morfologicamente normais, resultados do teste hiposmótico e porcentagem de espermatozóides com integridade de membrana na coloração com sondas fluorescentes... (Resumo completo, clicar acesso eletrônico abaixo). / Abstract: In this study 20 mixed breed dogs (15 to 20 Kg of body weight) of unknown fertility were used. The dogs were negative in leptospyrosis and brucelosis tests. After a conditioning time, were collected six ejaculated of each dog by digital manipulation of penis, being three before and three after deferentectomy. Immediately, the semen was analyzed considering the following parameters: volume, color, spermatic motility, vigor, morphological forms of the spermatozoa, pH and membrane integrity (hypoosmotic test and fluorescent stain). The concentrations of total proteins, total chlorides, calcium, potassium and sodium were determined. The SDS-PAGE of each seminal plasma sample was carried out using two polyacrylamide concentrations, 13% and 22%, in the separation gel. The gels were stained with coomassie blue brilliant R-250, the images were digitalized and evaluated in a analyzer software, where the molecular weights were estimated, in accordance with a standard applied in each gel. The integrated optic density of each band for each sample was determined. A negative correlation was verified between the potassium and the spermatic motility, vigor, results of the hypoosmotic test and percentage of spermatozoa with integrity of membrane in the fluorescent stain. The calcium concentrations after of deferentectomy were significantly higher than before. A total of 37 bands before deferentectomy and 35 after were observed. Before the deferentectomy, a positive correlation was verified between two bands, B4 (67 kDa) and the B5 (58,6 kDa), and the spermatic motility, vigor, percentage of normal spermatozoa, results of the hypoosmotic test and percentage of spermatozoa with integrity of membrane in the stain fluorescent. Three bands were significantly different before and after the deferentectomy (B3, of 73,5 kDa, B29, of 7,8 kDa and B27, 3,6 kDa).
