Orientador: Nereu Carlos Prestes / Resumo: O presente projeto pretendeu estudar o ciclo hormonal e o metabolismo de cálcio das cadelas durante a gestação e puerpério e testar a eficiência do método da quimioluminescência como uma alternativa ao RIA. Foram utilizadas 10 cadelas adultas com tamanho e peso semelhantes, mantidas sob as mesmas condições de manejo e alimentadas com ração de boa qualidade, sem qualquer suplementação até os 40 dias de gestação, após isto a ração foi substituída por ração de filhote até o final da lactação. As colheitas de sangue foram realizadas durante as fases gestacional e puerperal, nos momentos: M0 (início do diestro citológico), M1 (20 dias pós-M0), M2 (40 dias pós-M0), M3 (momento do parto), M4 (15 dias pósparto), M5 (45 dias pós-parto) e M6 (30 dias pós-desmame). O PTH, osteocalcina, calcitonina, progesterona, estradiol, T3 total, T4 total, T4 livre e calcitriol foram analisados pelo radioimunoensaio; T4 canino, T3 total, T3 livre, T4 total e T4 livre foram analisados pela quimioluminescência e o cálcio, fósforo, proteína total, albumina e fosfatase alcalina foram analisados pelo método colorimétrico. A osteocalcina e calcitonina foram testadas pelo método do RIA e não foram observadas diferenças entre os momentos. O T3 Livre testado pelo método de quimioluminescência e apresentou todos os valores abaixo do limite de detecção. As demais variáveis apresentaram diferenças significativas entre os momentos. Não foram observadas correlações que demonstrem envolvimento dos hormônios dosados com o metabolismo do cálcio e fósforo. Não houve correlação entre o número de filhotes e as demais variáveis estudadas; os resultados demonstram não ter havido tendência a hipocalcemia e correção hormonal; o estudo de correlações não demonstrou um padrão de acontecimento entre as diversas variáveis, principalmente entre o cálcio e as demais variáveis... (Resumo completo, clicar acesso eletrônico abaixo). / Abstract: The present project intended to study hormone cycle and calcium metabolism of bitches during gestation and the puerperal period and to test the efficiency of chemoluminescent immune assay as an alternative to RIA. Ten adult bitches of similar size and weight, maintained under the same environmental conditions and fed with high quality dry food without any supplementation until the fortieth day of gestation, then changed to puppy dry food until the end of lactation. Blood samples were collected during gestation and the puerperal period, on moments M0 (beginning of cytologic diestrus), M1 (20 days after M0), M2 (40 days after M0), M3 (at the time of parturition), M4 (15 days after parturition), M5 (45 days after parturition) and M6 (30 days after weaning). PTH, osteocalcin, calcitonin, progesterone, estradiol, total T3, total T4, free T4 and calcitriol were analyzed by RIA; canine T4, total T3, free T3, total T4 and free T4 were analyzed by chemoluminescence and calcium, phosphorus, total protein albumin and alkaline phosphatase were analyzed by colorimetric method. There was no difference between moments on values of osteocalcin and calcitonin. All values of free T3 were below the limit of detection of the test. The other variables studied presented no difference between moments. There was no correlation demonstrating the involvement of tested hormones on calcium and phosphorus metabolism. There was no correlation between litter size and the other parameters studied; the results showed no tendency to hypocalcaemia and hormone correction; the study of correlations did not suggest a pattern of happening among variables, specially between calcium and the other variables; osteocalcin is more specific as bone maker that alkaline phosphatase; nutritional correction may have been the responsible for the quiescence of minerals and related hormones, the same occurring to the values of total protein...
