Orientador: Frederico Ozanan Papa / Resumo: O presente trabalho teve os objetivos: avaliar laboratorialmente o sêmen congelado de asininos através do Hamilton Thorne (HTMA); determinar os indices de fertilidade do sêmen criopreservado em jumentas e éguas e verificar a resposta uterina das jumentas perante o sêmen "in-natura", diluente e congelado. No experimento 1 as amostras de sêmen, foram criopreservados em dois criotubos: meio macrotubo - 2,5 mL (M) ou palheta - 0,5 mL (P) com diluente MP50 e dez formulações de crioprotetores: 1= 2% DMSO e 2% DF; 2= 3% DF; 3= 2% MF e 2% de Glicerol; 4= 2% dimetil acetamida e 2% de glicerol; 5= 3% metil formamida; 6= 3% de dimetil acetamida; 7= 3% glicerol e 2% dimetil acetamida; 8= (Specific Patogen Free) 3% glicerol e 2% dimetil formamida; 9= 3% Dimetil sulfóxido e 2% glicerol; 10= 1% etilieno glicol + 3% dimetil sulfóxido e 1% dimetil formamida. As comparações realizadas entre os 2 tipos de envase, meio macrotubo e palhetas, não demonstraram superioridade (P>0,05) entre si, não havendo interação com as formulações utilizadas, nos parâmetros analisados. Para as características de motilidade total foi verificado que os tratamentos M4, P4 e P6 foram superiores ao M7 e P5 (P<0,05) a avaliação da motilidade progressiva mostrou qua a formulação M4 se distingüiu exclusivamente do M7 (P<0,05). O estudo da linearidade (LIN), apontou diferença exclusivamente entre os tratamentos M3 e M6 (P<0,05). Os valores obtidos de VCL (velocidade real), de VAP (velocidade do trajeto espermático) e de VSL (velocidade retílinea) proporcionaram resultados semelhantes entre os tratamentos (P>0,05). Na avaliação da membrana plasmática pela fluorescência, foi encontrado maior valor para o tratamento P6, que diferiu significativamente de P5 e M7, além disso o tratamento P4 se diferenciou de P5 (P<0,05)... (Resumo completo, clicar acesso eletrônico abaixo). / Abstract: The current paper aimed at conducting laboratoty tests in order to evaluate the frozen semen of the asinines, determining the fertility rate of semen cryopreserved in female donkeys and mares and also verifying the uterine response of female donkeys to the "in natura", MP50 extender and frozen semen. In experiment 1, the semen samples were cryopreserved in two cryotubes: half macro tube 2.5 mL (M) or pallets - 0.5 mL (P) with MP50 diluting and ten formulations of cryoprotectors: 1= 2% DMSO and 2% DF; 2= 3% DF; 3= 2% MF and 2% of glycerol; 4= 2% dimethyl acetamide and 2% of glycerol; 5= 3% methyl formamide; 6= 3% of dimethyl acetamide; 7= 3% glycerol and 2% dimethyl acetamide; 8= (Specific Patogen Free) 3% glycerol and 2% dimethyl formamide; 9= 3% dimethyl sulfoxide and 2% glycerol; 10= 1% ethylene glycol + 3% dimethyl sulfoxide and 1% dimethyl formamide. The comparisons made between the two types of packing, did not show superiority (P>0.05) between themselves, and there was no interaction with the used formulations within the analyzed parameters. Regarding the characteristic total motility, it has been verified that the M4, P4 and P6 treatments were superior to the M7 and P5 (P<0,05). The evaluation of the progressive motility showed that the M4 formulation differed exclusively from the M7 (P<0,05). The linearity study demonstrated difference particularly between the M3 and M6 treatments (P<0,05). The values of real velocity, sperm direction velocity and linear velocity did not show similar results between the treatments (P>0,05). Concerning the evaluation of the plasma membrane by fluorescence, a higher value was found for the P6 treatment, which differed significantly from the P5 and M7.
