Radiosensitization Strategies for Enhanced E-beam Irradiation Treatment of Fresh Produce

Fresh produce is increasingly implicated in outbreaks of foodborne illness. Internalization of bacterial pathogens into produce is of particular concern as internalized pathogens are unlikely to be removed by surface sanitizers. It is therefore necessary to develop treatments that will reduce their prevalence and numbers on fresh produce. Irradiation is a penetrating nonthermal treatment that effectively eliminates bacteria. Irradiated baby spinach leaves up to 1.0 kGy showed negligible (P>0.05) changes in color, texture, vitamin C, total carotenoids, and chlorophyll content compared to non-irradiated controls throughout storage (15 days at 4oC). This research also shows that irradiation effectively reduces viable Escherichia coli cells internalized in lettuce, and that decontamination is not influenced (P>0.05) by lettuce variety. Irradiation effectively reduced the population of internalized pathogens in a dose-dependent manner (3-4 log reduction at 1.0 kGy). Microscopy images suggest that the contamination sites of pathogens in leafy vegetables are mainly localized on crevices and in the stomata. A careful design of the treatment (understanding dose distribution) will effectively eliminate pathogens while maintaining produce quality. The use of modified atmosphere packaging increased (P<0.05) the sensitivity of pathogens (Salmonella spp. and Listeria spp.) to irradiation in baby spinach leaves (up to 25%). Increasing concentration of oxygen increased (P<0.05) sensitivity of both microorganisms. Radiosensitization could be affected (P<0.05) by production of ozone, which increases with increasing dose-rate and oxygen concentration, and reducing temperatures. Antimicrobial effectiveness of various active compounds was determined against Salmonella spp. and Listeria spp. Inclusion complexes were prepared with antimicrobial compounds and -cyclodextrin. The effectiveness of the microencapsulated compounds was tested by spraying them on the surface of baby spinach leaves inoculated with Salmonella spp. The increase in radiosensitivity (up to 40%) varied with the antimicrobial compound. Spherical poly (DL-lactide-co-glycolide) (PLGA) nanoparticles with entrapped eugenol and trans-cinnamaldehyde were synthesized for future antimicrobial delivery applications. All loaded nanoparticles proved to be efficient in inhibiting growth of Salmonella spp. and Listeria spp. The entrapment efficiency for eugenol and trans-cinnamaldehyde was 98% and 92%, respectively. Controlled release experiments (in vitro at 37oC for 72 hrs) showed an initial burst followed by a slower release rate of the antimicrobial entrapped inside the PLGA matrix.

Opportunistic Fresh-Produce Commercialization under Two-Market Disintegration

abstract: This thesis develops a low-investment marketing strategy that allows low-to-mid level farmers extend their commercialization reach by strategically sending containers of fresh produce items to secondary markets that present temporary arbitrage opportunities. The methodology aims at identifying time windows of opportunity in which the price differential between two markets create an arbitrage opportunity for a transaction; a transaction involves buying a fresh produce item at a base market, and then shipping and selling it at secondary market price. A decision-making tool is developed that gauges the individual arbitrage opportunities and determines the specific price differential (or threshold level) that is most beneficial to the farmer under particular market conditions. For this purpose, two approaches are developed; a pragmatic approach that uses historic price information of the products in order to find the optimal price differential that maximizes earnings, and a theoretical one, which optimizes an expected profit model of the shipments to identify this optimal threshold. This thesis also develops risk management strategies that further reduce profit variability during a particular two-market transaction. In this case, financial engineering concepts are used to determine a shipment configuration strategy that minimizes the overall variability of the profits. For this, a Markowitz model is developed to determine the weight assignation of each component for a particular shipment. Based on the results of the analysis, it is deemed possible to formulate a shipment policy that not only increases the farmer's commercialization reach, but also produces profitable operations. In general, the observed rates of return under a pragmatic and theoretical approach hovered between 0.072 and 0.616 within important two-market structures. Secondly, it is demonstrated that the level of return and risk can be manipulated by varying the strictness of the shipping policy to meet the overall objectives of the decision-maker. This study examines the price differences of commonly consumed produce between farmers’ markets and grocery stores in Utah. Our first objective is to compare price differences of a basket of produce between farmers’ markets and grocery stores. We compare these price differences in terms of low-income consumer affordability and if an individual can afford a market basket of produce using a combination of Farmers’ Market Nutrition Program (FMNP) dollars and Double Up Food Bucks (DUFB) incentive dollars. Our second objective for this study is to establish the price premiums of individual produce items based on where the produce was sold, time of season, market channel (farmers’ market versus grocery store), as well as production method used (conventional versus organic). The findings from this research can inform policy makers of the affordability of farmers’ market produce and apply incentive programs more effectively. We can inform consumers of the price differences so they can maximize their food budgets. We can use the research to help producers make market strategies that can then maximize their profits.
