The classics will be reinterpreted by an introduction of new ideas, and thus bringing innovations to the academia. Liaozhai zhiyi as the masterpiece of classical Chinese novels contains multiple meanings in its text. This thesis, inspired by theories such as structuralism and intertextuality, will adopt a novel methodology, namely spatial narration in initiating a discussion. Through interactive relationships among characters and among events, this thesis reveals pluralistic meanings of text while at the same time illustrating the process of constructing implied messages. / The research subjects of this thesis are different structural relationships in Liaozhai zhiyi. And this thesis is divided into five chapters, of which the first is introduction, summarizing previous scholarly research findings of and working out the concept of spatial narration as the main methodological concept and hence establishes research premise and context. Chapter two utilizes plot and human-fox relationship in the human-fox love story as focus, aims at discussing rules of narration formulated in Liaozhai zhiyi. Chapter three mainly uses interactions between the immortals and mortals to reveal author’s entailed messages and implied aesthetic standards (or standard of appreciation) in between intercommunication and obstruction of the immortals and mortals. Chapter four breaks though mere textual analysis, adopts the perspective of intertextuality to perceive Liaozhai zhiyi’s absorption and transformation of the Story of Liu and Ruan’s meeting with fairy maiden by chance. The final chapter intersects with the setting of double lovers to discuss the aesthetic principles of parallelism observed by Pu Songling and structure of narration constructed, and therefore explicating implied messages behind structure. / Detailed summary in vernacular field only. / Detailed summary in vernacular field only. / 王玲玲. / Parallel title from English abstract. / Thesis (M.Phil.) Studying on Development and Utilization Molds of National Land--Example pf Ting Pu High-tech Industrial Park

Chieh, Lin-maw 14 July 2004 (has links)
Abstract National lands are important resources of a country as well as important tools of a government for implement of its policies. National lands are usually released in different ways and for development and utilization of different purposes from short-term lease to medium- or long-term lease; appraisal for superficies right; sale of land ownership; and from appropriation of lands to the competent authorities in charge of the relevant industries to management and operation of the lands by land authorities. Different ways of land release will bring about different policy effects and financial effects. Therefore, to attain certain goals of policies, the central government will make use of different ways to release lands for meeting different needs. In the past, when it is necessary to develop and utilize National lands for special development purposes, the public lands will become the targets of such development in most cases, and the competent government authorities in charge of the relevant industries usually play a leading role in the development. However, this is not the case in the recent ¡§Ting Pu High-tech Industrial Park Development Project¡¨ of Taipei County Government. In the project, the non-public lands became objectives of development rather than the public lands under the control of National Property Administration (NPA), Ministry of Finance. The project, through public selection procedure, recruits investors to participate in the development of the lands. As a result, three high-tech companies including Hon Hai Precision Industry Co., Ltd. entered the park and built factories there. So the project should be considered as a successful case of land release. In recent years, NPA took over some lands from the Taiwan Provincial Government and commissioned the local governments to manage those lands. In addition, NPA started to take over the national assets from various organizations and state-run enterprises. It is anticipated that there will be a great deal of state-owned non-public lands to be released by NPA at the proper time and in proper ways for development and utilizations. In this way, the state-owned lands will have the opportunities to develop their functions. This study probes into the innovative development mold of Ting Pu High-tech Industrial Park and discuss the successful factors and existing problems of the park. It is hope that this study may provide the government a better method to create the utilization value of the lands and further to promote development of the country¡¦s economy and prosperity of the society. Finally, the research submits the following suggests: (1) The government should establish a land reserve system to enhance the power and function of land distribution and develop the functions of the public lands and its role in development of society and economy. (2) Currently, there are five types of industrial lands in the country, and they compete with one another to strive for resources and sources of customers. So, an integration of those lands becomes essential. (3) An incentive system should be established to encourage the local governments to take actions that suit local circumstances and develop the industries with local features. The government departments may also take the opportunities to release more public-owned lands for development and utilization of land in order to increase the land use value and create higher economic effect. Model pro studium regulace transkripce granulocytarních genů MPO a MMP9 rozdílnými koncentracemi transkripčního faktoru PU.1 / Model for study of transcription regulation of granulocytic genes MPO and MMP9 by different levels of PU.1 transcription factor

Chramostová, Kamila January 2018 (has links)
9 Abstract Enhancers are distal cis - regulatory DNA sequences that regulate (enhance) transcription of the respective gene driven by its promoter. Enhancers are found in non-coding DNA upstream or downstream of the gene coding sequence, or in introns or coding regions that are located up to hundreds kb away from the gene. Superenhancers are newly discovered clusters of multiple enhancers that play a vital role in activating tissue-specific genes, determining cell identity and regulating differentiation. PU.1 is the transcription factor (TF) that is necessary for normal haematopoiesis, specifically for the development of myeloid and lymphoid blood lineages. Distinct levels of PU.1 induce differentiation of hematopoietic cells into different cell lineages whereby disruption of PU.1 levels leads to leukemogenesis. High PU.1 levels stimulate macrophage development, while intermediate levels stimulate the development of granulocytes. This diploma thesis seeks to contribute to addressing the interesting biological question of what are the regulatory mechanisms to ensure that granulocytic genes are activated only at the intermediate concentration of PU.1, whereas macrophage genes are activated only at its high levels. The aim of this diploma thesis was to create a series of reporter vectors carrying regulatory...

Role transkripčních faktorů PU.1 a GATA-1 v leukemické diferenciaci / The role of transcription factors PU.1 a GATA-1 during leukemia differentiation.

Burda, Pavel January 2011 (has links)
Hematopoiesis is coordinated by a complex regulatory network of transcription factors among them PU.1 (Spi1, Sfpi1) and GATA-1 represent key molecules. GATA-1 and PU.1 bind each other on DNA to block each others transcriptional programs to prevent development of undesired lineage during hematopoietic commitment. Murine erythroleukemia (MEL) cells, transformed erythroid precursors that are blocked from completing the late stages of erythroid differentiation, co-express GATA-1 and PU.1 and as my and others data document, are able to respond to molecular removal (down-regulation) of PU.1 or addition (up-regulation) of GATA-1 by inducing terminal erythroid differentiation. We provide novel evidence that downregulation of GATA-1 or upregulation of PU.1 induces incompletely differentiation into cell cycle arrested monocytic-like cells. Furthermore, PU.1- dependent transcriptome is negatively regulated by GATA-1 in MEL cells, including CCAAT/enhancer binding protein alpha (Cebpa) and Core-binding factor, beta subunit (Cbfb) that encode additional key hematopoietic transcription factors. Chromatin immunoprecipitation and reporter assays identified PU.1 motif sequences near Cebpa and Cbfb that are co-occupied by PU.1 and GATA-1 in the leukemic blasts. Furthermore, transcriptional regulation of these loci by...

Small Business Tax Policy, Informality, and Tax Evasion - Evidence from Georgia

Bruhn, Miriam, Loeprick, Jan January 2014 (has links) (PDF)
Using a panel of administrative data and regression discontinuity analysis, this paper examines how the introduction of preferential tax regimes for Georgian micro and small businesses in 2010 affects formal firm creation and tax compliance. The results show that the new tax regime for micro businesses increased the number of newly registered formal firms by 18-30 percent below the eligibility threshold during the first year of the reform, but not in subsequent years. The analysis does not find an effect of the new tax regime for small businesses on formal firm creation in any year. Policy makers are often concerned about abuse risks stemming from differentiated tax treatment of micro and small businesses. The analysis in this paper reveals reduced tax compliance in 2010 around the micro business eligibility threshold, but does not find significant evidence of reduced compliance by Georgian firms in later years. The results also do not show any significant evidence of strategic sorting around the regime eligibility thresholds. (authors' abstract) / Series: WU International Taxation Research Paper Series

Synthèse d'alcynylcarbinols fonctionnels et évaluation d'analogues en tant que pharmacophores / Functional alkynylcarbinols synthesis and their analogous evaluation as pharmacophores

Listunov, Dymytrii 22 December 2015 (has links)
Among naturally occurring secondary metabolites isolated from marine sponges the compounds with chiral propargyl alcohol unite occupy a special place. They have been shown to display the different types of biological activities, for example: antibiotic, antiviral, and more generally cytotoxic. In these compounds, the propargyl alcohol unit hereinafter as the "alkynylcarbinol" (including substituted chiral carbinol center), is the key characteristic that can have a pharmacophore fragment function. Nevertheless, no systematic study to determine structure-activity relationships in this series of molecules had been carried until recently. In order to identify a minimum pharmacophore unit and identify structure-activity relationships, systematic structural variation was therefore considered as part of this work from the archetypal structure of a natural product isolated from the sponge Cribrochalina vasculum. The first chapter is a bibliographic summary describing the synthesis and biological activity of molecules with a propargyl alcohol moiety in their structure. The second chapter describes a study of structure-activity relationship in a series of compounds from the reference molecule ((E) -icos-4-en-1-in-3-ol). The four parameter variation was studied: the nature of each of the two unsaturations, the configuration of the asymmetric center, and the length of the saturated lipid chain. The third chapter focuses on the complete study of the synthesis of lipids in two symmetrical pharmacophore heads C2 and stereochemical composition characteristics of the obtained products. The fourth chapter is devoted to the description of the synthetic products in which an acetylene C2 unit is replaced by a C4 butadiyne fragment or C3 one of allene. The fifth chapter describes the synthesis of pharmacophore dialkynylcarbinol lipids with fluorophore fragment for cell imaging and the first cell imaging tests. In the sixth chapter has combined the results of measurements of cytotoxic activity of different types of synthesized molecules. The obtained compounds were subjected to measurements of cytotoxicity against the cell line HCT116 and most of them have shown remarkable levels of biological activity. The seventh and final chapter brings together the methods of synthesis and characterizations of all products obtained during this work.
