Harmonic Response Of Large Engineering Structures With Nonlinear Modifications

During the design and development stages of mechanical structures, after each modification made in order to satisfy design criteria, dynamic characteristics of the structure change and should be determined through reanalyzing the structure dynamically. Due to the significance of computational time and cost in design processes, it is inevitable for structural modification methods, especially for large systems, to become involved in predicting the dynamic behavior of modified structures from those of the original and modifying structures. Since most engineering structures are inherently nonlinear, linear approach may not be valid no more. Therefore, conventional structural modification methods can not be directly used, instead a nonlinear structural modification method needs to be employed. In this thesis, it is aimed to adapt an effective linear structural modification method to structures with nonlinear modification or coupling. The amplitude dependencies of nonlinearities are modeled by using describing function method. Mathematical formulations are embedded in a computer program developed in MATLAB&reg / with a graphical user interface. The software uses modal analysis results of ANSYS&reg / for the original structure and dynamic stiffness matrix and nonlinearity information that belong to the modifying structure in order to calculate dynamic response of the modified structure. The approach is verified by applying it to both discrete and real test structures previously studied in literature and generated discrete structures, then comparing the results with prior ones and ones obtained via time domain integration, respectively. Several other case studies are also included in order to demonstrate the applicability and to investigate the performance of the method. It is concluded in this study that the structural modification method proposed can be successfully and efficiently used for structures with nonlinear modification or coupling.

Le genre Bordetella comporte à l’heure actuelle neuf espèces, majoritairement responsables d’infections respiratoires. B. pertussis, agent de la coqueluche, est le modèle de travail de cette thèse avec d’autres espèces telles que B. holmesii et B. avium. Les endotoxines bactériennes sont les composants essentiels de la membrane externe des bactéries à Gram-négatif. Du point de vue chimique, ce sont des lipopolysaccharides (LPS) qui provoquent un grand nombre de désordres physiopathologiques allant de la simple fièvre, à faible dose, jusqu’au choc endotoxinique mortel, à forte dose. L’analyse structurale des LPS de Bordetella est la spécialité majeure du laboratoire et la structure des endotoxines de la plupart des espèces de ce genre y a été décrite. Il est admis que le lipide A, qui constitue la région hydrophobe des LPS, est responsable de la majorité des activités biologiques de ces molécules. Ainsi, la moindre variation structurale de ces molécules a une répercussion importante sur la reconnaissance hôte-pathogène, les activités biologiques et la virulence bactérienne. A titre d’exemple, il a été mis en évidence que la modification spécifique, par substitution des groupes phosphate du lipide A, avec de la glucosamine, jouait un rôle majeur dans la modulation de la réponse immunitaire. Cette originalité structurale qui a été mise en évidence dans notre équipe chez B. avium, B. bronchiseptica puis chez B. pertussis; elle semble être un trait spécifique des Bordetelles. Il faut savoir que la coqueluche fait des ravages dans les pays sous-développés et touche les nouveau-nés dans de nombreux pays, comme la France, d’une maladie infectieuse mortelle dans les cas graves. Le vaccin qui ne peut être injecté qu’à l’âge de 2-3 mois et dont les rappels ne sont pas régulièrement suivis est imparfait. Les spécialistes du domaine ont reconnu qu’un complément antigénique serait nécessaire pour le rendre plus efficace. Au cours de cette thèse, nous avons analysé la structure des LPS d’isolats cliniques de B. pertussis afin d’étudier leur évolution et leur adaptation avec le temps ainsi que leur application vaccinale potentielle. De plus, concernant deux souches de B. pertussis, BP338 et BP18-323 nous avons contribué à la mise en évidence de nouveaux gènes impliqués dans la biosynthèse de la GlcN substituant les groupes phosphate de la région lipidique et à expliquer la différence de longueur du seul acide gras distinct entre les deux souches. L’étude de l’influence de ces éléments structuraux sur l’activation du complexe, TLR4/MD-2 apporte de nouveaux éclairages sur les interactions entre les lipides A et ce récepteur. Nos études sur les isolats cliniques de B. holmesii, pathogène opportuniste responsable d’affections de type coqueluche, montrent une grande hétérogénéité structurale du lipide A au sein d’un même isolat. Nous avons montré dans cette souche la présence d’un marqueur spécifique des souches de Bordetella, il s’agit d’un acide gras présent uniquement chez les lipides A des isolats humains. Nos travaux effectués sur des isolats cliniques de B. pertussis appartenant aux ères pré- et post-vaccinales et provenant de différents pays, montrent une perte du matériel génétique avec une déficience de certains antigènes majeurs. Nous avons démontré, via des méthodes physico-chimiques, que ces modifications ne concernaient pas les LPS de ces isolats. La stabilité de ces antigènes ainsi que nos méthodes de purification, nous permettent de proposer que ces LPS détoxifiés soient de bons candidats pour améliorer l’efficacité des vaccins coquelucheux acellulaires. Enfin, toutes les études structurales présentées dans cette thèse ont permis de mieux comprendre la régulation de certains gènes en réponse à un stress extérieur. Elles participent, sur l’exemple d’un pathogène majeur, au déchiffrage des mécanismes moléculaires qui mènent à la virulence et à l’adaptation bactérienne. / The Bordetella genus is actually composed of nine species responsible for respiratory infections. B. pertussis, the agent of whooping cough, is the main model of this thesis along with other species such as B. holmesii and B. avium. Bacterial endotoxins are the major components of Gram-negative bacteria external membrane. From a chemical point of view, they are lipopolysaccharides (LPS) causing a high number of pathophysiological disorders ranging from low fever at weak doses, to lethal endotoxic choc at high ones. Structural analysis of the Bordetellae LPS is the major specialty of our group where the endotoxin structures of most species of the genus were described. It is well-known that lipid A, which constitutes the hydrophobic moiety of LPS, is responsible for the majority of biological activities of these molecules. Thus, any structural change of these molecules has an important impact on host-pathogen recognition, biological activities and bacterial virulence. For example, it has been demonstrated that the specific modification by grafting glucosamine on lipid A phosphate groups plays a major role in modulating the immune response. This structural peculiarity was highlighted by our team first in B. avium, B. bronchiseptica then in B. pertussis; it seems to be a unique trait of Bordetella. It should be noted that pertussis wreaks havoc in developing countries and affects newborns in several others, including France, where this infectious disease causes a significant death toll. The vaccine, which cannot be injected before the age of 2-3 months, could be improved and boosters are not regularly monitored. Experts in the domain have recognized the lack of an antigenic complement to make it more effective. In this thesis, we analyzed the structure of LPS from B. pertussis clinical isolates to study their evolution and adaptation over time along with their potential use in the design of new vaccines. In addition, regarding two strains of B. pertussis, BP338 and BP18-323, we have contributed to the identification of new genes involved in the biosynthesis of GlcN substituting the phosphate groups of the lipid moiety, which helped explaining the difference in the length of the single fatty acid differing between the two strains. The analysis of the influence of these structural elements on the activation of the receptor complex, TLR4/MD-2 sheds new light on the interactions between lipids A and this receptor. Our studies on clinical isolates of B. holmesii, an opportunistic pathogen responsible for pertussis-like illness, show great structural heterogeneity in the lipid A of these isolates. We showed the presence of a specific marker of Bordetella species, namely a fatty acid present only in the lipid A of human isolates. Our works on B. pertussis clinical isolates belonging to pre- and post-vaccine eras and coming from different countries show a loss of genetic material with a deficiency in certain major antigens. We have demonstrated, via physico-chemical methods, that these modifications did not affect the LPS of these isolates. The stability of these antigens and our ability to purify them, allow us to propose that detoxified LPS could be good candidates for improving the effectiveness of acellular pertussis vaccines. Finally, all structural studies presented in this thesis have provided insight into the regulation of certain genes in response to external stress. Our compiled work on a major pathogen is an important step in deciphering the molecular mechanisms leading to bacterial virulence and adaptation.

Development And Comparative Evaluation Of A New Structural Modification Method In Application To Aircraft Structures

In the development of engineering products, it is necessary to predict dynamic properties of the modified structures. Achieving such predictions by using the structural properties of the original structure and information on the modifications is commonly referred to as structural modification analysis. In this thesis, &Ouml / zg&uuml / ven&rsquo / s Structural Modification Method and Sherman-Morrison Method are selected as exact methods for structural modifications to predict the dynamics of a locally modified structure. Also, a new structural modification method named as &ldquo / Extended Successive Matrix Inversion Method&rdquo / is developed in this study. These three methods are implemented in a software developed herein, called &ldquo / Structural Modification Toolbox&rdquo / . The software uses modal analysis results of MSC Nastran&copy / for the original structure and calculates the modified frequency response functions by any of the methods above. In order to validate the software, direct modal analysis results of MSC Nastran&copy / for the frequency response functions of the modified structure are used. The methods are compared in terms of computational time, and the effectivity of each method is studied as a function of modification size to determine which of these methods is more suitable. In order to investigate the application of the methods and compare their results with experimental ones, modal tests are conducted on a scaled aircraft structure. The solutions are compared with test results obtained from modified test structure. Additionally, the software is used for comparison of real aircraft test results and frequency response functions of the modified structure.

Modelling Part Dynamicsin Machining Processes Considering Material Removal

Self-excited chatter vibration in machining is one of the most important limitations on utilizing the increasing productivity of modern machine tools. In order to predict stable depth of cuts at high cutting speeds, the stability lobe diagram for a spindle-tool holder-tool combination must be developed. The frequency response function (FRF) of the system must be known for analytical prediction of the stability lobe diagrams. When the flexibility of the workpiece is important, the workpiece itself should be included in the system model by considering the variation of its dynamics at different stages of the machining process. In this thesis, an exact structural modification method is used to find the frequency response functions of the workpiece to be machined at every stage of the machining process. In order to obtain the system matrices and the modal parameters of the original structure, a commercial finite element program MSC. Marc&copy / is used. The frequency response functions of workpiece are calculated by using the computer program developed in this thesis, and are compared with the ones found by MSC. Marc&copy / . The stability lobe diagram of the system is obtained by combining the FRFs of the tool with those of the workpiece. The effects of the dynamic of the workpiece on the stability lobe diagrams are studied extensively by using the results of case studies presented in this thesis. In order to increase productivity, minimum chatter-free machining times are also calculated for different cases. For this purpose the effects of the different radial depth of cuts and different cutting strategies on the stability and the machining time are examined with various case studies.

Structural Modification With Additional Degrees Of Freedom In Large Systems

In the design and development stages of mechanical structures, it is important to predict the dynamic characteristics of modified structures. Since time and cost are critical in design and development stage, structural modification methods predicting the dynamic responses of modified structures from those of the original structure and modification properties are very important, especially for large systems. In this thesis structural modification methods are investigated and an effective structural modification method for modifications with additional degrees of freedom is adapted to structures with distributed modifications and the performance of the method is investigated. A software program is developed in order to apply the structural modification method with additional degrees of freedom. In the software, the dynamic response of the modified structure is predicted by using the modal analysis results of ANSYS for the original structure and dynamic stiffness matrix of the modifying structure. In order to validate the approach used and the program developed, the dynamic analysis results obtained for modified structures by ANSYS are compared with those obtained by using the software. In order to investigate the performance of the structural modification method in real applications, the method is applied to a scaled aircraft model, and the results are compared with experimental results. In order to demonstrate the importance of using the structural modification method with additional degrees of freedom for distributed modification, lumped and distributed models are used for a distributed modification and results are compared. It is concluded in this study that using structural modification methods with additional degrees of freedom for a distributed modification increases the accuracy of the results, and it is observed that the method adapted is efficient for local modifications.

Development Of A Fast Analytical Method For Prediction Of Part Dynamics In Machining Stability Analysis

The objective of this study is to develop and implement practical and accurate methods for prediction of the workpiece dynamics during a complete machining cycle of the workpiece, so that FRFs of the workpiece can be used in chatter stability analysis. For this purpose, a structural modification method is used since it is an efficient tool for updating FRFs due to structural modifications. The removed mass is considered as a structural modification to the finished workpiece in order to determine the FRFs at different stages of the process. The method is implemented in a computer code and demonstrated on representative parts such as turbine blades. The predictions are compared and verified with the data obtained using FEA. The FRFs are used in chatter stability analyses, and the effect of part dynamics on stability is studied.

The Effects Of Structural Modifications On Acoustic Characteristics Of Enclosed Bodies

Low frequency noise caused by vibrating panels can pose problems for vehicles from noise, vibration and harshness (NVH) standpoint. In order to reduce interior noise levels in cars, some structural modifications are required on the car body. Structural modifications studied in this work are stiffeners welded on the walls of enclosed structure to change vibration characteristics. In this thesis, interaction between acoustic domain inside closed structures and their vibrating enclosing boundaries are analyzed. Analysis of vibro-acoustic behavior includes frequency response analysis of structure by Finite Element Method (FEM) and sound pressure level (SPL) prediction of the cabin interior by Boundary Element Method (BEM). The standard parts of the analyses are performed using available CAE (Computer Aided Engineering) software. It is demonstrated that the structural modification technique integrated with the vibro-acoustic model of the system reduces the computational effort considerably. The frequency response functions of a structure for each modification can easily be obtained in a fast and efficient way by using the structural modification technique. Thus, effects of design changes in the structure body on noise levels due to vibration of the structure can be very handily and efficiently studied. In the case studies presented, the effects of various different stiffeners applied on a simple closed structure are studied in detail.

Influência dos processos de engraxe e acabamento na modificação da estrutura e nas propriedades do couro

O estudo do colagênio na pele sobre sua estrutura e propriedades, composição e interações com substâncias levam a um maior entendimento das alterações químicas, micro e macroscópicas que ocorrem durante o processo de produção do couro. No processamento do couro são realizadas várias etapas que promovem a alteração da estrutura dérmica da pele, como acontece no acabamento molhado, onde reações químicas com a molécula de colagênio curtido são realizadas pela adição de produtos químicos, como recurtentes e óleos de engraxe, influenciando na estrutura macroscópica final, e ainda, dando propriedades específicas para o produto final, conforme a aplicação do couro. O acabamento final tem o objetivo de agregar valor ao artigo de couro, e as interações dos produtos aplicados sobre o couro nas camadas mais externas da estrutura que acontecem nesta etapa são determinantes quanto à qualidade do couro que se quer obter e suas propriedades, inclusive na parte estética e funcional. Ainda, durante seu processamento, o couro mantém propriedades importantes da estrutura fibrosa formada pelo colagênio, sendo este o principal motivo do couro, como produto industrializado, apresentar propriedades únicas e particulares, fazendo dele um material de vasta aplicação e de alto valor agregado. Neste trabalho, foram feitas determinações para comparar formulações de engraxe para couro estofamento automotivo (uma para couro hidrofugado e outra não), e verificar a influência dessas formulações utilizadas sobre as propriedades dos couros em estágio de processo wet-blue, semi-acabado e após aplicação de acabamento. Ainda, na determinação de propriedades, foi estudada a influência do local de retirada da amostra no couro nos experimentos realizadas com repetição em couros distintos, duplicata para três regiões do couro, e para cada etapa do processo. Para comparação entre as formulações, os couros utilizados foram meio gêmeos, usando o mesmo critério da região do couro. As variáveis de resposta de propriedades dos couros assim obtidos foram baseadas nas análises de resistência mecânica, caracterizações químicas e absorção de água, e ainda para evidenciar a modificação da estrutura do couro foram feitas análises de MEV (microscopia eletrônica de varredura) e superfície interna pelo método BET. Os resultados indicam que as técnicas analíticas utilizadas mostram reprodutibilidade para os diferentes couros testados, quando comparados região a região, uma vez que os resultados indicam que existe forte tendência da retirada do local de amostragem conforme experimento por regiões do couro, ficando assim indicado que em trabalhos futuros, essa variável seja controlada. Conforme esperado, o couro hidrofugado teve menor absorção de água do que o couro apenas engraxado, devido à sua menor área superficial, evidenciado pelo MEV e BET. O estágio de processo teve maior influência nos resultados dos testes físicos. As caracterizações químicas serviram para evidenciar as diferenças entre o couro wet-blue e os couros acabados de maneira geral. / The study of changes in collagen from the hide related to its structure, properties and chemical interactions should be accompanied during the process of leather production. In the processing of leather, some stages are carried out promoting changes on the dermic structure of the hide, as it happens in wet-finishing stage, where chemical reactions happen into collagen molecules by the addition of chemical products, as re-tanning agents and fatliquors, influencing the final macroscopic structure and still giving specific properties for the finished leather according to its application. The finishing stage has the objective to add value to the leather article, and the interactions of products applied on the leather in external layers of the structure are determinative to the desired quality and properties of the leather, as well to the aesthetic and other functions. Still during its processing, the leather keeps important properties by the structure of the fibers formed of collagen, being this the main reason of the leather, as industrialized product, to present particular properties and making of it a material of vast application and high aggregate value. In this work, analyses are made to compare formulations of fatliquoring and hidrofugation for automotive leather in order to verify the influence of these on the properties of leathers in stages of wet-blue, crust and finishing. Moreover, the influence of the region of withdrawal of the sample from the hide was studied with repetition in distinct leathers. Still, the determination of properties, we studied the influence of local removal of the leather sample in the experiments performed with repetition in different leathers, duplicate for three regions of leather, and for each process step. For comparison between the formulations, the leathers used were half twins, using the same criteria in the region of the leather. The response variables of the properties of leather thus obtained were based on analysis of mechanical strength, chemical characterization and water absorption, and also to highlight the changed structure of the leather were analyzed by SEM (scanning electron microscopy) and the inner surface the BET method. The results indicate that the analytical techniques used to show the reproducibility of different leathers tested compared region by region, since the results indicate a strong trend of withdrawal of the sampling location as experiment by regions of the leather, thus indicated that in future work, this variable is controlled.
