Growth and feeding of juvenile chinook salmon, Oncorhynchus Tshawytscha, in "in situ" enclosures

A feeding experiment was designed to examine how fish growth rates are affected by the size abundance of pelagic zooplankton. Juvenile chinook salmon, Oncorhynchus tshawytscha , were raised in 90 m³ mesh enclosures in Saanich Inlet, B.C. The enclosures permitted ample water and zooplankton circulation while retaining 5-6 gram juvenile salmon. The enclosed fish grew at an average rate of 1.8% wet body weight/day for a six week experimental period. Weekly growth rates ranged from 3.9%/day while food was abundant, to -0.5%/day when food was scarce. Several analytical methods were used to establish a relationship between fish gr.owth and the size and abundance of zooplankton in the enclosures. There is a strong relationship between the fish growth rates and the abundance of 1.4-4.5mm prey. Rates of successful search vary directly with the size and inherent contrast of a prey item. A minimum rate of successful search of 2.0m³/hour was estimated from a functional response curve for salmon feeding on 1.4-4.5mm zooplankton. This value is discussed in relation to a salmonids physical capabilities and results from previous field studies and tank experiments. Daily growth increments on the otoliths of the enclosed fish were examined with respect to daily variations in water temperature and zooplankton abundance. Extremes in food abundance appear to have a significant and consistent effect on the spacing of the growth rings. Contesting modernism : communities and the pacific salmon revitalization plan

This thesis explores the role for social work in addressing government policies that threaten the sustainability of small coastal communities. The response of government and industry to the globalization of trade and resource degradation is at odds with the needs of people. Utilizing a case study methodology the development and implementation of the Pacific Salmon Revitalization Plan is explored. This department of fisheries plan to rationalize the fishery was highly contested on the grounds that it took jobs out of small coastal communities. It was accused of benefiting the large fishing corporations and the urban based fishing fleet, which had the capital to profit from the plan. Concentrated opposition from coastal communities, fishers, advocacy groups and academics was unsuccessful in changing the plan. The assumptions of modernism - expert knowledge, scientific rationality and orthodox economics - as well as distorted communications, were postulated to be behind this lack of success. A post modern analysis suggests that a successful challenge to the plan would have incorporated the local knowledge of fishers and coastal communities within a process of fair and equitable public discourse aimed at reaching intersubjectively mediated understanding. For social work this demonstrates the need to work conjointly with communities and affected groups to identify the modernist assumptions on which policy decisions are based and develop locally derived alternatives to these assumptions. And most importantly, that the focus of social change efforts be on demanding a process for discussion and decision-making that ensures that the concerns of effected individuals will be fairly addressed. / Arts, Faculty of / Social Work, School of / Graduate

Examining fish quality : the evaluation of the use of lipids as a measure of condition in wild Atlantic salmon

Considering the response of organisms to their environment is difficult; it is made more so if population numbers cannot be closely monitored. In such cases different methods of population assessment are required. This thesis uses lipids as a measure of Atlantic salmon (Salmo salar L.) quality and investigates its usefulness in indicating fish condition. The first study examines the relationship between fish total lipid content and W[sub]R condition factor; this study clearly demonstrates that there is a significant positive relationship between the condition factor of a fish and its total lipid content. In the following study the lipid storage between the different tissues of the Atlantic salmon is considered. This indicates that the red muscle and the adipose tissues hold higher concentrations of lipid than the white muscle. However, the white muscle makes up the majority of lipid tissue mass in the Atlantic salmon so contains the bulk of stored lipid in a fish, at low concentration. The next study investigates the effect of spawning on Atlantic salmon condition. Salmon can be seen preferentially conserving lipid in their musculature and drawing down the lipid stored in their adipose tissues. The following study looked at one key lipid group, triacylglycerides, in salmon. Triacylglycerides are energetically important in fish and this study found that the spawning process depleted triacylglyceride reserves, but that the red muscle conserves triacylglycerides even after spawning. The final study considers the relationship between maternal quality and egg quality, identifying that longer Atlantic salmon produce eggs with more lipid after spawning migration. Egg lipid concentrations were comparably maintained between fish. Monitoring quality in this way is a useful tool to determine population wellbeing and help indicate where populations are compromised.

Trophically transmitted parasites as ecosystem indicators : relationships among parasite community structure, juvenile salmon diet composition, and ocean conditions

Recent research conducted throughout the Northern California Current (NCC) on the ecology of Pacific salmon (Oncorhynchus spp.) indicates that variable ocean conditions affect the community composition of zooplankton in the nearshore environment which, in turn, can affect the quality of prey for fish, sea birds and mammals. Interannual variability in the quality and composition of the copepod community in the NCC during early marine residency of some Pacific salmon populations is related to survival to adulthood. However, copepods make up a small portion of the diet of coho and Chinook salmon, and the mechanistic linkages between ocean climate, zooplankton composition and salmon prey remain unclear. Parasite analysis provides a supplement to traditional diet analysis that can describe the foraging history of a host species. Coho salmon (O. kisutch) and Chinook salmon (O. tshawytscha) serve as hosts to an array of marine parasites acquired through consumption of infected intermediate hosts such as copepods, euphausiids, and planktivorous fishes. Causing little or no harm to their salmon host, the presence of trophically transmitted parasites provides information on the dietary history of their salmonid host beyond the 24 hours associated with traditional diet analysis. This study (1) examined differences in feeding behavior of coho and Chinook salmon during their early marine residency using both stomach and parasite community analyses and (2) tested the hypothesis that variability in ocean circulation patterns (measured through the Pacific Decadal Oscillation, sea surface temperature (SST) and Bakun's upwelling index) and copepod species composition are related to variability in the community structure of trophically transmitted marine parasites found in juvenile salmon. I compared the abundance and species composition of parasites recovered from juvenile Columbia River coho and upper Columbia River summer and fall Chinook salmon captured off the coast of Washington from 2002 to 2009. I also compared interannual variability in parasite assemblages to physical and biological indices of ocean conditions. Coho and Chinook salmon consumed similar prey taxa; however, the species richness and abundance of trophically transmitted parasites indicated that Chinook salmon consumed a greater diversity and abundance of infected prey. In addition, differences in the abundance of fish in the diet and Anisakis simplex, a parasitic nematode known to infect salmon through fish consumption, suggest that Chinook salmon consistently consumed more fish prey than coho. In contrast, coho appeared to consume more euphausiids as indicated by stomach content analysis and increased abundance of the euphausiid parasite, Rhadinorhynchus trachuri. Shifts in the parasite community composition of both coho and Chinook salmon were related to interannual variability in SST and the biomass of southern-origin copepods (r > 0.7, P < 0.05). The acanthocephalan R. trachuri and a tetraphyllid cestode were associated with "warm" SSTs and greater biomass of lipid-poor, subtropical copepods while the nematode A. simplex was more abundant in years of "cold" SST and a relatively low biomass of subtropical copepods. These results provide novel insight into differences in the diet of Columbia River coho and Chinook salmon and illustrate linkages between ocean climate, zooplankton community composition and salmon diet during early marine residency. / Graduation date: 2012

Management strategies to control sexual maturation in sea-reared Atlantic salmon (Salmo salar L.) : biomass management, light-manipulation and sterility

Pre-harvest sexual maturation in farmed Atlantic salmon, Salmo salar, remains a key biological bottleneck compromising biomass and financial output, production predictability, environmental respect, stock welfare and the overall sustainability of the on-growing industry. The management practices currently in place are not optimized and events of high maturation rate are still sporadically observed. From an ecological perspective, the escape of reproductively competent, domesticated Atlantic salmon constitutes a threat to the integrity of wild stocks. The forecasted expansion of the Scottish salmon industry compels the need for a comprehensive and more reliable control of sexual maturation. The general aim of this research project was to optimize the current management strategy (windows of light-manipulation and quality grading) and test alternative practices (lighting-technologies, selective harvest and triploidization) in the control of pre-harvest sexual maturation within the Atlantic salmon on-growing industry. In that end, a number of trials were performed using stock reared in sea-cages on a full commercial-scale or in tanks on an experimental scale. The results of this project are organized around three experimental chapters dealing consecutively with body-size dimorphism, grading and harvest quality; light manipulations and triploidy. In each chapter, two original manuscripts either published or in review are included. In addition to these experimental results, a literature review chapter composed of two review papers on the photoperiodic synchronization and developmental regulation of maturation in salmonids and on morphological skin colour changes in teleosts (published) are presented. In the first experimental chapters, we aimed at investigating the possibility of detecting and selectively harvesting a high proportion of sexually recruited fish before flesh quality deterioration. Results clearly showed that body-size dimorphisms between maturity cohorts at the end of the anabolic window of reproduction (June/July) are strong and standard predictors of maturation among related populations with the same freshwater history. Dimorphism can therefore be modelled to easily and accurately estimate maturation rate in a number of discrete rearing-units. If required, a high proportion of sexually recruited fish can be selectively harvested as superior quality product while leaving the immature fish for further on-growing. This provides an alternative to visual grilse grading that is not feasible in large-scale aquaculture systems, prevents downgrading and increases production predictability as compared to emergency harvests. Furthermore, our results showed immature males grow faster than immature females which should be further investigated to fully determine gender specific performances and nutritional requirements. Weight-grading performed earlier in the cycle affects the sex-ratio within individual pens and in turn apparent performance. This work also revealed that Atlantic salmon can exhibit significant variations in skin colouration resembling the onset of nuptial display but that are not related to sexual recruitment and do not correlate with reduced flesh quality. This originates from a lack of purine (silver) pigments which was also identified, to a larger extent, as characteristic of the nuptial display. This suggests a degree of desmoltification in these histologically immature fish. The instrumental colouration of the altered phenotype was shown to be improved towards a more silver-like appearance by direct ice-contact. This knowledge could facilitate post-harvest quality grading towards the most appropriate market channel and increase product acceptance and attractiveness. The second experimental chapter investigated the possibility of improving photoperiodic manipulation used to suppress early maturation, currently applied for 6-months during the second winter at sea using wide-spectrum, high-intensity lighting systems. Our results showed that the window of continuous artificial-light (LL) exposure can be reduced to 4-months following its onset in early January without compromising its efficiency in suppressing pre-harvest maturation. In addition, alternative lighting technologies were also highly potent at suppressing sexual maturation. The mean-irradiance (intensity) generated within a commercial sea-cage was inversely proportional to the suppression of nocturnal plasma melatonin (light perception hormone) and negatively correlates with the maturation rate within the commercial sea-pen. Threshold levels of light-intensity required to achieve optimal (total) suppression of sexual maturation are suggested. Alternative, narrow band-width lighting-technologies (cold cathode and light-emitting diodes) present an array of technical, practical, economic and welfare benefits comparing to the system currently in use. Clear improvements of the photoperiod-manipulation strategy were demonstrated and these would reduce economic and environmental costs but also potential impacts on animal welfare. The third experimental chapter showed the strong potential of sterile-triploid Atlantic salmon stocks both in freshwater and seawater. Triploid out-of-season smolts were produced for the first time using a classical accelerated "square-wave" photoperiod. Triploidization affected the smoltification pattern but had no detrimental effects on freshwater and early seawater performances under both a S0+ and S1 regime. This illustrates the need to adapt the timing of seawater transfer for successfully producing triploid Atlantic salmon post-smolts. Following one year of seawater rearing, the prevalence of external deformities was higher in triploids but remained within acceptable levels. Importantly, the incidence of vertebral deformities and ocular cataract was higher in triploids possibly due to their specific requirements. It is suggested that tailoring the diet to the nutritional requirements of triploids holds strong potential for remediation. This must be addressed if the use of sterile-triploid stock is to become a commercial reality. The present research project provides means to optimize the maturation management strategy within the Atlantic salmon on-growing industry through light-manipulation, maturation detection and selective harvest, and quality grading. Proposed improvements have the potential to increase biomass and financial output, production predictability, environmental respect and animal welfare and will allow standardization of the overall control of pre-harvest sexual maturation. Their implementation provides a comprehensive strategy likely to favour a sustainable expansion of the Atlantic salmon industry. From a longer term perspective, the rearing of sterile-triploid stocks is promising and should be actively investigated to isolate domesticated strains from their wild conspecifics. This would also eliminate the need for on-growers to deploy a maturation management strategy that that might still affect stock welfare and remains, despite the strong improvements demonstrated, not 100% reliable, costly, technical and protracted.

Detection of QTL affecting flesh quality traits (body lipid percentage and flesh colour) using molecular markers (microsatellites and AFLP markers) in Atlantic salmon (Salmo salar L.)

Flesh colour and fillet fat percentage are the two most important attributes to salmon fillet quality. A medium genetic component to body lipid percentage within commercial lines has previously been shown (h2 = 0.17-0.24). A low level of heritability (h2 = 0.16) has also been reported for flesh colour in Atlantic salmon. To investigate whether this genetic component includes loci of major effect, a genome-wide QTL scan was performed with commercially bred Atlantic salmon (Landcatch Natural Selection). Five large full-sib families (10 parents with 153 offspring) were genotyped using microsatellite markers. To utilize the large difference between sire and dam recombination rate, a two-stage genotyping was employed. Initially, the parents and offspring were genotyped for two microsatellite markers per linkage group, and sire based QTL analysis was used to detect linkage groups with significant effects on those flesh quality traits. A linear-regression based interval as analytical method was applied for QTL detection. The results revealed evidence of QTLs affecting percentage fat percentage and flesh colour on linkage groups LNS16 and LNS1 respectively. To confirm the QTL and to provide an improved estimate of position, a dam-based analysis was then employed. One major QTL was located on the genome-wide significance level for percentage fat percentage. Microsatellite marker Ssa0016NVH (at position of 1.3 cM) was found to be tightly linked to QTL affecting percentage fat percentage. In addition, a QTL affecting flesh colour was found to be flanked by microsatellite markers Ssa9.44NUIG at position of 68.7 cM and Ssa0021NVH at position of 50.6 on linkage group LNS16. The evidence for suggestive QTL affecting flesh colour on linkage group LNS1 was also revealed. In order to increase marker density within these and other linkage groups, AFLP markers were employed, 24 primer combinations resulted in a total of 489 polymorphic fragments. Among 11 fragments that were found to be linked to the microsatellite markers on linkage group LNS16, four fragments (AAG-CAC328, AGG-CAG447, AGG-CTA237 and AGG-CTC237) were tightly linked to microsatellite marker Ssa9.44NUIG, but none were found to be linked to microsatellite Ssa0021NVH. Moreover, none of the AFLP markers were found to be linked to microsatellites residing on linkage group LNS1. Using a constructed map of microsatellite and AFLP markers for linkage group LNS16, the dam based analysis revealed a significant QTL for flesh colour at the location of 189 cM, while the sire based analysis detected a significant QTL for fat percentage at the location of 80 cM. Considering the dominant nature and clustering character of AFLP markers, it was concluded that a certain primer combination in AFLP markers could be of limited use for fine mapping and QTL detection in Atlantic salmon.

In this report mark-recapture data are analyzed with the use of weighted log-linear models, mosaics, and computer drawings of fish. The data are from salmon hatcheries, subsequent returns to the hatcheries and commercial catches of salmon. The log-linear models were used to study the effects of several variables on catches and returns. It is shown that these variables may have opposite effects depending on the brood year of the fish, that hatchery returns and commercial catches do not respond in the same way to the variables, and that more research is needed to determine the causes of the brood year differences. The mosaics and fish drawings were used to study the migration of the salmon in the ocean. The results confirm that chinook salmon decrease their food intake during the return trip to the hatchery, and they are consistent with theories of ocean migration of other species of salmon. / Thesis / Master of Science (MSc)
