It is often needed to predict the behavior of structures. With the helpof an FE model, it is possible to see the motion of the structure. Inthis, Master thesis work time domain analyses were carried out on afinite element model representing a window attached by click-ins. Theaim was to establish an FE model which gives a good correlation withtest data. The finite element modelling was carried out in MSC Apex.MSC Nastran was utilised to analyze the FEA model and the resultwas post-processed in Simxpert. Different vibrational tests were made. A crane was used to exciteand support the wall together with the window and the responseswere recorded with tri-axial accelerometers. During the thesis, it wasfound that the FE model requires more work and accurate boundaryconditions to provide better resemblance with the test data.The results deviate from the measured. This opens future possibilities tocarry on the project.

An In Vitro Evaluation of the Kontrolflex Accufile Hand FileTittle, Mia 01 January 2022 (has links)
The aims of this study were to determine the efficacy of the KontrolFlexTM AccufileTM compared to the READY•STEEL™ Flexofile® using an electronic apex locator and to evaluate the instrument’s design via scanning electron microscopy. Actual canal lengths of thirty extracted teeth were determined under magnification. An alginate model was used to determine experimental canal lengths with the Root ZX II apex locator and size #10 Accufiles and Flexofiles. Differences between actual lengths and experimental lengths were compared with Student’s t test. The average experimental lengths were short of the actual length by -0.10 mm (±0.34) and -0.12 mm (±0.16) for the Accufile and Flexofile, respectively with no statistical difference (p > 0.05). SEM images revealed similar non-cutting “batt” tips and a square cross-section for the Accufile and triangular for the Flexofile. Both files provided similarly high levels of reliability when used with the Root ZX II electronic apex locator.

Antibody discovery and engineering using the anchored periplasmic expression (APEx) Escherichia coli display system with flow cytometric selectionVan Blarcom, Thomas John 05 February 2010 (has links)
The development of recombinant proteins for therapeutic applications has revolutionized the pharmaceutical industry. In particular, monoclonal antibodies are the safest class of all therapeutic molecules and account for the majority of recombinant proteins currently undergoing clinical trials. A variety of technologies exist to engineer antibodies with a desired binding specificity and affinity, both of which are a prerequisite for therapeutic applications. This dissertation describes the implementation of a novel combinatorial library screening technology for the discovery and engineering of antibodies with unique binding properties. Combinatorial library screening technologies are used for the in vitro isolation of antibodies from large ensembles of proteins (libraries) typically produced by microorganisms using molecular biology techniques. Our lab has developed a powerful antibody discovery technology that relies on E. coli display by anchored periplasmic expression, otherwise known as APEx. First, I compared the effects of using combinatorial libraries comprising either smaller, monovalent single-chain antibody fragments (scFv), or the much larger, bifunctional full-length IgG antibodies. These technologies were used to isolate a small panel of antigen specific antibodies from the same library of antibody variable domains amplified from a mouse immunized with the Protective Antigen (PA) component from Bacillus anthracis, the causative agent of anthrax. Overall, IgG display resulted in the isolation of a broader panel of variable domain sequences. Most of these variable domains exhibited substantially reduced affinity when expressed as scFvs, which is consistent with the finding that none of these could be isolated from the equivalent scFv library. These results indicate that the antibody format used during in vitro selection affects which antibody variable domains will be discovered. Second, I developed several modifications of the APEx methodology to allow for more efficient recovery of antibodies with desired properties. Specifically, the system was reengineered to simultaneously account for antibody binding and expression levels in order to isolate the highest affinity antibodies with favorable expression characteristics. Third, the new approach, coupled with optimized fluorescence activated cell sorting (FACS) settings, was used to increase the affinity of an antibody by 35-fold resulting in a K[subscript D] of 100 pM. It was demonstrated that genetic transfer of this high affinity antibody specific for the V antigen of Yersinia pestis, the etiologic agent of the plague, conferred increased protection against intranasal challenge with a 363 LD₅₀ of Y. pestis in mice. / text

Este trabalho teve como objetivo avaliar a influência da espessura de dentina radicular apical, da constrição apical e do diâmetro do forame apical na precisão de leituras realizadas com os localizadores foraminais eletrônicos Mini Apex Locator e Root ZX II®. Foram utilizados 30 incisivos inferiores permanentes unirradiculados de humanos, extraídos, com raízes íntegras e ápices completamente formados e portadores de um único canal. Por meio de um paquímetro, as espessuras radiculares dos dentes foram medidas, no sentido mésio-distal a 1,0 e a 4,0mm aquém do forame apical. Após a abertura coronária, uma lima tipo K no 10, munida de limitador de penetração, foi introduzida no canal radicular até que sua extremidade pudesse ser visualizada na altura do forame, com o auxílio de um microscópio óptico com aumento de 7,8X. Dessa medida, subtraiu-se 1,0mm, estabelecendo-se o comprimento de trabalho. A dilatação do canal radicular foi feita, inicialmente, com brocas de Gates Glidden, em ordem numérica decrescente, da número 5 até a número1, até 4,0mm aquém do forame apical. Os dentes foram fixados em um modelo experimental especialmente desenvolvido para permitir a medição com os localizadores foraminais eletrônicos. Tal modelo era constituído por dois segmentos de PVC: um de menor calibre, com diâmetro correspondente a meia polegada por 2,0cm de comprimento, com as duas extremidades abertas e outro, de maior calibre, com uma das extremidades fechada e com diâmetro interno equivalente ao diâmetro externo do primeiro segmento (3/4 de polegada). No segmento de maior diâmetro, foi feito um orifício lateral que permitiu o posicionamento do eletrodo labial do localizador foraminal eletrônico e, para a medição, no seu interior, foi colocado alginato e, então, encaixado o componente de menor diâmetro, fazendo com que o ápice radicular ficasse imerso no alginato. Foram realizadas as leituras com os localizadores, iniciando-se com a lima tipo K no 10 e seguindo-se a seqüência de instrumentação e medida até a lima tipo K no 130; a lima tipo K no 10 foi utilizada em todos os diâmetros. Terminada essa fase, os dentes tiveram os canais sobreinstrumentados, isto é, a ponta da lima ultrapassou o forame apical em 1,0mm, a partir da lima tipo K no 25 e seguindo até a lima tipo K no 130; novas medidas foram obtidas com cada lima que sobreinstrumentou o forame e a lima no 10 foi utilizada em todos os diâmetros. Em todos os casos o canal radicular estava preenchido com hipoclorito de sódio a 1%. Para a análise estatística foram empregados os testes de Análise de Variância a dois critérios e de Tukey. Os resultados indicaram que as variáveis capazes de influenciar a recisão das leituras com os localizadores foraminais eletrônicos foram a eliminação da constrição apical com o consequente aumento do diâmetro do forame apical, ao contrário da espessura da parede dentinária do canal radicular, que não interferiu significativamente na precisão das leituras. / This study evaluated the influence of the apical root dentin thickness, apical constriction and diameter of the apical foramen in the accuracy of readings obtained using the electronic apex locators Mini Apex Locator and Root ZX II ®. The teeth were fixated in an experimental model especially designed to allow the measurement with the electronic apex locators. This model was composed of two PVC segments: one smaller, with diameter corresponding to half inch with 2.0cm length, with both ends open; and the other, with larger diameter, with one end closed and internal diameter similar to the external diameter of the first segment (3/4 inch). In the segment with greater diameter, a lateral orifice was made to allow positioning of the lip electrode of the electronic apex locator. For the measurement, alginate was poured and the component with smaller diameter was fitted, so as the root apex was immersed in alginate. Readings were performed using the electronic apex locators, initiating with a 10 K file and following the sequence of instrumentation and measurement up to 130 K file. The 10 K file was used in all diameters. After this stage, the root canals were overinstrumented, i.e. the file tip was introduced until 1.0mm beyond the apical foramen, beginning with 25 K file up to 130 K file; new measurements were obtained with each file overinstrumenting the apical foramen, and the 10 K file was used in all diameters. In all cases, the root canal was irrigated with 1% sodium hypochlorite. Statistical analysis was performed by two-way analysis of variance and the Tukey test. The results indicated that the variables that may influence the accuracy of readings with the electronic apex locators were the elimination of apical constriction with consequent increase in the diameter of the apical foramen, different from the thickness of the root canal dentinal wall, which did not significantly influence the accuracy of readings.

Ecology and evolution of tolerance in two cruciferous speciesBoalt, Elin January 2008 (has links)
<p>Tolerance to herbivory is the ability of plants to maintain fitness in spite of damage. The goal of this thesis is to investigate the genetic variation and expression of tolerance within species, determine whether and in what conditions tolerance has negative side-effects, and how tolerance is affected by different ecological factors. Tolerance is investigated with special focus on the effects of different damage types, competitive regimes, history of herbivory, and polyploidization in plants. Studies are conducted as a literature review and three experiments on two cruciferous species Raphanus raphanistrum and Cardamine pratensis.</p><p>In the tolerance experiments, plants are subjected to artificial damage solely, or in a combination with natural damage. A literature review was conducted in order to investigate the effects of damage method. We found that traits related to tolerance, such as growth and fitness were not as sensitive in regard to damage method as measures of induced chemical traits, or measures of secondary herbivory.</p><p>Genetic variation of tolerance was demonstrated within populations of R. raphanistrum and between subspecies of C. pratensis. In R. raphanistrum, traits involved in floral display and male fitness were positively associated with plant tolerance to herbivore damage. A potential cost of tolerance was demonstrated as a negative correlation between levels of tolerance in high and low competitive regimes. I found no evidence of other proposed costs of tolerance in terms of highly tolerant plants suffering of reduced fitness in the absence of herbivores or trade-offs in terms of a negative association between tolerance to apical and leaf damage, or between tolerance and competitive ability. In C. pratensis, higher ploidy level in plants involved higher levels of tolerance measured as clonal reproduction. Furthermore, populations exposed to higher levels of herbivory had better tolerance than populations exposed to lower levels of herbivory. In this thesis, I demonstrate evidence of different components for the evolution of tolerance in plants: genotypic variation, selective factors in terms of costs and ploidization, and selective agents in terms of changing environment or herbivore pressure.</p>

Previous issue date: 2013 / Resumo: O tratamento por meio de revitalização do canal radicular tem sido considerado para dentes com rizogênese incompleta e polpa necrosada. A revitalização do canal radicular compreende a descontaminação do canal e o preenchimento do mesmo com coágulo visando a invaginação do espaço do canal com tecido conjuntivo e consequente revitalização do espaço pulpar desvitalizado. O objetivo deste estudo foi analisar histologicamente os efeitos da ampliação do forame apical no procedimento de revitalização do canal radicular em dentes de cães, com polpa vital ou necrosada, e com ápices completamente formados. Foram utilizados 2 cães da raça Beagle, que foram divididos em 5 grupos: G.1 - necrose pulpar com coágulo sanguíneo; G.2 - necrose pulpar com medicação intracanal (associação de Ca(OH)2 com CLX gel 2%); G.3 - controle positivo polpa vital com coágulo sanguíneo; G.4 - controle positivo polpa vital com medicação intracanal (associação de Ca(OH)2 com CLX gel 2%); G.5 - controle negativo - canais radiculares com lesões apicais induzidas. Para a indução de necrose pulpar e desenvolvimento das lesões periapicais, os dentes foram mantidos abertos para contaminação dos canais radiculares pelo período de 180 dias. Os canais foram tratados pela técnica crown-down e os forames apicais ampliados até a lima K #40. Os resultados demonstraram que no grupo G.1 houve revitalização do canal radicular, total ou parcial, em 63,6% dos dentes. No grupo G.2 em 77,8% dos casos houve revitalização parcial ou total do canal radicular. No grupo G.3 houve revitalização do canal radicular, parcial ou total, em 100% dos casos. No grupo G4 não houve revitalização do canal radicular em 100% dos casos (houve selamento biológico apical). No grupo G.5 em 100% dos casos houve formação de lesão periapical. Desta forma, pode se concluir que a revitalização do canal radicular em dentes com necrose pulpar ocorreu em maior percentual no grupo com medicação intracanal, enquanto que, nos dentes com polpa vital a revitalização do canal radicular ocorreu em maior porcentagem no grupo com coágulo / Abstract: Treatment through revitalization of the root canal has been considered for teeth with incomplete root formation and necrotic pulp. The revitalization of root canal includes canal decontamination and filling with clot aiming the invagination of canal space with connective tissue and consequent revitalization of the devitalized pulp space. The aim of this study was to histologically analyze the effects of apical foramen enlargement in root canal revitalization procedure in dogs' teeth, with vital or necrotic pulp, with completely formed apices. Two Beagle dogs were divided into 5 groups: G.1 - pulp necrosis with blood clot; G.2 - pulp necrosis with intracanal medication (association of Ca(OH)2 with 2% chlorhexidine gel); G.3 - positive control vital pulp with blood clot; G.4 - positive control vital pulp with intracanal medication (association of Ca(OH)2 with 2% chlorhexidine gel); G.5 - negative control - root canals with induced apical lesions. For induction of pulp necrosis and periapical lesion development, the teeth were kept open to root canals contamination for a period of 180 days. The root canals were treated by crown-down technique and apical foramen was enlarged to #40 K file. The results showed that in G.1 group there was root canal revitalization, partial or total, in 63.6% of teeth. In G.2 group, partial or complete root canal revitalization occurred in 77.8% of cases. In G.3 group, partial or total revitalization of root canals occurred in 100% of cases. In G.4 there was no revitalization of the root canal in 100% of cases (there were biological apical sealing). In the group G.5, in 100% of cases there was formation of apical lesions. Trocantheric Femoral Fractures - a retrospective cohort study comparing reoperation rates before and after implementation of new treatment protocolSmith, Sean January 2020 (has links)
Introduction: Hip fractures are very common in Sweden. Keeping reoperation rates as low as possible is critical. One factor correlated to mechanical complications and thereby reoperation rates is the tip-apex distance. In April of 2017 the Orthopedic clinic at Karlstad’s Central Hospital in Sweden implemented new routines for the choice of treatment method of pertrocantheric and subtrocantheric femoral fractures. Aim: The primary aim of this study was to compare reoperation rates of pertrocantheric and subtrocantheric femoral fractures before and after implementation of new routines. Secondly, we wanted to study the association between the tip-apex distance and reoperation rates for pertrocantheric femoral fractures treated at Karlstad’s Central Hospital. Method: This was an internal quality control designed as a retrospective cohort study. Data on patients treated for pertrocantheric or subtrocantheric femoral fractures were retrieved from the Swedish Fracture Registry. Tip-apex distances were measured on patients’ radiographic images. P-values for comparison of the two study periods were created using the chi2 test. Association between the tip-apex distance and reoperation rate was presented as odds ratio. Result: The reoperation rates were the same for both time periods. The reoperation rates of unstable fractures were lower for the study period after implementation of new routines. Neither of these results were statistically significant. There was also no statistically significant association between the tip-apex distance and reoperation rates. Conclusion: No statistically significant decrease in reoperation rates was seen after implementation of new routines. There was no association between the tip-apex distance and reoperation rates.
