Gondor e Zarado: valutazione degli effetti di nuovi coadiuvanti antideriva sull'attività degli erbicidi

L’irrigidimento del contesto regolamentare europeo dovuto all’attuale condizione di contaminazione diffusa dell’ambiente, riscontrata in Italia e in molti altri paesi europei, ha visto l’esigenza sempre più pressante di razionalizzare le dosi dei fitofarmaci utilizzati in agricoltura. Lo sviluppo e l’utilizzo di nuovi prodotti coadiuvanti come specifici antideriva per erbicidi, rappresenta in questo senso, un’importante risorsa su cui si inizia a fare affidamento. In Francia, per esempio, già da alcuni anni ci sono normative che obbligano l’utilizzo in agricoltura di tali prodotti, mentre in Italia non si hanno ancora direttive precise a riguardo. In tal contesto l’obiettivo principale di questa ricerca, effettuata in collaborazione con la ditta Intrachem, è stato quello di studiare alcune caratteristiche funzionali relative a due prodotti, che verranno lanciati a breve sul mercato, come specifici antideriva per erbicidi. In particolar modo è stato fatto uno studio per verificare se ed eventualmente come, questi coadiuvanti (Gondor e Zarado) possono influenzare l’attività del principio attivo a cui vengono aggiunti, apportando variazioni relative alla sua efficacia. Lo schema di lavoro seguito ha previsto una prima fase di saggio dove venivano effettuati test dose-risposta, utilizzando diversi erbicidi a diverse concentrazioni. I test sono stati effettuati su alcune malerbe mono e dicotiledoni. In ciascuna di queste prove è stata valutata e confrontata la percentuale di sopravvivenza e il peso dei sopravvissuti tra le tesi trattate. Le tesi prevedevano trattamenti con erbicida e trattamenti con erbicida più uno dei due coadiuvanti. Nella seconda fase si è effettuato un approfondimento sulle tesi che hanno mostrato i risultati più interessanti, per capirne possibilmente le basi fisiologiche. In particolare si è verificato se l’aggiunta dei due antideriva potesse determinare cambiamenti durante la fase di assorbimento e di traslocazione del principio attivo all’interno della piantina, utilizzando molecole radiomarcate con C14. Dai risultati ottenuti si è potuto evidenziare come l’aggiunta dei coadiuvanti possa rendere più efficace l’azione dell’erbicida nei casi in cui le infestanti non vengono completamente controllate dagli stessi (stadio vegetativo troppo avanzato e resistenza all’erbicida). Non è stato sempre verificato che ad un miglioramento dell’efficacia coincida un aumento dell’assorbimento e della traslocazione del principio attivo, all’interno della pianta. In conclusione si è potuto constatare che Gondor e Zarado oltre a svolgere la loro funzione antideriva, non influenzano negativamente l’efficacia dell’erbicida, salvo poche eccezioni, ma al contrario possono potenziarne l’azione, nelle situazioni “border line”.

Analisi ambientale della coltivazione di biomasse a scopo energetico con metodologia Life Cycle Assessment (LCA)

Lo scopo dello studio è un'analisi comparativa degli impatti ambientali, calcolati utilizzando la metodologia del Life Cycle Assessment, della fase agricola di 9 colture dedicate (lignocellulosiche, oleaginose e cereali) da biomassa, con diifferenti destinazioni energetiche (biocarburanti di I e II generazione ed energia elettrica). E' infine stata eseguita un'analisi "from cradle to grave" considerando anche le diverse tecnice di trasformazione possibili, con dati bibliografici. Sotto tutti i profili (impatto per ettaro, impatto per unità energetica generata, e impatto totale della filiera, risulta un netto vantaggio delle coltrue lignocellulosiche, e fra queste specialmente le poliennali.

Emerging contaminants in agricultural ecosystems: impact of selected pharmaceutical on water and soil ecology and pratical implications

Pharmaceuticals are useful tools to prevent and treat human and animal diseases. Following administration, a significant fraction of pharmaceuticals is excreted unaltered into faeces and urine and may enter the aquatic ecosystem and agricultural soil through irrigation with recycled water, constituting a significant source of emerging contaminants into the environment. Understanding major factors influencing their environmental fate is consequently needed to value the risk, reduce contamination, and set up bioremediation technologies. The antiviral drug Tamiflu (oseltamivir carboxylate, OC) has received recent attention due to the potential use as a first line defence against H5N1 and H1N1 influenza viruses. Research has shown that OC is not removed during conventional wastewater treatments, thus having the potential to enter surface water bodies. A series of laboratory experiments investigated the fate and the removal of OC in surface water systems in Italy and Japan and in a municipal wastewater treatment plant. A preliminary laboratory study investigated the persistence of the active antiviral drug in water samples from an irrigation canal in northern Italy (Canale Emiliano Romagnolo). After an initial rapid decrease, OC concentration slowly decreased during the remaining incubation period. Approximately 65% of the initial OC amount remained in water at the end of the 36-day incubation period. A negligible amount of OC was lost both from sterilized water and from sterilized water/sediment samples, suggesting a significant role of microbial degradation. Stimulating microbial processes by the addition of sediments resulted in reduced OC persistence. Presence of OC (1.5 μg mL-1) did not significantly affect the metabolic potential of the water microbial population, that was estimated by glyphosate and metolachlor mineralization. In contrast, OC caused an initial transient decrease in the size of the indigenous microbial population of water samples. A second laboratory study focused on basic processes governing the environmental fate of OC in surface water from two contrasting aquatic ecosystems of northern Italy, the River Po and the Venice Lagoon. Results of this study confirmed the potential of OC to persist in surface water. However, the addition of 5% of sediments resulted in rapid OC degradation. The estimated half-life of OC in water/sediment of the River Po was 15 days. After three weeks of incubation at 20 °C, more than 8% of 14C-OC evolved as 14CO2 from water/sediment samples of the River Po and Venice Lagoon. OC was moderately retained onto coarse sediments from the two sites. In water/sediment samples of the River Po and Venice Lagoon treated with 14C-OC, more than 30% of the 14C-residues remained water-extractable after three weeks of incubation. The low affinity of OC to sediments suggests that the presence of sediments would not reduce its bioavailability to microbial degradation. Another series of laboratory experiments investigated the fate and the removal of OC in two surface water ecosystems of Japan and in the municipal wastewater treatment plant of the city of Bologna, in Northern Italy. The persistence of OC in surface water ranged from non-detectable degradation to a half-life of 53 days. After 40 days, less than 3% of radiolabeled OC evolved as 14CO2. The presence of sediments (5%) led to a significant increase of OC degradation and of mineralization rates. A more intense mineralization was observed in samples of the wastewater treatment plant when applying a long incubation period (40 days). More precisely, 76% and 37% of the initial radioactivity applied as 14C-OC was recovered as 14CO2 from samples of the biological tank and effluent water, respectively. Two bacterial strains growing on OC as sole carbon source were isolated and used for its removal from synthetic medium and environmental samples, including surface water and wastewater. Inoculation of water and wastewater samples with the two OC-degrading strains showed that mineralization of OC was significantly higher in both inoculated water and wastewater, than in uninoculated controls. Denaturing gradient gel electrophoresis and quantitative PCR analysis showed that OC would not affect the microbial population of surface water and wastewater. The capacity of the ligninolytic fungus Phanerochaete chrysosporium to degrade a wide variety of environmentally persistent xenobiotics has been largely reported in literature. In a series of laboratory experiments, the efficiency of a formulation using P. chrysosporium was evaluated for the removal of selected pharmaceuticals from wastewater samples. Addition of the fungus to samples of the wastewater treatment plant of Bologna significantly increased (P < 0.05) the removal of OC and three antibiotics, erythromycin, sulfamethoxazole, and ciprofloxacin. Similar effects were also observed in effluent water. OC was the most persistent of the four pharmaceuticals. After 30 days of incubation, approximately two times more OC was removed in bioremediated samples than in controls. The highest removal efficiency of the formulation was observed with the antibiotic ciprofloxacin. The studies included environmental aspects of soil contamination with two emerging veterinary contaminants, such as doramectin and oxibendazole, wich are common parasitic treatments in cattle farms.

"Crop elicitation": approccio innovativo per la valorizzazione delle proprietà funzionali in leguminose da granella

Crop elicitation: innovative approach for the valorization of grain legume functional properties. In Italy grain legume cultivation has encountered a drastic decrease due to several causes (productive, economic, social). In this regard, studies aimed at the setting up of agronomic techniques able to guarantee high and constant in planta yields of health-promoting compounds may concur at re-launching legume production. In this context, 22 accessions of grain legumes (17 Phaseolus vulgaris, 3 Phaseolus coccineus, 1 Vigna unguiculata and 1 Glycine max genotypes) were screened with the aim of identifying genotypes rich in health beneficial phytochemicals (α-amylase inhibitors, α -glucosidase inhibitors, polyphenols) and with low anti-nutritional compounds (lectins). A wide variability was observed among investigated accessions. Four genotypes (Verdone, Kidney Cina, Roviotto and DG) showed a α -amylase inhibitory activity significantly higher (approximately 30% more) than all other tested accessions. The α -amylase inhibitory activity was not correlated neither with the protein nor with the polyphenol contents. Conversely, the α -glucosidase inhibitory activity was positively correlated with grain color and polyphenol content: dark-colored seeds had a mean inhibitory activity of 83.64 ± 22.07%, whereas light-colored seeds had mean values of 21.11 ± 9.36%. As regards the anti-nutritional compounds, out of all common bean accessions, only DG showed no erythro-agglutination activity (lectins). Preliminary experiments, performed in controlled environment, permitted to highlight that different germination conditions markedly affect the synthesis and accumulation of functional compounds in legume seedlings. Those findings were confirmed with field trials performed in two different locations (Bologna and Pisa), on two bean genotypes (Verdone and Zolfino), during the 2004-2005 cropping season. Results showed that the application of abiotic stresses (no fertilization and /or no irrigation) lead to a significant increase of flavonoids in grains, but a decrease (up to 50%) in legume yields was also observed. Crop elicitation, even if valuable for boosting health-promoting compound synthesis in crops, must necessary cope with economically acceptable crop yields.

Practical implications of aflatoxin contamination in corn

Aflatoxin (AFL) contamination of corn is a serious economic and food security issue. Although a variety of technical solutions for reducing AFL contamination of corn have been proposed, only a few have produced satisfactory results. A successful approach is a biocontrol strategy consisting of using non-flatoxigenic strains of Aspergillus flavus to replace indigenous AFL-producing isolates. The main objective of the present thesis was to investigate the dynamic and contamination of AFL/A. flavus in corn in Northern Italy. The study also investigated the role of the key-pest of corn, the European Corn Borer (ECB), on AFL contamination and dispersal of A. flavus propagules in corn. Finally, the study evaluated the feasibility of bioplastic-based granules entrapping a non-aflatoxigenic A. flavus strain for the biocontrol of this fungus in corn. The 2-year field study demonstrated the efficacy of the bioplastic formulation to reduce AFL contamination in corn. More precisely, although AFL contamination varied among the two years, application of 15 and 30 kg ha-1 of granules reduced AFL contamination to up 60 and 85% in 2009 and 2010 respectively. Microbiological analysis showed that the relative abundance of non-aflatoxigenic soil isolates significantly increased after 1 month from granules application (mid-May) and throughout the corn-growing season. These findings were consistent with data obtained using a bioplastic-based bait specifically developed to selectively isolate Aspergilli from soil and other environmental samples. In addition, field and laboratory evaluations showed that the level of damages produced by ECB larvae were not significantly correlated to A. flavus infestation and AFL contamination. Taking together, these findings demonstrated that AFL contamination of corn in Northern Italy was variable, but above the EU limit for human consumption. First proposed in the USA, this study showed the practical possibility of this formulation to be use for reducing AFL contamination in corn in the EU.

Isolamento, variabilità ed espressione del gene PIMT in Girasole / Isolation, variability and expression of PIMT gene in Sunflower

Il vigore è un aspetto rilevante della qualità delle sementi, strettamente connesso al loro status fisiologico, al genotipo e alle condizioni di stoccaggio. Stress ossidativi e danni alle macromolecole sono alla base del deterioramento del seme, che è equipaggiato con sistemi protettivi e di riparazione. Uno di questi coinvolge l’L-isoaspartil metiltransferasi (PIMT) che ripara i misfolding proteici catalizzando la riconversione in aspartato dell’isoaspartile anomalo accumulato. Scopo di questo studio era valutare il possibile ruolo del meccanismo di riparazione di PIMT nel vigore del seme in girasole. Per questo il relativo gene è stato isolato e caratterizzato, la variabilità allelica determinata su un campione di linee inbred e l’espressione genica misurata in risposta all’invecchiamento accelerato (aging) e al priming. La sequenza codificante ottenuta è costituita da 4 esoni e contiene i 5 domini caratteristici delle metiltransferasi. Il gene mostra elevata similarità con gli ortologhi vegetali e scarsa diversità nucleotidica nei genotipi coltivati rappresentativi della variabilità della specie. Nella sequenza aminoacidica, comunque, sono state rinvenute tre sostituzioni che potrebbero influenzare la funzionalità enzimatica. Dal punto di vista fisiologico i genotipi considerati hanno esibito notevole variabilità di risposte ai trattamenti, sia in termini di vigore che di espressione genica. Aging e priming hanno prodotto generalmente gli effetti attesi, rispettivamente negativi e positivi, sulla germinabilità e sulla sua velocità. In generale l’espressione di PIMT è risultata massima nel seme secco, come riportato altrove, e ridotta dall’aging. Anche il priming ha diminuito l’espressione rispetto al seme quiescente, mentre il suo effetto dopo l’aging è risultato genotipo-dipendente. Tuttavia, nelle condizioni descritte, non si sono evidenziate correlazioni significative tra vigore ed espressione di PIMT, tali da suggerire un chiaro ruolo di questo meccanismo nella qualità fisiologica del seme. / Vigour is a relevant aspect of seed quality which depends on genetic make-up, physiological status and enviromental conditions encountered during storage. Oxidative stress and damage to macromolecules is a main cause of deterioration in seeds which for this are equipped with antioxidant and repair mechanisms. One of these, involves L-isoaspartyl methyltransferase (PIMT) which counteracts protein misfolding resulting from L-isoaspartyl formation by catalyzing the conversion of abnormal residues to their regular form. The aim of this work was to investigate the possible role of the repair pathway of PIMT in relation to seed vigour in Helianthus annuus. For this purpose the PIMT gene was isolated and characterized, the allelic variability investigated on a panel of inbred lines, and its expression assessed in response to accelerated aging and priming treatments. The coding sequence of PIMT in suflower consists of 4 exons and contains the 5 conserved domains of metiltransferases. The gene shows a high similarity with its orthologs isolated from other plant species, and a rather low nucleotide diversity among cultivated accessions representative of a broad spectrum of genetic variability. At the peptide level, however, three aminoacidic substitutions were detected that could potentially affect enzyme activity. At the physiological level, different responses both in terms of vigour as well as of PIMT expression were observed between genotypes. Overall, aging and priming produced, as expected, negative and positive effects respectively, on germination percentage and rate. The PIMT transcripts reached maximum levels in quiescent seeds and minimum in aged seed, confirming previous findings. Priming also reduced gene expression compared to dry seed, but its effect after aging was strongly dependent on genotype. However, no evidence of correlation between vigour and PIMT expression was found that could suggest a clear role of this mechanism in seed vigour in the experimental conditions considered.

Methane production through anaerobic digestion of dedicated energy crops

Methane yield of ligno-cellulosic substrates (i.e. dedicated energy crops and agricultural residues) may be limited by their composition and structural features. Hence, biomass pre-treatments are envisaged to overcome this constraint. This thesis aimed at: i) assessing biomass and methane yield of dedicated energy crops; ii) evaluating the effects of hydrothermal pre-treatments on methane yield of Arundo; iii) investigating the effects of NaOH pre-treatments and iv) acid pre-treatments on chemical composition, physical structure and methane yield of two dedicated energy crops and one agricultural residue. Three multi-annual species (Arundo, Switchgrass and Sorghum Silk), three sorghum hybrids (Trudan Headless, B133 and S506) and a maize, as reference for AD, were studied in the frame of point i). Results exhibit the remarkable variation in biomass yield, chemical characteristics and potential methane yield. The six species alternative to maize deserve attention in view of a low need of external inputs but necessitate improvements in biodegradability. In the frame of point ii), Arundo was subjected to hydrothermal pre-treatments at different temperature, time and acid catalyst (with and without H2SO4). Pre-treatments determined a variable effect on methane yield: pre-treatments without acid catalyst achieved up to +23% CH4 output, while pre-treatments with H2SO4 catalyst incurred a methanogenic inhibition. Two biomass crops (Arundo and B133) and an agricultural residue (Barley straw) were subject to NaOH and acid pre-treatments, in the frame of point iii) and iv), respectively. Different pre-treatments determined a change of chemical and physical structure and an increase of methane yield: up to +30% and up to +62% CH4 output in Arundo with NaOH and acid pre-treatments, respectively. It is thereby demonstrated that pre-treatments can actually enhance biodegradability and subsequent CH4 output of ligno-cellulosic substrates, although pre-treatment viability needs to be evaluated at the level of full scale biogas plants in a perspective of profitable implementation.

Wheat and low-input agriculture: agronomic, nutritional and nutraceutical implications

Recently, the increasing interest in organic food products and environmental friendly practices has emphasized the importance of selecting crop varieties suitable for the low-input systems. Additionally, in recent years the relationship between diet and human health has gained much attention among consumers, favoring the investigations on food nutraceutical properties. Among cereals, wheat plays an important role in human nutrition around the world and contributes to the daily intake of essential nutrients such as starch and protein. Moreover, whole grain contains several bioactive compounds that confer to wheat-derived products unique nutraceutical properties (dietary fibre, antioxidants). The present research provided interesting insights for the selection of wheat genotypes suitable for low-input systems and the development of specific breeding programs dedicated to organic farming. The investigation involved 5 old not dwarf genotypes (Andriolo, Frassineto, Gentil rosso, Inallettabile, Verna) and 1 modern dwarf variety (Palesio), grown under biodynamic management, over two consecutive growing seasons (2009/2010, 2010/2011). Results evidenced that under low-input farming some investigated old wheat genotypes (Frassineto, Inallettabile) were comparable to the modern cultivar in terms of whole agronomic performance. As regards the nutritional and nutraceutical properties, some old genotypes (Andriolo, Gentil rosso, Verna) emerged for their relevant content of several investigated phytochemicals (such as insoluble dietary fibre, polyphenols, flavonoids, in vitro antioxidant activity) and nutrients (protein, lipid, minerals). Despite of the low technological features, the six wheat varieties grown under low-input management may efficiently provide raw material for the preparation of traditionally processed bread with valuable sensory and nutritional properties. Results highlighted that old wheat varieties have peculiar phytochemical composition and may be a valuable source of nutraceutical compounds. Some of the genetic material involved in the present study may be used in breeding programs aimed at selecting varieties suitable for low-input farming and rich in health-promoting compounds.
