Gewebsspezifische Genexpression wird zum größten Teil durch das Zusammenspiel einer Reihe von Transkriptionsfaktoren bewerkstelligt. Um die transkriptionelle Kontrolle muskelspezifischer Genexpression näher zu untersuchen, wurde ein Promoterfragment des procinen Skelettmuskel-Ryanodinrezeptors 1 analysiert (RyR1). Für dieses Promotorfragment konnte in früheren Untersuchungen Bindungsaktivität eines unbekannten Transkriptionsfaktors nachgewiesen werden. Die Anwendung molekularbiologischer und biochemischer Isolationsmethoden zeigte, daß die beschriebene Bindungsaktivität in Gesamtzellextrakten auf Interaktion eines unspezifischen Proteins beruht. Die Analyse von Skelettmuskel-Kernextrakten ergab keine spezifische Bindung eines Faktors im Retentionsgel. Allerdings ist nicht völlig auszuschließen, daß ein für die Expression von RyR1 benötigter Faktor mit diesem Promotorabschitt interagiert. Sehr geringe Konzentrationen oder entwicklungsspezifische Expressionsmuster wären durch di! e vorliegende Untersuchung nicht identifiziert worden. Weiters konnte über PCR ein im procinen Skelettmuskel stark exprimiertes Gen isoliert und charakterisiert werden. Bei der untersuchten cDNA handelt es sich um eine neue Variante des LIM Proteins FHL1 (Four and a half LIM Protein 1). Neben der cDNA wurden 14 kb genomischer Bereich sequenziert, der Transkriptionsstart mittels Primer Extension Analyse festgelegt und die chromosomale Lokalisation von FHL1C auf Chromosom Xq23-24 bestimmt. Eine hohe gewebsspezifische Expression im Skelettmuskel und im Gewebe von Blutgefäßen konnte über RT-PCR nachgewiesen werden.
