C2- and C3-symmetric ligands via ring-opening of aziridines

This thesis deals with the design and synthesis of chiralenantiopure nitrogencontaining ligands and the use of theseligands in asymmetric catalysis. A modular synthetic approachto enantiopure nitrogen-containing ligands was developed. Thesynthetic method is based on the ring-opening of activatedchiral aziridines by nitrogen nucleophiles. The aziridines areconveniently prepared from amino alcohols. The structure oftheaziridine and of the nucleophile can be extensively varied andlibraries of ligands are easily prepared. The use of primaryamines affords C2-symmetric bis(sulfonamides), whereas the use ofammonia affords C3-symmetric tris(sulfonamides) that can beelaborated into the corresponding tetra-amines. The C2- and C3-symmetric ligands were used in the asymmetrictitaniummediated addition of diethylzinc to benzaldehyderesulting in modest enantioselection, 76% ee. A thoroughinvestigation of the reaction conditions revealed that theamount of Ti(OiPr)4has a decisive effect on the reaction rate and thestereochemical outcome of the reaction. The reaction timedecreased from about 90 hours to 15 minutes and theenantioselectivity changed from 26% of the (R)- enantiomer to72% of the (S)-enantiomer when the Ti(OiPr)4:benzaldehyde ratio was increased from 0.125:1 to1.48:1. Moreover, the titanium-mediated addition of diethylzincto benzaldehyde was studied in the presence of chiraladditives. The bis(sulfonamides) were also used in thecyclopropanation of cinnamyl alcohol. However, only lowenantioselection was observed, 27% ee. The C3-symmetric tetra-amines were reacted to formazaphosphatranes. These weak acids were only partiallydeprotonated by the strong base KOtBu to form the correspondingproazaphosphatranes. The unexpectedly strong basicity of theproazaphosphatranes was believed to be due to steric effects assuggested by DFT calculations. In 1908, H. Emil Fisher attempted to prepare glycinal, an unprotected amino aldehyde, which he found to be inherently unstable and prone to polymerization. This instability arises from the propensity of amines to condense with aldehydes. Accordingly, amino aldehydes require protection of the amine functional group. On the contrary, aziridines do not condense with aldehydes; the aziridine ring-strain precludes the formation of an iminium ion. Predicated upon this orthogonal reactivity, a stable class of unprotected amino aldehydes has been prepared, and an in-depth investigation into their chemical reactivity has been undertaken. O anel aziridínico é conhecido desde o final do século XIX e apresenta alta reatividade devido a sua alta energia de ligação e tensão do anel, o que o torna um valioso aliado na síntese orgânica, especialmente em reações com nucleófilos. Estas características também o tornam reativo frente à nucleófilos biológicos, tais como os ácidos nucléicos. Tal fato leva a uma toxicidade celular e pode ser dirigida a substratos específicos, como no caso de quimioterápicos, levando à alquilação de DNA em células tumorais. Este anel pode ser sintetizado a partir de ?-aminoálcoois, cuja classe de compostos apresenta conhecidas atividades, presentes em importantes moléculas biologicamente ativas. Diante disto, realizouse neste trabalho um estudo a partir de um padrão estrutural envolvendo as duas classes mencionadas, aziridinas e ?-aminoálcoois, a fim de se identificar uma possível atividade antitumoral frente à enzima LSD1. Para este caso, se explora a similaridade estrutural entre aziridina e ciclopropano, presente na estrutura da tranilcipromina. Além disso, realizou-se o estudo deste padrão estrutural frente a dois outros alvos: células de câncer de mama e o parasita T.cruzi, com o intuito de expandir o conhecimento sobre a reatividade de tais compostos sobre estas células. O desenvolvimento desta biblioteca de compostos permitiu uma análise preliminar das relações entre a estrutura química e atividade biológica observada. Neste estudo, foi possível identificar aziridinas com atividade antumoral, mais potentes que cisplatina (IC50 abaixo de 40?M). O mecanismo de ação, por sua vez, ainda deverá ser estudado, considerando que os compostos não inibiram LSD1. / The aziridine ring is known since the end of the XIX century and presents high reactivity due to its high strain energy and ring tension, which makes it a valuable ally in organic synthesis, especially with nucleophiles. These characteristics also culminate in reactivity towards biological nucleophiles, such as nucleic acids. This fact leads to cellular toxicity and can be directed to specific sites, which is the case in chemotherapy agents, resulting in DNA alkylation in cancer cells. This ring can be obtained from ?-amino alcohols, which also present known activities and are present in important molecules with biological activity. As a result, this work performed a study of a structural pattern involving those two classes mentioned above, targeting a potential antitumoral activity towards the LSD1 enzyme. In this case, it is explored the structural similarity between the aziridine ring and the ciclopropane, present in the tranylcipromine structure. Furthermore, this structural pattern was studied for other two targets: breast cancer cells and the T.cruzi parasite, in order to expand the knowledge about the reactivity of those compounds in those biological systems. The development of this library allowed a preliminary analysis between the chemical structure and the observed biological activity. In this project it was possible to identify aziridines with antitumoral activities, showing higher potency than cisplatine (IC50 below 40?M). However, the mechanism is yet to be studied, considering that those compounds didn\'t inhibit the LSD1 enzyme.

Résumé Cette thèse consiste en trois thèmes résumés dans les paragraphes ci-dessous. L’influence de différents groupements protecteurs du groupe hydroxyle lors des réactions d’ouverture des cis- et trans- 3-hydroxy-1,2-époxycycloalcanes a été étudiée. Il a été montré que Yb(OTf)3 constituait un catalyseur doux pour l’ouverture régiosélective de cycles afin d’obtenir les -anilino cycloalcanols correspondants avec de bons rendements. Le chauffage du milieu réactionnel dans le toluène comme solvant a permis d’augmenter la cinétique de la réaction, au dépend du rendement. La partie aniline a été régiosélectivement introduite en position vicinale du groupe hydroxyle ou éther afin d’obtenir un unique régioisomère. La même tendance a été observée avec les époxydes du 3-azidocyclohex-1-ène et du 3-carbamate correspondant. Le temps de réaction a été réduit lorsque Yb(OTf)3 a été dissous dans l’acétonitrile. Le triflate d’ytterbium (III) a également été utilisé comme catalyseur pour l’ouverture de cycle régiosélective d’aziridines non-activées sur des cyclohexanes portant des substituants azotures ou éthers de benzyle. L’ion azoture ou l’aniline forment les produits trans correspondants, donnant alors accès à des diamines vicinales avec de bons rendements. Un éther ω-alcoxy p-méthoxybenzylique racémique, inhibiteur de HDAC, a été ainsi préparé en huit étapes synthétiques (rendement total de 26%) à partir du 1-((tert-butyldiphénylsilyl)oxy)hept-6-èn-2-ol. Ceci représente un progrès par rapport à la précédente méthode (9 étapes, rendement total de 16%). La métathèse croisée se montre particulièrement efficace et pratique dans cette stratégie et l’alkylation par le trichloroacétimidate de p-méthoxybenzyle en présence de Sc(OTf)3 améliore le rendement global de la synthèse. Un aminoalcool présent dans la pactamycine et contenant le squelette carboné, les groupements fonctionnels et la stéréochimie requise a été synthétisé en 27 étapes à partir de la L-thréonine. La méthodologie décrite dans cette thèse permet la synthèse de cet intermédiaire clé à l’échelle multigramme. / Abstract Ring-opening reactions of epoxides and aziridines have been extensively studied. This is an improvement over the published method (9 steps, 16% overall yield). The cross-metathesis method proved to be efficient and practical in this strategy, and alkylation using p-methoxybenzyl trichloroacetimidate in the presence of Sc(OTf)3 improved the overall yield of the synthesis. An amino alcohol that contains all the core carbons, functional groups and the required stereochemistry present in pactamycin was obtained starting from L-threonine over 27 steps. The methodology described in this thesis allows for a synthesis of this key intermediate on a multigram scale.

Síntese de um fragmento precursor do fármaco Indinavir / Synthesis of a precursor fragment of drug Indinavir

Neste trabalho foram aprofundados nossos estudos para obtenção da (S)-2-terc-butilamida-4-(3-picolil)piperazina, pela abertura da (S)-2-terc-butilcarboxamida-N-p-tosilaziridina seguida de ciclização, em 78% de rendimento, com o triflato de vinildifenilsulfônio. A aziridina foi preparada por um processo de ciclização, em condições de transferência de fase, partindo-se da L-serina, um aminoácido natural de baixo custo. Esta rota sintética rendeu um material que apresenta a mesma estereoquímica S do fragmento piperazínico usado na síntese do Indinavir, podendo vir a constituir uma via alternativa para a obtenção deste fármaco. / In this work we performed a deeper study for obtaining (S)-2-tert-butylamide-4-(3-picolyl)piperazine by opening (S)-2-tert-butylcarboxamide-N-p-tosylaziridine followed by cyclization, in 78% yield, with diphenylvinylsulfonium trifluoromethanesulfonate. The aziridine were prepared by a cyclization process in phase transfer conditions, starting from L-serine, a low cost amino acid. This synthetic route yielded a material which has the same S piperazinic fragment stereochemistry used in the synthesis of Indinavir, and may constitute an alternative route for obtaining this drug.
