Modification on glycosidic enzyons as strategy to alter tumor

Alterations in the expression of carbohydrates and the enzymes involved in their synthesis (glycosidases and glycosyltransferases) can be considered as universal feature of malignant transformation. Iminosugars, monosaccharide analogues compounds, have emerged as a new class of glycosidase inhibitors and have been suggested as therapeutic tools to inhibit tumour metastasis. The addition of an N-alkyl chain to iminosugars seems to increase their efficiency. We have test the cytotoxicity of D-fagomina and (2R,3S,4R,5S)-2-(hydroxymethyl)-5-methylpyrrolidine-3,4-diol and their N-alkyl derivatives. The N-alkyl derivative have shown an inhibition of α-L-fucosidases i α-D-glucosidases. Lewis antigens are fucosylated oligosaccharides carried by glycoproteins and glycolipids, synthesized by the sequential action of fucosyltransferases (FucT) and sialyltransferases (ST). Their aberrant expression such as the over expression of sialylated antigens (sLea and sLex) has been viewed as one of the underlying mechanisms for metastasis in different carcinomas, due to the interaction of those antigens with the E-selectin present on endothelial cells that mediate the extravasation of cancer cells. Clinical studies have described that the presence of chronic inflammation in the digestive tract increase the risk of carcinogenesis. Therefore we have described the regulation by pro-inflammatory cytokines (IL-1β and IL-6) of fucosyltransferases involved in Lewis antigen synthesis. In the other hand, we have studied FucT III and FucT V implications in the synthesis of Lewis antigens and how changes on Lewis antigens expression pattern alter the adhesion capacities of MKN45 cells. / Un tret característic del procés carcinogènic es l’alteració de l’expressió i estructura dels carbohidrats i dels enzims que els sintetitzen (glicosidases i glicosiltransferases). Els iminosucres, compostos anàlegs als monosacàrids, s’han descrit com a potents inhibidors de glicosidases pel que han estat proposats com a possible teràpia per inhibir la metàstasis. La incorporació d’una cadena N-alquilada als iminosucres sembla incrementar la seva eficàcia, nosaltres hem testat la citotoxicitat de la D-fagomina i del (2R,3S,4R,5S)-2-(hydroxymethyl)-5-methylpyrrolidine-3,4-diol i els seus derivats N-alquilat, aquest últims han mostrat una inhibició in vitro de la α-L-fucosidases i de la α-D-glucosidases. Els antígens Lewis són oligosacàrids presents en posicions terminals dels carbohidrats de glicoproteïnes i glicolípids, sintetitzats per l’acció seqüencial de fucosiltransferases (FucT) i sialiltransferases (ST). Canvis en l’expressió d’aquests antígens com la sobre-expressió dels antígens sialitzats (sLea i sLex) faciliten la metàstasis de les cèl•lules tumorals, degut principalment a la interacció entre aquest antígens i la E-selectina present a la superfície de les cèl•lules endotelials. Estudis clínics han demostrat que la inflamació crònica incrementa el risc de carcinogènesis en càncers de l’aparell digestiu. Pel que hem descrit la regulació per citoquines inflamatòries (IL-1β i IL-6) de les fucosiltransferases implicades en la síntesis dels antígens Lewis. Per altra banda hem estudiat la implicació de FucT III i FucT V en la síntesis dels antígens Lewis, i com aquest canvi en el patró d’expressió d’aquests antígens afecta la capacitat d’adhesió de les cèl•lules tumorals.

Cellular mRNA decay factors involved in the hepatitis C virus life cycle

The group of positive strand RNA ((+)RNA) viruses includes numerous plant, animal and human pathogens such as the hepatitis C virus (HCV). Their viral genomes mimic cellular mRNAs, however, besides acting as messengers for translation of viral proteins, they also act as templates for viral replication. Since these two functions are mutually exclusive, a key step in the replication of all (+) RNA viruses is the regulated exit of the genomic RNAs from the cellular translation machinery to the viral replication complexes. By using a model system that allows the replication of the plant (+) RNA Brome Mosaic Virus in yeast, it was shown by our group that the cellular decapping activators Dhh1, LSm1 and PatL1 play a key role in such transition. The LSm1 protein is a subunit of the LSm1-7 complexes. We have recently shown that these complexes interact directly with sequences in the BMV genome and that these interactions regulate the translation and replication of BMV. Interestingly, the LSm1 homolog in bacteria, Hfq, is required for the replication of the (+) RNA bacteriophage Qβ in E. coli.Since all (+)RNA viruses need to regulate the exit of the viral genome from the cellular translation machinery to replication and these proteins are conserved from yeast to humans, We hypothesized that the human homologues of Lsm1-7/Dhh1/Pat1 are required for the replication of human (+) RNA viruses. In this work we proved this hypothesis by showing that HCV translation and replication depend on these cellular proteins. Moreover, the requirement of these factors for efficient HCV RNA translation was linked exclusively to the 5´ and 3´ nontranslated regions (NTRs) of the viral genome. Furthermore, the LSm1-7 complex specifically interacts in vitro with essential cis-acting HCV RNA elements located in the NTRs. El grupo de virus de RNA de cadena de polaridad positiva ((+)RNA) incluye numerosos patógenos de plantas, animales y humanos, tales como el virus de la Hepatitis C (VHC). Sus genomas virales imitan a los RNAm celulares, sin embargo, además de actuar como mensajeros para la traducción de proteínas virales, también actúan como moldes para la replicación viral. Debido a que estas dos funciones son mutuamente excluyentes, un paso clave en la replicación de todos los virus (+) RNA es la salida regulada del RNA genómico desde la maquinaria de traducción celular hacia las complejos de replicación virales. Utilizando un sistema modelo que permite la replicación del virus del mosaico del bromo (VMB), un virus (+) RNA de plantas, en levaduras, nuestro grupo ha demostrado que los activadores de decapping celulares Dhh1, LSm1 y Pat1 juegan un papel clave en este paso. La proteína LSm1 es una subunidad del complejo celular LSm1-7. Hemos demostrado recientemente que dichos complejos interactúan directamente con secuencias específicas en el genoma del VMB y que dichas interacciones regulan la traducción y replicación del VMB. De manera interesante, la proteína homologa de LSm1 en bacterias, Hfq, es necesaria para la replicación del bacteriófago (+) RNA Qβ en E. coli.Debido a que todos los virus (+) RNA necesitan regular la salida del genoma viral desde la maquinaria traduccional de la célula hacia los complejos de replicación virales, y que las proteínas Dhh1, LSm1-7 y Pat1 están conservadas de levaduras a humanos, nuestra hipótesis es que lo homólogos humanos de Lsm1-7/Dhh1/Pat1 son requeridos para la replicación de virus (+)RNA de humanos. En este trabajo, hemos demostrado esta hipótesis mostrando que la traducción y la replicación del virus de la hepatitis C (VHC) dependen de estas proteínas celulares. Más aún, el requerimiento de estos factores para una eficiente traducción del genoma del VHC está vinculado de manera exclusiva a las regiones no traducidas (RNT) 5´ y 3´ del genome viral. Además, el complejo cellular LSm1-7 interactúa in vitro de manera específica con elementos esenciales del RNA del VHC que actúan en cis localizados en las RNTs.

Functional analysis of JmjC+N histone demethylases in Drosophila melanogaster

La metilació de les lisines de les histones està implicada en les funcions associades a la cromatina, com l’expressió gènica o la formació d’heterocromatina. És una modificació covalent afegida per metiltransferases d’histones i eliminada per desmetilases d’histones, que han estat descobertes recentment. Hem caracteritzat els grups de desmetilases que contenen dominis Jumonji C i Jumonji N a l’organisme model Drosophila melanogaster. Observem que les dues dKDM4 actuen sobre H3K36me3 i H3K9me3 i que LID/dKDM5 desmetila H3K4me3. La distribució d’HP1a es veu afectada per la sobreexpressió de dKDM4A. A més, hem analitzat la localització de LID/dKDM5 als discos imaginals d’ala i quins efectes té LID sobre l’expressió gènica i la trimetilació de H3K4 en aquest teixit. Hem observat que LID es troba als llocs d’inici de la transcripció de gens actius relacionats amb diferenciació i desenvolupament. Els gens diana de LID contenen H3K4me3 i H3K36me3 i RNAPII fosforilada a la serina 5 i a la serina 2. Una disminució de lid provoca una lleu baixada en l’expressió dels seus gens diana, la qual cosa suggereix que la desmetilasa té un paper en l’ajust dels nivells d’expressió. / Histone lysine methylation is involved in chromatin related cell functions, such as gene expression or heterochromatin formation. It is a covalent modification added by histone methyltransferases and removed by histone demethylases, discovered in the recent years. We characterized the groups of demethylases that contain both Jumonji C and Jumonji N domains in the model organism 2 Drosophila melanogaster. We show that dKDM4s act on H3K36me3 and H3K9me3 and LID/dKDM5 targets H3K4me3. HP1a distribution is affected by dKDM4A overexpression. In addition, we analyzed LID/dKDM5 localization in wing imaginal discs and how LID affects gene expression and trimethylation of H3K4 in this tissue. We observed that LID localizes at the transcription start sites of active genes related to development and differentiation. LID target genes contain H3K4me3 and H3K36me3 and RNAPII, phosphorylated at serine 5 and serine 2. Downregulation of lid produces a weak decrease in the expression of LID targets, suggesting a role of the demethylase in fine-tuning expression levels.

Participación del sistema opioide endógeno en la adicción a la nicotina

El tabaquismo es una enfermedad crónica y recurrente caracterizada por la necesidad imperiosa o compulsiva de consumir nicotina para experimentar la sensación de placer característica de su uso, a pesar de las consecuencias negativas de su consumo. La adicción a la nicotina es un proceso en el que están implicados diferentes sistemas de neurotransmisión, entre ellos el sistema opioide endógeno. Dicho sistema está compuesto por una serie de familias de péptidos opioides que actúan sobre tres tipos de receptores denominados receptores opioides. Este sistema está implicado, entre otras respuestas, en el control del dolor así como en la motivación y el refuerzo inducido por las drogas de abuso.El objetivo de la presente Tesis Doctoral fue determinar la implicación del sistema opioide endógeno en las propiedades adictivas de la nicotina. Los estudios que hemos realizado demuestran que los diferentes componentes del sistema opioide endógeno participan en las respuestas comportamentales de la nicotina, principalmente aquellas relacionadas con la adicción. Así, tanto los péptidos opioides derivados de la proencefalina, como los receptores opioides δ, modulan los efectos reforzantes de la droga mientras que los péptidos opioides derivados de la prodinorfina intervienen en los aspectos aversivos y/o disfóricos de la misma. Además, es necesaria la participación de las encefalinas endógenas y de los receptores opioide μ para que la nicotina induzca sus efectos antinociceptivos agudos y los fenómenos de dependencia física. Los receptores opioides μ y κ modulan de manera opuesta la tolerancia a los efectos antinociceptivos inducidos por la nicotina.Nuestros estudios revelan que los efectos adictivos de la nicotina están mediados por el sistema opioide endógeno. Con nuestros datos aportamos una perspectiva para el desarrollo de compuestos que podrían ser utilizados como una alternativa terapéutica en el fenómeno de la adicción a la nicotina. / Tabaquism is a chronic and recurrent disease that is characterized by the overwhelming and compulsive need for nicotine consumption. Nicotine addiction involves different neurotransmission systems. Among them, the Endogenous Opioid System is postulated to play an important role in the nicotine addiction process, although the specific mechanisms by which this role may be exerted have not been clearly elucidated yet. The Endogenous Opioid System comprises a series of families of opioid peptides that bind to three different types of opioid receptors: μ, δ and κ opioids receptors. Among other features in the nervous system, the Endogenous Opioid System has been related to the motivation and reinforcing properties of drugs of abuse as well as pain control. The aim of the present PhD Thesis has been to dissect the role of the Endogenous Opioid system in the addictive properties of nicotine. Our results demonstrate that different components of the endogenous opioid system are involved in the behavioral responses induced by nicotine, and very importantly in those responses related to nicotine addiction. Thus, both opioids derived from pro-enkephalin and δ-opioids receptors modulate the reinforcing effects of nicotine whereas opioid peptides derived from pro-dinorphin participate in the aversive and/or disphoric effects of this drug. Furthermore, as we have observed the participation of the endogenous enkephanins and µ-opioid receptors are necessary in order to experience both nicotine dependence and acute antinociceptive effects. The μ and κ opioid receptors are involved in different manners in the antinociceptive tolerance of nicotineThis study shows that the addictive effects of nicotine are mediated by the endogenous opioid system. In addition, our data provides a new perspective that might be used as a therapeutic alternative for the development of new drugs targeting nicotine addiction phenomenon.

Biochemoinformatics: integrative computational tools at the interface between chemistry and biology

The work on this thesis has focused on the development of integrated databases and visualization tools for three related areas of biomedical sciences involved in the drug discovery process. An annotated chemical library including drugs and other bioactive compounds with reported affinities over protein targets has been extended and optimized for its use in different knowledge based analysis and virtual profiling methodologies. A second database relating drugs with their side effects has also been developed in preparation for future studies. At the same time, two web based visualization architectures have been designed and implemented as tools for the analysis of relevant data. / Aquesta tesi ha comprès el desenvolupament de bases de dades i eines de visualització relacionades amb tres àrees de la biomedicina implicades en el procés de disseny de fàrmacs. S'ha estès i optimitzat una llibreria química que inclou fàrmacs i altres compostos bioactius anotats a proteïnes diana pel seu ús en diferents anàlisis i metodologies de perfilat virtual. També s'ha desenvolupat una segona base de dades relacionant fàrmacs comercialitzats amb els seus efectes adversos com a base per a futurs estudis. A l'hora han estat dissenyades i implementades dues eines web de visualització d'informació rellevant en aquest camp.

Molecular mechanisms underlying the effects of cannabinoids in the brain

The endocannabinoid system is an endogenous neuromodulatory system that regulates a plethora of physiological functions, including the control of movement, memory, anxiety, and pain, among others. Cannabinoid compounds are mainly found in the Cannabis sativa plant and exert their effects by acting at the endocannabinoid system. Cannabinoids are potential therapeutic agents, mainly for multiple sclerosis, pain, and emesis, although an important caveat to their use is the possible adverse effects, such as memory impairment and anxiety. This thesis mainly addresses the molecular mechanisms underlying some of the physiological processes controlled by the endocannabinoid system as well as specific pharmacological effects triggered by 9-tetrahydrocannabinol, the main psychoactive compound of marijuana plant. The combination of biochemical, pharmacological, and behavioral approaches allowed the elucidation of certain signaling cascades responsible for particular effects induced by cannabinoids. / El sistema endocannabinoid és un sistema neuromodulador endogen que regula diverses funcions fisiològiques, incloent el control del moviment, la memòria, l’ansietat i el dolor, entre altres. Els compostos cannabinoids es troben principalment a la planta Cannabis sativa i exerceixen els seus efectes actuant al sistema endocannabinoid. Els cannabinoids tenen potencial terapèutic, principalment per l’esclerosi múltiple, el dolor i l’èmesi, tot i que una limitació important pel seu ús recau en els possibles efectes adversos, tal com l’alteració de la memòria i l’ansietat. Aquesta tesi exposa principalment els mecanismes moleculars responsables d’alguns processos fisiològics controlats pel sistema endocannabinoid així com efectes farmacològics desencadenats pel 9-tetrahidrocannabinol, el principal compost psicoactiu de la planta de marihuana. La combinació d’aproximacions bioquímiques, farmacològiques i comportamentals ha permès revelar algunes cascades de senyalització responsables de determinats efectes induïts pels cannabinoids.

Qualitative and quantitative study of the effect of osmotress on cell cycle of Saccharomyces cerevisiae

Control of cell cycle by Stress Activated Protein Kinases (SAPKs) is an essential aspect for adaptation to extracellular stimuli. In Saccharomyces cerevisiae, the activation of the Hog1 SAPK, results in a delayed transcription of the G1 cyclins CLN1,2 and the stabilization of the B-type cyclin inhibitor SIC1, therefore postponing entry into S phase. The results displayed here, show, by a combination of mathematical modelling and quantitative in vivo experiments, that, before Start, the control of G1-S transition is mainly exerted by inhibiting expression of cyclins, both G1 (CLN1,2) and S phase (CLB5,6) cyclins. On the other hand, after Start, it is the phosphorylation and stabilization of Sic1 by Hog1 that becomes imperative to prevent inadequate firing of replication before adaptation. Therefore, we found that there is a distinct temporal role for Sic1 and cyclins on the G1 regulation by a SAPK in response to stress. We have also found that Hog1 induces a G2 delay, by down-regulating CLB2 transcription and phosphorylating Hsl1 to promote Hsl7 delocalization and subsequent accumulation of Swe1, an inhibitor of Clb1,2-Cdc28, and thus postponing anaphase. Altogether, we demonstrate novel Systems Biology approaches are useful to better understand how an intracellular signalling pathway incises on cell cycle control, beyond a mechanistic description, as well as showing how a single MAPK modulates different cell cycle checkpoints to improve cell survival upon stress. / El control del cicle cel·lular per Proteïna Cinases Activades per Estrès (SAPKs) es un aspecte essencial per a l'adaptació als estímuls extracel·lulars. A Saccharomyces cerevisiae, l'activació de la SAPK Hog1, resulta en un retardament de la transcripció de les ciclines de G1 (CLN1,2) i l'estabilització del inhibidor de les ciclines del tipus B, SIC1, i per tant posposa l'entrada en fase S. Els resultats que aquí s'exposen, mostren, mitjançant la combinació de modelatge matemàtic i experiments quantitatius in vivo, que, abans d'Start, el control de la transició es duu a terme principalment inhibint l'expressió de les ciclines, tant les de G1 (CLN1,2) com les de la fase S (CLB5,6). Per altra banda, després d'Start, la fosforilació i estabilització de Sic1 per part de Hog1 esdevé un fet necessari per a prevenir la iniciació de la replicació abans d'adaptar-se. Per tant, hem descobert aquí que la regulació de Sic1 i les ciclines juguen un paper diferent segons el moment en que apareix l'estrès. Hem descrit també, que Hog1 produeix una parada a G2 a través de la inhibició de la transcripció de CLB2 i la fosforilació d'Hsl1, la qual promou la deslocalització d'Hsl7 i la subsegüent estabilització de Swe1, un inhibidor específic de Clb1,2-Cdc28, i d'aquesta manera es posposa l'entrada en Anafase. Tot plegat, demostra que l'ús d'aproximacions pròpies de la Biologia de Sistemes és útil per a entendre de quina forma una via de senyalització intracel·lular incideix sobre el control del cicle cel·lular, mes enllà de la pura descripció de la mecànica del sistema. D'aquesta forma, proposem que una sola MAPK modula distints punts de control del cicle cel·lular per millorar la probabilitat de supervivència en front de l'estrès osmòtic.

Além disso, foram realizados testes visando-se determinar a sobrevivência dos ácaros adultos, quando submetidos as condições adversas de umidade e alimentação. Com esse objetivo foram definidas três épocas denominando-se a primeira outono-inverno (20/05/90 a(26/08/90), a segunda inverno (26/05/90 a 20/09/90 e a terceira verão (15/11/90 a 10/01/91) obtendo-se para os diversos parâmetros biológicos os seguiintes valores respectivamente : período de incubação 9,57,13,51 E 7,38 dias de estágio larval 5,20,4,10 e 2,72 dias estágios nifais 26,62,30,65 e 11,32 dias, período ovo-adulto 40,33,45,40 e 21,44 dias , períodos de pré-oviposição 4,44,10,50 e 3,59 dias, número de ovos/fêmea/dia 0,47, 0,39 e 0,61 e longevidade 47,25,53,75 e 34,57 dias. No verão sob condições não controladas de laboratótio os valores obtidos foram os seguintes : período de incubação, 5,98 dias; estágio larval 2,47 dias;fases ninfais 9,20 dias, período ovoadulto 17,81 dias;periodo de pré-oviposição 1,71 dias; número de ovos;fêmea/dia 1,37 e longevidadse 23,12 dias. Neste contexto, a grande variação das condições climáticas, mostrou exercer uma considerável influência sobre o ciclo biológico do ácaro. Com relação aos testes de sobrevivência, verificou-se para a primeira situação, ou seja, ausência água e alimento, que 72 horas após o inicio do teste cerca de 10% dos ácaros estava,m, mortos, atingindo o máximo de mortalidade (95%) após 144 horas,... / The biology of Brevipalpus phoenicis ((Geijskes,1939) (Acari: Tenuipalpidae ) was studied at three different periods of the year to evaluate possible alterations in this development compared to results obtained under laboratory conditions by in previous studies. The fallowing values were obtained for the biological parameters at thorn periods, respectively: incubation period 9.75,15.51 and 7.38 days, larval stage 11.32 days, egg to adult 40.33,45.40 and 21.44 days; pre-oviposition 4.44, 10.50 and 3.59 days;daily oviposition rate 0.47, 0.39 and 0.61 and longevity 47.25,53.75 and 34.57 days.
