Terapia fotodinâmica sobre carcinoma mamário de cadela cultivado in vitro

Conseqüentemente é crescente a busca por alternativas terapêuticas que cessem ou retardem o crescimento tumoral. Uma terapia promissora, difundida para o tratamento de alguns tipos de é a terapia fotodinâmica que se baseia na combinação de um fármaco fotossensibilizante que quando ativado provoca morte celular. O objetivo do trabalho foi avaliar a aplicação e citotoxicidade da Terapia Fotofinâmica mediada pelo Alumínio-Cloro-Ftalocianina em formulação liposomal (TFD-AlClFt) em um sistema de tratamento experimental in vitro de amostras tumorais extraídas de carcinoma mamário de cadela submetida ao tratamento cirúrgico tradicional. Para isso foram formados 04 grupos experimentais: grupo controle, LED, fármaco fotossensibilizador AlClFt 2,5μM e TFD. O grupo O controle (1) foi aquele em que as células não foram submetidas a nenhum tratamento; O grupo LED (2) recebeu iluminação com a fonte de luz LED na intensidade 10 J/cm2 e comprimento de onda 660nm. O grupo AlClFt (3) apenas administração do fármaco AlClFt e grupo TFD (4), que consistiu na terapia fotodinâmica. Foram analisados viabilidade celular, citotoxicidade, via de morte celular e possíveis alterações morfológicas através dos ensaios ciitotóxicos; azul tripan, dupla coloração de alaranjado de acridina e brometo de etídio, MTT e avaliações morfológicas que evidenciaram efetividade da TFD nas condições experimentais deste trabalho. _______________________________________________________________________________ ABSTRACT / Breast cancer is the most common malignant neoplasm in dogs and treatment currently consists of surgical excision, with or without adjuvant chemotherapy. However, the search for alternative therapies to stop completely or to slow down tumor growth is increasing. Photodynamic therapy (PDT) is based on the combination of a photosensitizing agent that once activated causes cell death. The objective of this study was to evaluate PDT mediated by Aluminum-Chlorine-Phtalocyanine (AlClPc) in an experimental in vitro treatment system of tumor samples taken from mammary carcinoma of dogs subjected to surgical excision . The primary cell culture was divided into four experimental groups: control, LED, photosensitizer AlClPc 2.5 μM and PDT. The control group (1) represented the cells which had not undergone any treatment, the LED group (2) was exposed to a LED light source of 10 J/cm2 intensity and wavelength of 660nm. The AlClPc group (3) was treated with only the photosensitizer agent and the PDT group (4), was the one that undergone the photodynamic therapy. Cell viability, cytotoxicity and cell death pathways were evaluated through the cytotoxicity assays: trypan blue, double staining of acridine orange and ethidium bromide and MTT. Cultures were also subjected to morphological evaluation by optic microscopy. It can be observed from the results that the amount of cell death was significantly higher (P <0.05) in the PDT group when compared to the other groups by all tests, and the cell death was due to necrosis. It can be concluded that the PDT is an effective treatment in cell culture of canine mammary carcinoma.
