Cellulose is the most abundant polysaccharide on earth and it is relatively a simple homopolymer with three hydroxyl groups, differing only subtly in reactivity. The position of substitution has a powerful influence on physical properties of cellulose derivatives. To better understand the structure and property relationships of cellulose derivatives, it is critical to have all homopolymers related to important cellulose ethers and esters available. However, regiocontrol in cellulose chemistry is still a difficult, mostly unconquered frontier.
In this dissertation, the main objective is to develop novel synthetic methods to synthesize regioselectively substituted cellulose derivatives including cellulose ethers and esters, and apply advanced characterization tools to understand structure and its influence on properties, which will give us deep insights into the composition of more random commercial derivatives, maximizing the content of advantageous monosaccharides. Several strategies to regioselectively synthesize cellulose derivatives are discussed in detail. The obtained regioselective cellulose derivatives were fully characterized analytically. Structure-property relationships of these regioselectively substituted cellulose derivatives were also studied. / Ph. D.

Regioselective Synthesis of Novel Cellulose Derivatives for Drug DeliveryFox, Stephen Carter 30 November 2011 (has links)
New methods were developed for the regioselective synthesis of new classes of cellulose derivatives with properties that could help improve the delivery of pharmaceutical drugs within the human body. The specific synthetic targets of this research were regioselectively carboxylated and regioselectively aminated cellulose derivatives. While different avenues to the carboxylated cellulose were ultimately explored without success, a new method for the synthesis of selectively <i>O</i>-acylated 6-amino-6-deoxy-cellulose esters was devised.
A key reaction that enabled the synthesis of the new cellulose derivatives described in this dissertation was the one-pot conversion of microcrystalline cellulose to 6-bromo-6-deoxy-cellulose esters. This reaction resulted in the highly selective displacement of the primary hydroxyl groups attached to the 6-carbon (<i>C</i>-6) on each anhydroglucose unit (AGU) in cellulose with bromide, with little or no bromination occurring at carbons 2 and 3 (<i>C</i>-2 and <i>C</i>-3). The brominated cellulose was then completely esterified by adding acetic, propionoic, or butyric acid anhydride to the reaction solution. The reaction products were readily soluble in many common organic solvents, including acetone, dimethyl sulfoxide, dimethylformamide, tetrahydrofuran, and chloroform. It was shown that the bromides could be converted to iodides under Finkelstein reaction conditions.
The presence of halides at <i>C</i>-6 allows a variety of new functional groups to be regioselectively introduced to cellulose via nucleophilic substitution. In one case, the 6-bromo-6-deoxy-cellulose esters were reacted with sodium cyanide to produce regioselectively synthesized cellulose nitriles. These compounds were synthesized with the idea that they could be converted to regioselectively carboxylated cellulose derivatives as an alternative pathway to the rhodium-catalyzed carbonyl insertion reactions also attempted in this research. However, the cellulose nitriles were highly susceptible to alkaline degradation, and conversion to the carboxylated cellulose was not achieved.
The 6-bromo-6-deoxy-cellulose esters were also reacted with sodium azide to successfully produce 6-azido-6-deoxy-cellulose esters. The azide groups were then reduced to amines using the Staudinger reaction. This very mild and selective reaction allowed the conversion of the azides to amines in the presence of the ester groups still attached to the cellulose backbone. Such derivatives could have properties useful for a range of biomedical applications, including the delivery of anionic drugs. / Ph. D.

Entretanto, na literatura, há divergências se ocorre realmente uma interação entre um polímero não iônico, como os derivados aquo-solúveis de celulose, em conjunto com tensoativos catiônicos. Desta forma, neste projeto de Mestrado, foram investigadas as interações entre três derivados de celulose (2-hidroxietil celulose, 2-HEC; hidroxipropil celulose, HPC e hidroximetilpropil celulose, HMPC) e três tensoativos catiônicos (cloreto de benzil-hexadecildimetilamônio, CBz; cloreto de fenil-hexadecildimetilamônio, Fenil e cloreto de 2-feniletil hexadecildimetilamônio, 2-Feniletil) obtendo-se informações sobre a estrutura dos agregados formados por este sistema através de medidas de condutividade, fluorescência no estado estacionário e viscosidade. Estas técnicas forneceram os parâmetros de concentração de agregação crítica (cac), concentração micelar crítica (cmc), grau de dissociação micelar e viscosidade das soluções. Em uma segunda etapa, foi investigada a variação da cmc do tensoativo zwitteriônico, coco amido propil betaína (CAPB), em soluções aquosas com diferentes pHs, visando futuros estudos de interação deste compostos com os biopolímeros derivados ou não de celulose. / There are many industrial formulations that are consisted of a biopolymer, such as cellulose derivatives, and surfactants. Synthesis and Characterization of Drug-Containing, Polysaccharide-Based Nanoparticles for Applications in Oral Drug DeliveryMazumder, Sonal 08 August 2013 (has links)
Amorphous solid dispersions of polysaccharide-drug nanoparticles were produced by a rapid precipitation process known as flash nanoprecipitation and the formulation process and properties of nanoparticles were investigated. In this thesis, several novel cellulose derivatives and a pullulan derivative were studied. Among these polymers, carboxymethyl cellulose acetate butyrate (CMCAB)-drug nanoparticles were investigated in detail. Previous work has shown that the presence of different chemical groups in CMCAB could aid in complexation with hydrophobic drugs with low solubility, forming an amorphous matrix which can increase the effective solubility and, hence, bioavailability of the drug in physiological conditions. An antibacterial drug and two less soluble anti-viral drugs were selected as model drugs for this study. A separate study was conducted with several other cellulose derivatives like cellulose acetate propionate adipates with two different degree of substitution 0.33 and 0.85 (CAP-Adp 0.33 and CAP-Adp 0.85), cellulose acetate sebacate (CA-320S Se) and butyl pullulan-6-carboxylate (BPC) polymers. The effect of polymer interaction with drug molecule on release of antiviral drugs was studied with these latter polymers.
The purpose of this research was two-fold. First, the methodology for producing drug-polymer nanoparticles with well-defined particle size distributions was developed. Second, the factors affecting drug loading and release properties of these nanoparticles were investigated. The nanoparticles were processed using two methods of solvent removal and drying to investigate their effects on drug loading and particle size: (a) various combinations of rotary vacuum evaporation (rotavap) and acid-induced flocculation were used and (b), dialysis followed by freeze drying. Dynamic light scattering showed particle sizes were between 150-400 nm with polydispersity index values as low as 0.12. The antibiotic drug loading efficiencies ranged from 14-40%, whereas drug loading efficiency as high as 85 % was attained with the antiviral drug. The dissolution studies showed an increase in the solution concentration and release of the amorphous drug nanoparticles. The high glass transition temperature helped to stabilize the drug in an amorphous form, thus increasing the effective solution concentration of the drug in an aqueous medium. / Ph. D.

Esta tese apresenta os resultados da obtenção de eletrólitos sólidos poliméricos a partir dos derivados de celulose - hidroxipropilcelulose (HPC) e hidroxietilcelulose (HEC), ambas comerciais. Para atingir os objetivos do projeto, os dois derivados passaram por diferentes processos sendo que a HEC foi modificada fisicamente por meio de plastificação com glicerol e HPC foi alterada quimicamente. A transformação química consistiu nas reações de oxidação de grupos hidroxila da HPC em grupos cetona que, em seguida, foram submetidos as reações de enxertia com diamina de poli(óxido de propileno) [Jeffamina] resultando em redes por meio de ligações imina. A adição do sal perclorato de lítio, em diferentes concentrações, na matriz plastificada ou entrecruzada, resultou na obtenção de eletrólitos sólidos poliméricos, todos na forma de filmes. A caracterização destes eletrólitos foi realizada com técnicas básicas de caracterização de materiais como: análises térmicas (DSC, TGA), análise térmica dinâmico-mecânica (DMTA), análises estruturais (raios-X), medidas espectroscópicas (IR, UVNIS), análise elementar, microscopia eletrônica de varredura (SEM), e, como a mais importante, medidas de condutividade iônica utilizando a técnica de espectroscopia de impedância eletroquímica (EIS). / The present thesis reports the preparation and characterization of new types of solid polymeric electrolytes (SPE) based on cellulose derivatives such as hydroxypropylcellulose (HPC) and hydroxyethylcellulose (HEC), both commercial products. The rheological properties of the carboxymethylcellulose (CMC) – water, hydroxyethyl cellulose (HEC) – DMF and hydroxypropyl cellulose (GPC) – DMSO and GPC – ethanol have been investigated in the dynamic conditions with a frequency scan and in a stationary shear. The concentration dependences of activation enthalpy of viscous flow for HEC and CMC solutions were obtained. It is established that the nature of GPC solutions varies with the increase in concentration from Newtonian to non-Newtonian: the stationary and dynamic viscosity of fluids decrease when the shear rate and frequency increase. Solutions of the GEC are viscoelastic liquids the concentration larger a certain one. For the first time the calculations of the activation parameters of the solution flow from data obtained by essentially different methods are compared. It is shown that the magnitude and the nature of the activation enthalpy of viscous flow are determined by the phase composition of the system, i.e. the dependence of the activation enthalpy on the concentration allows to determine the boundaries of the "corridor" in which the isotropic and anisotropic (LC) phase coexist. The concentration dependences of the dynamic modules are correlated with those for the activation enthalpy of the flow. / В работе исследованы реологические свойства растворов карбоксиметилцеллюлозы в воде, гидроксиэтилцеллюлозы в ДМФА и гидроксипропилцеллюлозы в ДМСО и этаноле в динамическом режиме с разверткой по частоте и в режиме стационарного сдвига. Построены концентрационные зависимости энтальпии активации вязкого течения для растворов ГЭЦ и КМЦ. Установлено, что характер течения растворов ГПЦ меняется с ростом концентрации с ньютоновского на неньютоновский: стационарная и динамическая вязкости растворов уменьшаются с увеличением скорости сдвига и с ростом частоты. Растворы ГПЦ выше определенной концентрации являются вязкоупругими жидкостями. Впервые проведено сопоставление расчетов активационных параметров течения для растворов из данных, полученных принципиально различными методами. Показано, что величина и характер изменения энтальпии активации вязкого течения определяются фазовым составом системы, т.е. зависимость энтальпии активации от концентрации позволяет определять границы «коридора», в котором сосуществуют изотропная и анизотропная (ЖК) фазы. Концентрационные зависимости динамических модулей коррелируют с таковыми для энтальпии активации течения.

O processamento da cana gera vários co-produtos tais como, o bagaço e a palha, sendo esta considerada um dos principais resíduos gerados devido à mecanização da colheita. Assim, a disponibilidade e a composição desse resíduo de cana-de-açúcar têm impulsionado muitas pesquisas para o desenvolvimento de tecnologias que proporcionem o seu aproveitamento efetivo. Este trabalho teve como objetivo a separação dos principais componentes da palha de cana-de-açúcar para a obtenção de insumos químicos com maior valor econômico, sendo proposta a obtenção, caracterização e modificação da celulose desta biomassa vegetal, a ser utilizada na síntese da carboximetilcelulose (CMC) partindo da celulose branqueada por meio da combinação do processo químico com o enzimático, utilizando xilanase. Foi também avaliada a influência nas propriedades mecânicas, termogravimétricas e morfológicas das poliuretanas, ao adicionar-se celulose a esses polímeros obtidos a partir de óleo de mamona (Ricinus communis L.). A palha de cana foi pré-tratada por explosão a vapor e submetida à deslignificação alcalina. Foi realizado, posteriormente, o tratamento utilizando-se xilanase e então, a polpa foi branqueada com peróxido de hidrogênio. A polpa branqueada obtida foi utilizada para obtenção de CMC. Foi determinado o ganho de massa após a reação de obtenção e o grau de substituição da CMC obtida. A polpa deslignificada (celulose) foi utilizada como reforço para obtenção de biocompósitos com poliuretana obtida a partir do óleo de mamona. Os biocompósitos obtidos foram analisados por calorimetria exploratória diferencial (DSC), termogravimetria (TGA), foram determinadas as propriedades mecânicas, microscopia eletrônica de varredura e estudo de absorção de água. A adição de fibras de palha em matriz de PU de óleo de mamona melhorou as propriedades mecânicas comparado à matriz pura, e uma possível redução no custo sugere uma aplicação industrial do produto final. / The sugarcane processing generates various agricultural byproducts such as bagasse and straw, which is considered one of the main waste generated due to the mechanization of the harvest in Brazil. Thus, the availability and composition of sugarcane straw have driven a great effort to develop technologies that provide its effective utilization. This study aimed to separate the main components of sugarcane straw to obtain chemical products with higher economic value, and the proposed obtainment is the modification and characterization of cellulose biomass to be used in the synthesis of carboxymethylcellulose (CMC) based on the bleached pulp through a combination of chemical process with the enzyme, using xylanase. It was also studied the influence in mechanical, thermal and morphological properties of polyurethane obtained from castor oil (Ricinus Communis L.) reinforced composites. Sugarcane straw was pretreated by steam explosion, delignified, treated with xylanase and bleached with hydrogen peroxide. The bleached cellulose obtained was used to produce CMC. It was determined the mass gain and the degree of substitution of the obtained CMC. The delignified pulp (cellulose) was used as reinforcement in composites of polyurethane obtained from castor oil. The biocomposites were analysed by differential scanning calorimetry (DSC), thermogravimetry (TGA), and it was also determined the mechanical properties, scanning electron microscopy (SEM) and water absorption. The incorporation of sugarcane straw fibers in the polyurethane matrix improved the mechanical properties compared with the pure matrix, and a possible reduction in the costs of production suggests a industrial application of the material.
