This diploma thesis deals with dynamic of accumulation of non-structural carbohydrates and activity of Rubisco enzyme at elevated concentration of CO2 on beech (Fagus sylvatica L.). Three years old seedlings of beech were cultivated in minisphere with ambient (385 µmol•mol-1, variant A), and with elevated concentration CO2 (700 µmol•mol-1, variant E) for four months. In every variant the first half of plants was fertilized by nitrogen (variant N+) and the second half was control (variant N-). Plants used for experiment were at first adapted for darkness for 12 hours. Subsequently tested leaves were cut off, leafstalk including short segment of branch (approximately 1 cm) was inserted into 0.7 M solution of sucrose (variant S) or water (variant V) and exposed to radiation 200 mol•m-2•s-1 for 0, 30, 60, 120, 180 minutes. Then leaf area and fresh mass of leaf blade were established, samples were fixed in liquid nitrogen and stored in deep freezer to analysis in –70 °C. Rubisco content was determined by SDS-PAGE method, Rubisco activity spectrofotometrically and content of non-structural carbohydrates by anthrone method and HPLC method. Rubisco content was significantly lower in the N- variant than in N+ variant. Rubisco content was also significantly lower in E than in A variant, which is an evidence of down-regulation. Rubisco activity is moderately stimulated at E variant with time, but differences between variant A and E are not statistically significant. Influence of sucrose feeding to Rubisco activity was not proved. Significant differences were detected by anthrone method in non-structural carbohydrates content between variants S and V, but not between variants A and E. Statistically significant increase of sucrose content with time was detected by HPLC method at variant AS, but not at variant ES.

This diploma thesis deals with a draft of wireless weather station, whose task is to measure basic values inside and outside the home. Measured variables are for example temperature, atmospheric pressure, relative humidity, wind speed and direction or concentration of carbon dioxide (CO2). These data will be transmitted to a web server for later processing. Possible technical solutions are described and the outcome is a designed printed circuit boards, which has been assembled and tested. Resulting system was tested in trial operation.

In this diploma work changes of initial and total activities and content of Rubisco in beech and Norway spruce were studied. The plants were cultivated in conditions with ambient CO2 concentration (350 mol·mol–1) and elevated CO2 concentration (700 mol·mol–1). Three series of samples (at the beginning, in the middle and at the end of growing season) were taken. Initial and total Rubisco activities were measured spectrophotometrically and activation state was calculated. Rubisco content was determined by SDS–PAGE method. Rubisco activity in beech cultivated in elevated CO2 concentration decreased during the whole growing season while in beech growing in ambient CO2 concentration Rubisco activity decreased up to middle of growing season and then increased. Rubisco content in beech in ambient CO2 concentration slightly increased and in beech in elevated CO2 concentration decreased up to middle of growing season and then increased. Rubisco activities in Norway spruce both in ambient and elevated CO2 concentration decreased. Rubisco content in Norway spruce in ambient CO2 concentration decreased but in Norway spruce in elevated CO2 concentration first decreased and then increased.

The first part of the final thesis is focused on the use of thermovision for diagnosing defects and failures in building industry. Documented history, physical principles and conditions for the measuring of thermographic camera are described in this part. Thermal images of the object described below were used for demonstrating the defects. The second part is dedicated to the elaborated energy assessment for a nine-storey slab block building. In total, there are five energy economic measures developed for the building. Three of those are of structural character and two of those are for building equipment. There are two variants designed based on the measures, one of which is recommended and evalu-ated. The final section is focused on the measurement of slab block building indoor environment (CO2, air temperature, relative humidity, dew point) and its assessment in concern with the current legislation.

The effect of elevated CO2 concentration (EC CO2) on photosynthesis has been observed on many hierarchical levels. There was a significant increase in the rate of photosynthesis of examined trees observed in previous studies thus I hypothesised these changes are accompanied by changes of chloroplast ultrastructure and photosystem content and function and the main aim of this study is to evaluate these adjustments. In this study 13 - 14 years old seedlings of Norway spruce (Picea abies L. Karst.) grown in glass domes with adjustable windows - one with ambient CO2 concentration as a control, the other one with simulated EC CO2 (700 ppm) - during the vegetative season were examined. Pigment content, fluorescence and reflectance indexes, activity of photosystem 1 (PS1) and 2 (PS 2) of isolated chloroplasts, size of cross-sectional area of chloroplast and proportion of stromal to granal thylakoids under EC CO2 treatment were investigated. Although there was a significant increase in the maximum rate of photosynthetic assimilation of trees from EC CO2 (observed by other researchers of our team), decreased chlorophyll and carotenoid content as well as the activity of both photosystems were observed, which is usually atributed to photosynthetic acclimation. La fixation symbiotique d’N par les légumineuses est très sensible aux ravageurs, provoquant des dommages sur les racines nodulées, avec un impact sur la fixation d’N et la croissance qui demeure mal connu. Nous avons alors analysé la réponse adaptative de la fixation symbiotique et de la croissance du pois Frisson sauvage et 3 de ses mutants hypernodulants P64, P118 et P121, respectivement mutés sur les gènes SYM28, SYM29 et NOD3, à une ablation de la moitié du système racinaire, en fin de phase végétative. La réponse adaptative a été mesurée 8 jours après ablation, dans des conditions d'alimentation en carbone par la photosynthèse variées. A 380 ppm, le mutant P118 a montré la plus faible diminution de l’activité spécifique de fixation (-17%) suite à l’ablation comparé au sauvage et aux 2 autres mutants (-36% à -62%) associé à une accélération chez les mutants P118 et P121 et un maintien (sauvage et P64) de la croissance des nodosités. A 150 ppm, suite à l’ablation, l’activité spécifique de fixation symbiotique par les nodosités a été diminuée (sauvage), maintenue (P64 et P118) ou augmentée (P121), associée à une accélération (sauvage et P121) ou un maintien (P64 et P118) de la croissance des nodosités. A 750 ppm, l’activité spécifique de fixation a diminué suite à l’ablation pour tous les génotypes, associée à un ralentissement (P64), un maintien (P118, sauvage) ou une faible accélération (P121) de la croissance des nodosités. Les résultats montrent une plus grande capacité de la fixation symbiotique des mutants hypernodulants (P118 et P121 essentiellement) à résister au stress provoqué par l’ablation. / Symbiotic N fixation of legumes is very sensitive to environmental stresses, like pea pests damaging nodulated roots. However, the impact on their N uptake capacity and plant growth has not been studied so far.We analyzed the adaptive response symbiotic N2 fixation and plant growth of pea wild type Frisson and hypernodulating mutants P64, P118 and P121 mutated respectively on genes SYM28, SYM29 and NOD3 to root pruning of half the root system at the end of the vegetative stage. The adaptive responses of pea: cv. Frisson and 3 of its hypernodulating mutants were compared under varying carbon supplies from photosynthesis.At 380 ppm, mutant P118 showed the lowest decrease of the specific activity of N fixation (-17%) following root pruning compared to the wild type and the 2 others mutants (-36% to -62%), associated to an acceleration (P118 and P121) and a maintained (wild type and P64) nodule growth. At 150 ppm, following root pruning, specific activity of N fixation of nodules decreased in wild type, was maintained in P64 and P118 and increased in P121. At 750 ppm, specific activity of N fixation of nodules decreased for all genotypes following root pruning, associated to a maintained nodule growth in wild type and P118, a slower growth in P64 and acceleration in P121.Our results showed a greater capacity of hypernodulating mutants P118 and P121 to withstand the stress induced by root pruning of half the root system.
