Asymmetric Hydrogenations of Chiral Acyclic Alkenes for Important Chiron Syntheses

Hydrogenation of "largely unfunctionalized" alkenes has been an active area of research for about a decade. Many catalysts have been prepared but we noticed that comparatively few substrates have been studied and none of these hydrogenations provided useful chirons for the organic synthesis area. That motivated us to investigate asymmetric hydrogenations of chiral acyclic alkenes, which are seldom used for hydrogenations and usually the reactions are fully substrate controlled. It emerged that such reactions could provide a concise entry points into chirons that can be used to prepare many natural products. Asymmetric hydrogenations of functionalized, but not coordinatively functionalized, alkenes have been used to prepare several chirons for syntheses ofpolyketide natural products using our N,carbene Crabtree's catalyst analog. Starting from optically active starting materials (eg Roche esters, lactic acid, glyceraldehyde dimethyl ketals, amino acids), highly optically active chiral alkenes can be made in several steps with high yield. With the iridium catalyzed asymmetric hydrogenations, chiral ethers, 1,3-hydroxymethyl chiron, alpha-methyl-beta-hydroxy-gamma-methyl chiron, alpha-methyl-gamma-alkyl-gamma-amino acid can be obtained with high stereoselectivities. With those well developed methodologies, (-)-dihydromyoporone, (-)-spongidepsin, (-)-invictolide have been prepared with high efficiency. Not like the vinyl acetate, which can be hydrogenated quite well with many Rh catalysts, the alkyl vinyl ether does not have a coordination functional group nearby, hence it is a difficult substrate for asymmetric hydrogenation and there are relatively few iv reports. Also the simple alkyl enol ether is quite acid sensitive and the Pfatlz's type N,PIr catalysts cannot hydrogenate the simple alkyl enol ethers well under the standard hydrogenation conditions. We explored many alkyl enol ethers and found some of them can be hydrogenated efficiently (50 bar H2, 1 mol percent N,carbene-Ir catalyst, 25 degree C) with high enantioselectivities (up to 98 percent ee). This study led us to suspect that more protons were produced when N,P-Ir catalyst precursors were used relative to the corresponding carbene catalyst since the former only gave complex mixture when being used. DF calculations and several other experiments supported this postulation.

Conception, synthèse et activité anticancéreuse d’analogues basés sur la molécule FTY720 (Gilenya)

FTY720 (aussi connu sous le nom de Fingolimod ou Gilenya) agit sur les récepteurs sphingosine-1-phosphate (S1P) et induit la suppression du système immunitaire (immunosuppression). Cette molécule est reconnue pour avoir une activité contre plusieurs cellules cancéreuses. Cette activité est indépendante de l’action sur les récepteurs S1P et on attribue plutôt la mort (apoptose) des cellules cancéreuse à la capacité que possède la molécule à réduire le transport des nutriments dans la cellule. Toutefois, malgré ses nombreux avantages, FTY720 ne peut pas être utilisé afin de traiter des humains puisque l’activation secondaire des récepteurs S1P1 et S1P3 mènent à une diminution du rythme cardiaque (bradycardie) chez les patients. Notre groupe s’est donc concentré sur la synthèse d’analogues qui potentiellement n’activeraient pas le récepteur S1P tout en gardant une activité biologique contre plusieurs cellules cancéreuses. Malgré le fait que nos analogues agissent également sur la diminution du transport des nutriments dans les cellules, nous ne connaissons pas le mécanisme d’action par lequel ceux-ci agissent. Au passage, le projet de recherche ci-présenté nous aura par ailleurs permis de développer une grande variété de sondes photo-actives dans l’espoir d’isoler une ou plusieurs protéines qui seraient impliquées dans le mécanisme d’action. / FTY720 (also known as Fingolimod, or Gilenya) functions as an immunosuppressant due to its effect on sphingosine-1-phosphate receptors. FTY720 also exhibits anticancer activity in a number of cancer cell lines. This result is independent of its effects on S1P receptors and due instead, to its ability to induce nutrient transporter down-regulation. However, FTY 720 cannot be used in human patients due to bradycardia that occurs secondary to the activation of S1P1 and S1P3 receptors. Our group focused on the design and synthesis of constrained FTY720 analogues that do not activate S1P receptors and also have the potential to be safely used in humans due to their potent antiproliferative activity against selected cancer cell lines. Although we know that our analogues, like FTY 720, act as nutrient transporter down-regulators, the proteins involved in the process are not known. We are developing a wide variety of Photo-Cross-Linking Probes to hopefully isolate one or more proteins that may be involved in one of the pathways.

REASONING ABOUT DEFINEDNESS - A DEFINEDNESS CHECKING SYSTEM FOR AN IMPLEMENTED LOGIC

<p>Effective definedness checking is crucial for an implementation of a logic with undefinedness. The objective of the MathScheme project is to develop a new approach to mechanized mathematics that seeks to combine the capabilities of computer algebra systems and computer theorem proving systems. Chiron, the underlying logic of MathScheme, is a logic with undefinedness. Therefore, it is important to automate, to the greatest extent possible, the process of checking the definedness of Chiron expressions for the MathScheme project. This thesis provides an overview of information useful for checking definedness of Chiron expressions and presents the design and implementation of an AND/OR tree-based approach for automated definedness checking based on ideas from artificial intelligence. The theorems for definedness checking are outlined first, and then a three-valued AND/OR tree is presented, and finally, the algorithm for reducing Chiron definedness problems using AND/OR trees is illustrated. An implementation of the definedness checking system is provided that is based on the theorems and algorithm. The ultimate goal of this system is to provide a powerful mechanism to automatically reduce a definedness problem to simpler definedness problems that can be easily, or perhaps automatically, checked.</p> / Master of Science (MSc)

Les alcaloïdes Daphniphyllum constituent une vaste famille de produits naturels isolés à partir de plantes à feuillage persistant couramment utilisés dans la médecine chinoise traditionnelle. Ils affichent une gamme impressionnante d'activités biologiques; antipyrétique, anti-inflammatoire, antioxydant et même anticancéreux. La calyciphylline B appartient à cette famille et possède un motif original comprenant sept stéréocentres adjacents, dont un stéréocentre quaternaire tout carbone, avec un échafaudage hexacyclique. Sa structure a été déterminée par données spectroscopiques, plus précisément par des techniques de RMN 2D. Malgré le peu d'information sur son activité biologique, sa synthèse représente sans le moindre doute un grand défi pour les chimistes organiciens. Le groupe de recherche du Prof. Hanessian a entrepris la synthèse totale de la calyciphylline B en 2010, laquelle est toujours en cours. Une nouvelle approche a été développée pour la préparation d'un intermédiaire azabicyclo[3.3.0]octane avancé. Ce mémoire résume les travaux de recherche de l'auteur sur les progrès réalisés pour la voie alternative élaborée par le groupe du prof. Hanessian. These alkaloids display an impressive range of biological activities, including antipyretic, anti-inflammatory, antioxidant, and even anticancer properties. Calyciphylline B is a structurally unique member of this family containing seven contiguous stereocenters including an all-carbon quaternary stereocenter with a fused-hexacyclic ring scaffold. Its structure was determined by spectroscopic methods, especially 2D NMR techniques. Despite the sparse availability of information on its biological activity, its synthesis is undoubtedly a great challenge for synthetic chemists. The research group of Prof. Hanessian embarked on the total synthesis of calyciphylline B in 2010 and the project is still ongoing. A new route was developed for the preparation of an advanced azabicyclo[3.3.0]octane intermediate. This thesis summarizes the research work of the author on the progress made for the synthetic route developed by the Hanessian group. The work done includes the formation of a quaternary stereocenter, alkylation of an enolate using a secondary alkyl triflate, diastereoselective reduction, reductive cyclization, and a regioselective Wacker oxidation.
