The demonstrative nature of the Hindi/Marwari correlative

One of the main features of the correlative construction is the necessity of an appropriate correlate (either a demonstrative or a pronoun) in the main clause. While the syntactic features of the correlative construction are well established, the relationship between the correlative clause and its correlate remains unclear. In this dissertation, I propose that the correlative clause is the overt pronunciation of the index of the demonstrative. The correlative, therefore, does not adjoin to IP (Dayal 1996) or the demonstrative (Bhatt 2003) but enters the syntax as the indexical argument of the demonstrative phrase (Nunberg 1993; Elbourne 2008). I then turn to the adverbial correlative clause, which involves an adverbial relative phrase, and show that it is also the overt pronunciation of the index and, further, that it is interpreted as a definite description and contributes an individual of type e. Having established the relationship between the correlative clause and its correlate, I develop a new analysis of the semantic contribution of both the single headed correlative, involving one relative phrase, and the multi-headed correlative which involves multiple relative phrases. I propose that the correlative gets its interpretation through a Q particle, QCOR, which raises from the relative phrase to Spec-CP. It is QCOR which allows both adverbial and nominal correlatives to have a definite interpretation. I present new data from Hindi and Marwari which shows that the multi-headed correlative is basegenerated inside of the main clause, at the highest demonstrative or below, and denotes an ordered pair. Each member of that set is then an argument of one of the demonstratives in the main clause. Finally, if the proposed analysis is correct, then it should be follow that other types of phrases can occur in the same position. Not only is this possible in Hindi and Marwari, but sign languages and Mandarin Chinese allow overt indices as well.

Studying cellulose nanostructure through fluorescence labeling and advanced microscopy techniques

As the major component of the plant cell wall, cellulose is produced by all plant species at an annual rate of over a hundred billion tonnes, making it the most abundant biopolymer on Earth. The hierarchical assembly of cellulose glucan chains into crystalline fibrils, bundles and higher-order networks endows cellulose with its high mechanical strength, but makes it challenging to breakdown and produce cellulose-based nanomaterials and renewable biofuels. In order to fully leverage the potential of cellulose as a sustainable resource, it is important to study the supramolecular structure and hydrolysis of this biomaterial from the nano- to the microscale. In this thesis, we develop new chemical strategies for fluorescently labeling cellulose and employ advanced imaging techniques to study its supramolecular structure at the singlefibril level. The developed labeling method provides a simple and efficient route for fluorescently tagging cellulose nanomaterials with commercially available dyes, yielding high degrees of labeling without altering the native properties of the nanocelluloses. This allowed the preparation of samples that were optimal for super-resolution fluorescence microscopy (SRFM), which was used to provide for the first time, a direct visualization of periodic disorder along the crystalline structure of individual cellulose fibrils. The alternating disordered and crystalline structure observed in SFRM was corroborated with time-lapsed acid hydrolysis experiments to propose a mechanism for the acid hydrolysis of cellulose fibrils. To gain insight on the ultrastructural origin of these regions, we applied a correlative super-resolution light and electron microscopy (SR-CLEM) workflow and observed that the disordered regions were associated nanostructural defects present along cellulose fibrils. Overall, the findings presented in this work provide significant advancements in our understanding of the hierarchical structure and depolymerization of cellulose, which will be useful for the development of new and efficient ways of breaking down this polymer for the production of renewable nanomaterials and bio-based products like biofuels and bioplastics. / Thesis / Doctor of Philosophy (PhD) / In this dissertation, we have studied in unprecedented detail the structure of cellulose – a polymer that is found in every plant. As the main structural component of the plant cell wall, cellulose endows trees with their strength and resilience while storing sunlight energy in its chemical bonds. Since plant biomass represents eighty percent of all living matter on Earth, cellulose is an abundant resource that can be used to produce sustainable and environmentally benign nanomaterials and bioproducts, like biofuels and bioplastics. Our ability to use cellulose as a renewable source of structural materials and energy is intimately linked to our capacity to break apart its tight structural packing. Deconstructing cellulose into various forms demands that we understand the multi-level organization of its structure and the susceptible regions within it. To gain this information, in this thesis we develop new labeling methods and apply state-of-the-art microscopy tools to directly visualize the arrangement of cellulose fibrils at the nanoscale (comparable to 1/10,000 the width of a human hair) and study their breakdown by acid treatment. The findings presented in this work furthers our fundamental understanding of the natural structure of cellulose, which has important implications on the development of industrial strategies to break down this abundant and renewable biomaterial.

Correlative multiscale imaging and quantification of bone ingrowth in porous Ti implants

Additive manufactured porous titanium scaffolds have been extensively investigated for orthopaedic applications. The quantification of tissue response to the biomaterial implants is primarily achieved by analysing a two-dimensional (2D) stained histological section. More recently, three-dimensional X-ray micro-computed tomography (μCT) has become increasingly applied. Although histology is the gold standard, μCT allows non- destructive quantification of 3D tissue structures with minimal sample preparation and high contrast. A methodology to correlate information from both histology and μCT of a single sample might provide greater insights than either examining the results separately. However, this task is challenging because histology and μCT provide different types of information (stained tissue morphology vs. greyscale dependent on the X-ray absorption of material) and dimensionality (2D vs 3D). A semi-automated methodology was developed to directly quantify tissue formation and efficacy within an additive manufactured titanium implant using histology and μCT. This methodology was then extended to correlatively integrate nano-scale elemental information from nano- secondary ion mass spectroscopy (NanoSIMS). The correlative information was applied to investigate the impact of silver release on bone formation within a nano-silver coated additive manufactured implant. The correlative imaging methodology allowed for the quantification of the significant volumetric shrinkage (~15%) that occurs on histology slice preparation. It also demonstrated the importance of the location of the histological sectioning of the tissue and implant, revealing that up to 30% differences in bone ingrowth can be found along the entire length of the porous implant due to preferential bone ingrowth from the periphery to the centre. The quality and quantity of newly formed bone were found to be comparable between the uncoated and nano-silver coated Ti-implants, suggesting that the layer of silver nanoparticles on the Ti-implant does not negatively impact bone formation. Further, the newly formed bone at 2 weeks had a trabecula morphology with bone at the interface of Ti-implant as well as at a distant. This indicates that both contact (bone apposition on implant) and distance (bone ingrowth from host bone) osteogenesis were present in both types of implants. Finally, nanoscale elemental mapping showed silver was present primarily in the osseous tissue and was co-localised to sulphur suggesting that silver sulphide may have formed.

Coordinations de prédicats en mandarin contemporain / Coordinations of predicates in contemporary Mandarin

Dans ce travail, nous examinons les propriétés syntaxiques et sémantiques des coordonnants de prédicats du mandarin contemporain ainsi que les structures syntaxiques des coordinations qu’ils forment. Nous nous intéressons aux coordinations simples et corrélatives. D’une part, nous découvrons que comme Zhang Ning (2010) l’a avancé les coordinations simples du mandarin ont une structure de type spécifieur-tête-complément : le coordonnant est la tête de la coordination, le conjoint externe le spécifieur et le conjoint interne le complément. Mais d’autre part, nous démontrons que, contrairement à l’affirmation de Zhang Ning (2008), la composition de la structure syntaxique des coordinations corrélatives du mandarin est différente de celles des langues germaniques : les premières sont composées d’une conjonction nulle et de plusieurs particules de focus tandis que les secondes, comme le montrent des travaux antérieurs, sont constituées d’une conjonction prononcée et d’une particule de focus. Au long de notre étude, nous comparons par ailleurs les coordonnants de prédicats du mandarin sur les plans syntaxique et sémantique. Nous remarquons qu’ils diffèrent l’un de l’autre selon au moins l’un des aspects suivants : les catégories syntaxiques des éléments qu’ils peuvent coordonner, les relations sémantiques qu’ils peuvent établir entre leurs conjoints et la possession d’un trait [V] ou d’un trait [-V]. Enfin, nous notons que les prédicats reliés par certains coordonnants doivent, dans la plupart des cas, avoir les mêmes traits aspectuels. Nous expliquons que cette tendance est liée au type de relation sémantique que ces coordonnants établissent entre les prédicats qu’ils relient. / In this dissertation, we study the syntactic and semantic features of several coordinators of predicates in contemporary Mandarin and analyze the syntactic structures of the coordinate constructions formed by these coordinators. We investigate not only simple but also correlative coordinations. We notice that, on the one hand, the Mandarin simple coordination has most likely a head-specifier-complement structure, as Zhang Ning (2010) has proposed: the coordinator is the head, the external conjunct is the specifier and the internal conjunct is the complement. On the other hand, in contrast to Zhang Ning’s (2008) claim, we discover that the correlative coordinations of Mandarin and those of Germanic languages do not have the same syntactic configurations: the former are composed of one silent conjunction and at least two focus-sensitive particles while the latter, as previous research has noted, are made up of one overt conjunction and one focus-sensitive particle. In addition, we find out that the Mandarin coordinators of predicates differ from one another in at least one of the following respects: the syntactic categories of the elements that they can coordinate, the semantic relationships that they may build between their conjuncts and the possession of a [V] feature or a [-V] feature. Last but not least, we notice that the predicates coordinated by some coordinators must, in most of the cases, have identical aspectual features. We argue that this tendency is related to the type of semantic relationship that these coordinators build between their conjuncts.

In vivo Analysis of the Role of FtsZ1 and FtsZ2 Proteins in Chloroplast Division in Arabidopsis thaliana

Chloroplasts divide by a constrictive fission process that is regulated by FtsZ proteins. Given the importance of photosynthesis and chloroplasts in general, it is important to understand the mechanisms and molecular biology of chloroplast division. An FtsZ gene is known to be of prokaryotic origin and to have been transferred from a symbiont's genome to host genome via lateral transfer. Subsequent duplication of the initial FtsZ gene gave rise to the FtsZ1 and FtsZ2 genes and protein families in eukaryotes. These proteins co-localize mid-chloroplast to form the Z-ring. Z-ring assembly initiates chloroplast division, and it serves as a scaffold for other chloroplast division proteins. Little is known, however, about the FtsZ protein subunit turnover within the Z-ring, the effects of accessory proteins on Z-ring turnover assemblies, as well as the in vivo ultrastructure of the Z-ring in plants. To investigate the Arabidopsis thaliana FtsZ subunit turnover rate within the Z-ring, a section of the Z-ring in the chloroplasts of living plants expressing fluorescently tagged FtsZ1 or FtsZ2 proteins was photobleached and the recovery rate was monitored. The results show that the fluorescence recovery half times for the FtsZ1 and FtsZ2 proteins are 117s and 325s, respectively. This is significant as these data mirror their differences in GTP hydrolysis rates. To elucidate in vivo structure and ultrastructure of the Z-ring, a protocol was established that maintained fluorescence during high pressure freezing, freeze substitution and low temperature embedding. Afterwards, a correlative microscopy approach was employed to visualize and identify fluorescently labeled puncta, circular structures, at the light microscopy level. These puncta were further resolved as mini-rings using optical microscopy eXperimental (OMX) superresolution microscopy. Electron microscopy (EM) analysis imaged mini-rings and filament assemblies comprised of dense subunits. Electron tomography (ET) showed mini-rings composed of protofilaments.

That is, the political tradition and institution, how the two actors got close relations with the historical frames of Han dynasty. The historical institutionalism, being the school of New institutionalism, try to catch a clear mark of power elites and meaningful events, so we can have clear idea of Han history, because we are using a useful approach to study the political development of Han history. We think the most basic rule of ancient Chinese history is one man rule. Wang Mang use the popular concept of correlative cosmology, the school of yin and yang, to break one man rule of ancient Chinese history. But things are not exactly the same of eastern Han. Cao Cao has to fight to survive, because the rule of correlative cosmology was not working so well in eastern Han.

Studies of macromolecular trafficking across Arabidopsis homografts

Micrografting was used to study the restoration of symplasmic transport at the graft union and to examine the long-distance transport of macromolecules between scion and rootstock. New techniques were established, such as correlative imaging and single-cell analysis in microfluidic devices, to study graft development both in vivo and in vitro. Imaging of Arabidopsis homografts showed that a symplasmic domain develops in the callus stele whose function may be to contain the spread of auxin into the surrounding ground tissue. It was demonstrated, also, that recent reports of organelle transfer at the graft union cannot be explained by the formation of secondary plasmodesmata (PD) at the graft interface. While fused calli did not exchange organelles in vitro, large aggregates of the SIEVE-ELEMENT OCCLUSION RELATED protein fused to YFP (SEOR-YFP; 112 kDa) were unloaded from mature sieve tubes into living cells of the graft partner in vivo, suggesting that vascular remodelling may be a prerequisite for the exchange of organelles at the graft interface. Fusion proteins expressing organelle-targeting signals were found to translocate across the graft junction, unloading into cell files adjacent to the root protophloem. The phloem mobility of a given fusion protein was assessed using bioinformatic and statistical analysis of publicly available data. The size of a protein and its relative abundance in CCs both emerged as defining factors for subsequent phloem transport. The recipient tissue for phloem-unloaded macromolecules was identified as the phloem-pole pericycle (PPP). This cell layer is required to remove macromolecules from the terminus of the protophloem. Induced callose deposition at the PD that connect protophloem SEs to the PPP caused a restriction in unloading and a subsequent arrest in root growth. A non-cell autonomous protein of CC origin, NaKR1-1, is proposed to affect the unloading of macromolecules either by increasing the size exclusion limit (SEL) of PD within the PPP or by enabling a build-up in pressure at the protophloem terminus, due to SUC2 activity, thus allowing phloem unloading.

Amostras da liga de alumínio 7075-T6 passaram por um processo de recozimento pleno seguido de um estágio de compressão uniaxial, obtendo níveis de deformações entre 25 e 65%. As microestruturas das amostras foram avaliadas em função dos parâmetros morfológicos dos precipitados, da reorientação dos planos cristalográficos dos grãos e da orientação das subestruturas formadas durante o processo de deformação. Para a caracterização foram utilizadas técnicas de difração de raios-X, microscopia eletrônica, microscopia óptica utilizando técnicas de polarização linear e circular, microscopia correlativa e processamento digital de imagens. Os resultados de difração de raios-X indicaram uma reorientação do plano cristalográficos (200) para o (220) após a deformação, e as técnicas de microscopia eletrônica identificaram precipitados de Mg2Si, Al7Cu2Fe e Al6(FeCu) na liga. A análise morfológica dos precipitados indicou uma maior fragmentação dos precipitados devido à maior ativação do plano (331) a partir de 39% de deformação. Por meio do processamento de imagens foi encontrada uma tendência de correlação entre os planos cristalográficos e a fração de área das fases, enquanto que os parâmetros morfológicos das subestruturas formadas durante o processo de deformação permitiram avaliar apenas qualitativamente o nível de encruamento das amostras. / Digital image processing and analysis tools were developed to perform the AA 7075 crystallographic and morphologic texture evaluation under different level of plastic deformation by uniaxial compression. Samples of AA 7075-T6 were submitted to full annealing process followed by uniaxial compression, thus obtaining deformations between 25 and 65% of thickness. The samples microstructure evaluation was performed considering: precipitates morphological parameters, crystallographic lattices reorientation and deformation substructure orientation. The characterization technics were: X-ray diffraction, electron microscopy, optical microscopy with polarization light, correlative microscopy and digital images processing. Xray diffraction results showed that the crystallographic plane (200) was reoriented to (220) after compression. The EDS analysis identified precipitates of Mg2Si, Al7Cu2Fe e Al6(FeCu). The precipitates morphological analysis showed an increase in fragmentation due to plane (331) at 39% of deformation. The digital image process of the samples etched with Barker reagents indicated a correlation between area fraction and the diffraction peaks, and the deformation substructures analysis made viable a qualitative characterization of the hardening process. / FAPESP: 2011/00403-2
