Exploiting stable isotope imaging with high resolution secondary ion mass spectrometry for applications in biology

This thesis presents applications of high resolution secondary ion mass spectrometry (NanoSIMS) analysis for stable isotope imaging in biological samples. These projects were designed to explore the potential applications of NanoSIMS analysis, and to develop protocols and novel methodologies to visualize and quantify biological processes. Working with collaborators in the UK and USA, I have applied NanoSIMS analysis to study 3 research areas, including molecule interactions, single cell metabolisms and lipid imaging in tissues. Antimicrobial peptides (AMPs) play important role in the immune system, and understanding how AMPs interact with cell membranes can provide useful information to design new therapies to control infection. The pore structures and dynamics of the interaction of AMPs with membranes has been visualized for the first time and confirmed with combined AFM and NanoSIMS analysis. A correlative backscattered electron (BSE) imaging and NanoSIMS analysis methodology has been developed to study glutamine metabolism in single cancer cells. This method enables us to measure the chemical information in specific organelles in these cells and can be widely applied to study metabolisms and to trace the uptake of labelled molecules in biological matrices. Quantitative analysis on the effects of hypoxic conditions and the PYGL gene were studied. Applying correlative BSE and NanoSIMS analysis, I also studied lipid uptake mechanisms in various mouse tissues, including brown adipose tissue, heart, intestines, liver and skeletal muscle, mainly focused on a recently discovered protein, GPIHBP1, and its function in the lipid uptake process. TRL margination was proved to depend on the GPIBP1-LPL complex, and 3 stages of lipid transport from capillary lumen to lipid droplets was also visualized by combined BSE and NanoSIMS analysis.

Darbo tikslas – ištirti aplinkos georodiklių sąsajas su skirtingų susirgimų grupių rizika ir ranguoti Lietuvos regionus pagal palankumą (nepalankumą) susirgimams atskiromis ligų grupėmis. Tyrimų duomenys – teritorijos georodikliai paimti iš Lietuvos teritorijos geologinių tyrimų duomenų, o duomenys apie susirgimus iš Higienos instituto. Tyrimų objektas – Lietuvos teritorija. Tyrimų metodai – matematinė statistinė analizė, daugiakriterinė analizė ir daugiatikslių sprendimų priėmimo metodai. Tyrimo rezultatų mokslinis naujumas – pirmą kartą atliktas Lietuvos teritorijos rangavimas pagal riziką susirgti tam tikromis ligomis ir sukurta rangavimo metodika galėtų teikti papildomą informaciją kadastrams. Galimas praktinis rezultatų panaudojimas – vertinant teritorijas sveikatingumo požiūriu, nustatant nekilnojamojo turto rinkos vertę, pasirenkant gyvenamąją vietą. Darbą sudaro: įvadas, 3 skyriai, išvados, literatūros sąrašas, priedai. Darbo apimtis – 58 p. teksto be priedų, 35 paveikslų, 10 lentelių. / The research purpose – to analyze the correlation between the geographical environment indexes and risk of different illnesses and to rank Lithuanian regions by disfavor and disfavour of particular illnesses groups. The research data – the environment geoindexes were taken from Lithuanian Geology Service of GIS date base and data about illnesses were taken from Hygiene Institute. The object of the research – the territory of Lithuania. The method of the research – the mathematical statistical analysis, the multiobjective analysis and the multipurpose awards accepatnce method. The scientific novelty of results – The first time Lithuanian territories were arranged by the risk to developing certain illnesses and the created method of arrangement can give an additional information for the Cadastres. Its is possible the practical results using – to value the territories according as the attitude of healthy; evaluated the market value of the realty, choosing the living place. It includes: an introduction; 3 chapters; the conclusion, references, additions. The size of the thesis - 58 pages of the text without additions, 35 illustrations, 10 charts.

Posouzení vybraných ukazatelů firmy pomocí analýzy časových řad / Assessing Selected Indicators of a Company Using Time Series Analysis

The diploma thesis deals with an assessing selected indicators of a KAVALE company s. r. o. using time series analysis. Issues of financial analysis, analysis of period lines, regressive analysis and correlative analysis are described in a theoretical part. Practical part is focused on statistic analysis of selected indicators of financial analysis. Its aim is to reveal dependance between indicators and prediction of their future development for the next two periods of time. Comparison of the analyzed company with an average sector and with two rival companies is also part of the practical section. On the basis of these analyzes I recommend proposals to improve the financial situation of the company.

Dans le cadre de la conservation et de la réhabilitation des bâtis anciens, les matériaux utilisés pour leur construction doivent être caractérisés vis-à-vis de leurs propriétés de transferts thermiques et hydriques. La première étape du travail de recherche a concerné la mise au point d'un capteur de conductivité thermique et de teneur en eau des matériaux inorganiques poreux (pierres, bétons, enduits) constitutifs des monuments. Les méthodes d’investigation doivent rester non intrusives. Dans cet objectif, la voie suivie a été d'adapter la mesure de conductivité thermique par une méthode simplifiée du fil chaud, précédemment développée pour les fluides corrosifs, aux pierres de taille utilisées dans le bâti ancien. Afin de transférer cette technologie des fluides vers les solides, plusieurs étapes ont été effectuées : (1) des mesures avec le fil intégré, (2) des mesures de surface, (3) estimation de l'influence de la rugosité de la surface et (4) utilisation de la méthode pour l'estimation de la teneur en eau des matériaux tests.La seconde étape de la recherche a consisté en deux suivis microclimatiques effectués sur deux sites historiques : l'Hypogée des Dunes et l'abbatiale de Saint-Savin-sur-Gartempe. Le travail de thèse a consisté à évaluer l'impact des travaux effectués sur les conditions climatiques dans l'hypogée, et à déterminer l’influence des variations climatiques sur la formation d'un voile biologique dans la crypte de Saint-Savin. Pour ce faire, l'analyse corrélatoire est appliquée au traitement des donnés climatiques. / In the framework of preservation and rehabilitation of ancient buildings, materials used for their construction must be characterized with regards to thermal and hydric transfer properties. The first stage of our research work focused on the development of a sensor of thermal conductivity and moisture content of the inorganic porous materials (stone, concrete, render) making up the monuments. The methods of investigation have to remain non-invasive. For this purpose, the path followed was to adapt the measure of thermal conductivity by a simplified transient hot-wire method, previously developed for corrosive fluids, to dressed stones used in built heritage. In order to transfer this technology from fluids to the solids, several stages were carried out : (1) measures with the integrated wire, (2) surface measures, (3) estimation of the influence of surface roughness and (4) use of this method to estimate the moisture content of several test materials.The second stage of the research consisted in the microclimatic monitoring of two historical sites: the Hypogeum of the Dunes in Poitiers and the crypt of the abbey church of Saint-Savin-sur-Gartempe. The thesis work sought to evaluate the impact of construction work performed in the Hypogeum on the internal climatic conditions, and to determine the influence of climatic variations on the formation of a biological veil in the crypt of Saint-Savin. To this aim, correlative analysis is applied to the treatment of climatic data.
