Tabebuia aurea is a species of the family Bignoniaceae typical of the Cerrado, having large sized bees as its main pollinator. The dispersal of pollen by bees has been well-studied and genomic techniques can facilitate this type of study. This work aimed to develop strategies for the extraction and amplification of DNA from pollen grains of Tabebuia aurea. For this purpose, we collected pollen grains of bee legs caught in Parque Nacional de Bras??lia ??? DF, which subsequently underwent two different techniques of WGA (Whole Genome Amplification). The WGA amplification products were used to amplify 15 microsatellite loci, which were used for characterization of the genetic diversity of the pollen grains. The amplified fragments were separated by capillary electrophoresis in an automatic sequencer and the results were analyzed using the program Geneious. Twenty-two trees were evaluated and the characterization of the allelic diversity was held for groups of pollen grains, each containing 20 grains, from 31 bees; single pollen grains were also evaluated. The WGA technique OmnPlex was the one that produced the largest amount of genomic DNA allowing the amplification of different microsatellite loci. Using the DNA obtained by this technique as a template, all groups of pollen grains showed high rate of polymorphism with 10 or more alleles per locus. In addition, it was observed that most of the bees carry pollen from only one donor, indicating that bees remain for a long time in the same tree. / As abelhas d??o uma grande contribui????o na manuten????o da variabilidade gen??tica de plantas do Cerrado. Tabebuia aurea ?? uma esp??cie da fam??lia Bignoniaceae, t??pica do Cerrado, tendo as abelhas de grande porte como seu principal polinizador. A dispers??o de gr??os de p??len por abelhas vem sendo bastante estudada e as t??cnicas da gen??mica podem facilitar esse tipo de estudo. Este trabalho teve como objetivo desenvolver estrat??gias para a extra????o e amplifica????o de DNA de gr??os de p??len de Tabebuia aurea. Para isto, foram coletados gr??os de p??len das patas de abelhas capturadas no Parque Nacional de Bras??lia ??? DF, que posteriormente foram submetidos a duas t??cnicas diferentes de WGA (Whole Genome Amplification). Os produtos da amplifica????o WGA foram utilizados para amplificar 15 locos microssat??lites, os quais foram usados para caracteriza????o da diversidade al??lica dos gr??os de p??len. Os fragmentos amplificados foram separados por eletroforese capilar em um sequenciador autom??tico e os resultados foram analisados utilizando o programa Geneious. Foram avaliadas 22 ??rvores e a caracteriza????o da diversidade al??lica foi realizada para conjuntos de gr??os de p??len, cada um contendo 20 gr??os, provenientes de 31 abelhas; foram avaliados tamb??m gr??os de p??len ??nicos. A t??cnica WGA OmnPlex foi aquela que produziu a maior quantidade de DNA gen??mico permitindo a amplifica????o espec??fica dos diferentes locos microssat??lites. Utilizando o DNA obtido por essa t??cnica como molde, todos os grupos de gr??os de p??len apresentaram alta taxa de polimorfismo com 10 ou mais alelos por loco, al??m disso, observou-se que a maioria das abelhas carregavam gr??os de p??len de apenas um doador, indicando que as abelhas permanecem por muito tempo em uma mesma ??rvore.
