Expresión de catepsinas durante el desarrollo embrionario temprano en el pez dorado (Seriola lalandi Valenciennes 1833)

Memoria para optar al Título Profesional de Médico Veterinario / El pez dorado S. lalandi, es una especie marina pelágica, que posee características favorables para su producción intensiva, tales como altas tasas de crecimiento y de conversión alimenticia, resistencia a las altas densidades de los cultivos, docilidad y demanda internacional creciente (Poortenaar et al., 2001). Sin embargo, en su acuicultura se ha observado una baja sobrevivencia en las etapas de hatchery, la que se atribuiría principalmente a la producción de huevos y embriones de baja calidad (Carnevali et al., 2001). Una de las principales causas de esta deficiencia, sería una deficiente acumulación y procesamiento del vitelo durante la ovogénesis y el desarrollo embrionario temprano. Se ha visto, que las catepsinas B, D y L son las principales enzimas lisosomales involucradas en la proteólisis del vitelo en los vertebrados ovíparos (Carnevali et al., 2006). La expresión de estas enzimas, así como su actividad durante la ovogénesis y la embriogénesis, parecen ser especie-específicas, desconociéndose estos antecedentes en miembros del género Seriola. En este trabajo, se evaluó la expresión de las catepsinas B, D y L durante diferentes etapas del desarrollo embrionario temprano de S. lalandi (E1: huevos recién fecundados, E2: 4 células, E3: 16 células, E4: bástula, E5: 24 HPF y E6: 48 HPF). Se utilizó la técnica RT-qPCR y como estrategia de cuantificación, la normalización de la expresión con los genes constitutivos Tubulina (TUB) y -actina (ACTB), los cuales se validaron como referencia entre seis genes constitutivos candidatos. Las tres catepsinas se expresaron durante los distintos estadíos, presentando un nivel de expresión alto en los primeros estadíos, representando probablemente los niveles de transcritos maternos. Luego, durante la blastulación donde ocurre la MBT, hubo una disminución brusca de la expresión. Posteriormente, la expresión presentó una disminución en catepsina B, no así para catepsina D y L, que aumentaron sus niveles de expresión indicando la expresión cigótica de estas enzimas. Estos resultados concuerdan con la función de estas proteasas en generar las condiciones osmóticas necesarias para la hidratación final del ovocito y además, proveer al embrión de energía durante su periodo de alimentación endógena, a través de los aminoácidos libres generados de la proteólisis del vitelo / Financiamiento: Proyecto Fondecyt Postdoctorado 3120211 y PDACH Área Genética

This diploma thesis studies the transformation of the image of conquistador Lope de Aguirre in selected pieces of Hispanic literature. The introductory compilation explains the historical context of the colonial background in which Aguirre's rebellion took place. Some historical facts about Aguirre himself are added as well. The analytical part of this thesis concentrates on Aguirre's image in literature written in Spanish, from the second half of the 16th century till present. The four hundred year development of a historical persona into a myth is very briefly captured. Given the amount of possible research samples we choose to primarily focus on Aguirre's image in 16th century chronicles written by witnesses, namely the chronicle of Francisco Vázquez. Secondarily, we also look into the image created by post-boom writers in the seventies in the 20th century, with special consideration of Abel Posse's novel Daimón. As a result, we can make a limited comparison between one of the very first and one of the very last literary images of Aguirre. Based upon these images we also try to deduce the influence of contemporary values and mentality on the creation of said images.

O Rio dos Sinos está listado como um dos oito rios mais poluídos do país e atualmente programas de revitalização estão sendo desenvolvidos para recuperar os danos causados. O dourado (Salminus brasiliensis) é uma espécie ameaçada de extinção no Rio Grande do Sul amplamente estudada na bacia. O estudo tem como objetivo reintroduzir juvenis de dourado nas cabeceiras do Rio dos Sinos e testar o efeito da densidade na dispersão. Na avaliação foi utilizada radiotelemetria, onde nanotags foram introduzidos em 25 dourados, sub-amostra de 200 peixes liberados. Os peixes foram soltos em dois eventos um em menor densidade (0,017 peixes/m-2) e outro em maior densidade (0,040 peixes/m-2). Foram rastreados durante 120 dias. A taxa de permanência foi de 68%, inoperância 20% e emigração de 12%. O pico de deslocamento ocorreu nos dias iniciais à introdução. The Sinos River are listed as one of the eight most polluted rivers in the country, and now, revitalization programs are being developed to recover the damages. The dorado (Salminus brasiliensis) is an endangered species in Rio Grande do Sul, widely studied in the basin. The objectives of that study is reintroduce dorado juvenile in headwaters of the Sinos River and test the effect of density on displacement. This thesis examines the account of the discovery of the Amazon River written by Gaspar de Carvajal in the sixteenth century. In his Relación del descubrimiento del famoso río grande que, desde su nacimiento hasta el mar descubrió el capitán Francisco de Orellana, Carvajal describes the nine-month journey in which Orellana and his men crossed South America from the Andes to the Atlantic Ocean. In makeshift boats, they traveled through the river that we now know as the Amazon. The fact that the river has kept the name of the mythical warriors of European classical culture shows how the Amazon region has been, for the past five centuries, a mixture of legends from the new and the old worlds. Therefore, I analyze how the myth of the Amazons came to be such an intrinsic part of the New World. In order to do so, I trace the origin of the myth in Ancient Greece and how the Amazons made it into the Spanish books of chivalry during the Middle Ages and the Renaissance. These books were widely read by the conquistadors, who then thought they had found these mythical warriors in the heart of the unfathomable Amazon jungle. There are two other myths that were of particular importance in Orellana's expedition, the "Cinnamon Country" and "El Dorado," which I also analyze. Finally, I recount the history of the manuscript, which was not known until the nineteenth century, and how Carvajal used a large variety of sources, as well as an amalgam of American indigenous terms, to write his chronicle. In an appendix at the end of this study, I have included my transcription of the manuscript, which is the first semi-paleographic transcription of Carvajal's account.
