ENZYMATIC TRANSESTERIFICATION OF WASTE ANIMAL FATS FOR PRODUCTION OF BIODIESEL

The process of transesterification is the exchange of the organic group R” of an ester with the organic group R’of an alcohol, often catalyzed by acid, base or enzyme. Biodiesel, a mixture of monoalkyl esters of long chain fatty acids, is produced from vegetable oils, animal fats and fish oils by transesterification in presence of alcohol. Biodiesel is a fuel which can be used in a mixture of other fuels or alone. The base catalyzed transesterification method of biodiesel production is not suitable for waste animal fat as it contains 10–15% free fatty acids which result in higher soap formation and cause extensive downstream processing. Enzyme catalyzed transesterification can overcome the problem of soap formation and multi-step purification of end products and results in a higher purity biodiesel. Lipase is the enzyme widely used in the process of enzymatic transesterification. Various lipases have been used to transesterify triglycerides with short chain alcohols to alkyl esters. The objectives of this study were to screen lipase enzymes for the transesterification process and to use the best lipase for biodiesel production from waste animal fat. Enzymatic transesterification by individual and combined enzyme catalysts (Novozyme 435 and NS88001) was first carried out to investigate the effects of reaction time (4, 8, 12 and 16 hour), oil : alcohol molar ratios (1:1, 1:2, 1:3, 1:4 and 1:5), the effects of alcohol type (methanol and 2-butanol) and reaction temperature (35, 40, 45 and 50°C) on biodiesel yield in solvent and solvent-free systems. The highest conversion yield of biodiesel (96.67%) was obtained from a combination of Novozyme and NS88001 lipase with the optimal reaction condition of oil : 2-butanol molar ratio of 1:4, enzyme concentration of 25% (12.5% w/w of each enzyme), hexane as solvent, a 45°C reaction temperature, a reaction time of 16 h and a mixing speed of 200 rpm. The reusability of lipase enzymes by individual and combination of enzyme catalysts (Novozyme 435 and NS88001) with solvent and solvent-free systems was also investigated in order to reduce the cost of the process. The lipase enzymes lost their activity after being reused for 30 cycles in solvent-free systems and after 10 cycles in solvent system.

Synthesis Of Biodiesel In Supercritical Fluids

Alternative fuels are becoming important due to diminishing fossil fuel reserves and the environmental hazards associated with exhaust gases. Biodiesel is an attractive alternative fuel because it is environmentally friendly and can be synthesized from edible/non-edible oils. Though several methods are available for synthesis, transesterification is the preferred route for biodiesel synthesis. The current techniques for transesterification of the oils to biodiesel are based on acid/alkali catalysis. However, these methods do not work for oils with high free fatty acid content and also require an additional downstream step for separation of catalyst from the product. In this work, we investigate the synthesis of biodiesel by two techniques: in supercritical methanol and ethanol without any catalyst; in supercritical carbon dioxide using enzyme as catalyst. In the first technique, the transesterification of refined grade edible oil like sunflower oil, palm oil and groundnut oil, and crude non-edible oils like Pongamia pinnata and Jatropha curcas was investigated in supercritical methanol and ethanol without using any catalyst from 200 oC to 400 oC at 200 bar. The variables affecting the conversion during transesterification, such as molar ratio of alcohol to oil, temperature and time were investigated in supercritical methanol and ethanol. The conversion to biodiesel increased from 30-40% at a molar ratio of 10 to 80-85% at a molar ratio of 45 to 50. Very high conversions (96-97%) were obtained in supercritical methanol and ethanol within 30 minutes at 350 oC. The kinetics of the reaction was modeled, the rate coefficients and activation energies were determined. As an enzymatic transesterification in organic solvents is mass transfer limited, supercritical fluids are found to be a better alternative because of their gas like diffusivity. Among potentially interesting solvents for enzymatic synthesis, carbon dioxide is the most obvious choice in supercritical fluids, because it is non-toxic, nonflammable and easily available. Because the products and the enzyme do not dissolve in carbon dioxide at room conditions, separation can easily be achieved by reduction of pressure Thus, the enzymatic transesterification for production of biodiesel in supercritical fluids under mild conditions is of commercial interest. Therefore, biodiesel was synthesized using immobilized enzyme (Lipase, Novozym-435) in supercritical carbon dioxide. The effect of reaction variables like temperature, molar ratio, enzyme loading and kinetics of the reaction was investigated. The conversion to biodiesel was found to be lower in supercritical carbon dioxide than that in supercritical alcohol.

Mise en œuvre des lipases végétales issues des graines dans la catalyse enzymatique d’esters éthyliques d’huiles végétales pour la production de biodiesel / Use of plant lipases from seeds for enzymatic catalysis of ethyl esters of vegetable oils for the production of biodiesel

Les lipases présentent un grand intérêt pour la synthèse du Biodiesel, carburant alternatif au gasoil, généralement obtenu d’une transestérification des triacylglycérols avec un alcool, la plupart du temps le méthanol. Pour avoir un ester issu totalement de la biomasse végétale, l’éthanol peut être utilisé comme accepteur d’acyle. L’objectif de cette étude est de développer des procédés enzymatiques de synthèses d’esters éthyliques catalysés par les lipases végétales sous leur forme brute avec des intrants (huile et alcool) d’origine végétale. D’abord, elle a consisté à la mise en évidence d’une activité lipasique pour des réactions d’éthanolyse et d’hydrolyse par les graines d’A. suarezensis, d’A. grandidieri, de J. curcas, de J. mahafalensis, de M. oleifera et de M. drouhardii. Ensuite, les influences de certains facteurs sur la capacité des extraits le(s) plus actif(s) à réaliser des réactions d’éthanolyse en milieux non aqueux, aqueux et en utilisant comme substrat leurs lipides natifs ont été étudiées. Enfin, des essais de combustion ont été menés sur un moteur monocylindre à injection directe pour l’étude des performances, des émissions et de la combustion du biodiesel produit et de ses mélanges avec le gasoil. Toutes les graines germées sont dotées d’une activité en hydrolyse et éthanolyse. La poudre d’A. grandidieri est la plus active en éthanolyse. Avec cette dernière, deux procédés ont pu être développés : un en milieu non aqueux et un en milieu aqueux (respectivement un rendement de 96,2 % et 96,3 %). Elle est aussi capable de transformer ses lipides natifs sans extraction au préalable en esters éthyliques (rendement de 91,6%). Les performances et la combustion du biodiesel et de ses mélanges sont similaires à celle du gasoil. Une réduction significative des émissions de CO, NOX, CO2 et SO2 au cours de la combustion du biodiesel et de ses mélanges est observée. Ces résultats montrent que les lipases végétales exploitées sous leurs formes brutes peuvent être une alternative aux lipases microbiennes et aux catalyseurs chimiques. / There is a great interest in the use of lipase in the production of Biodiesel, alternative diesel fuel, usually obtained from a transesterification of triacylglycerol with an alcohol which is mostly methanol. To have a biodiesel derived totally from vegetable biomass, ethanol must be explored as acyl acceptor. The objective of this work is to develop enzymatic processes for the synthesis of ethyl esters catalyzed by plant lipases in their crude form with all inputs (oil and alcohol) of origin plant. Firstly, the hydrolysis and ethanolysis activities of A. suarezensis, A. grandidieri, J. curcas, J. mahafalensis, M. oleifera and M. drouhardii seeds were assessed. Subsequently, the most active(s) plant lipase(s) was selected to study the effects of some factors on their ability to carry out ethanolysis reactions in nonaqueous, aqueous media and using as substrate their lipids. Finally, combustion tests were carried out on a single cylinder direct injection engine to study the performance, emissions and combustion of biodiesel and its mixture with diesel. All germinated seeds have hydrolysis and ethanolysis activity. The most active in ethanolysis is the powder from A. grandidieri seed. With this powder, two processes were developed: one in nonaquous medium and the other in aqueous medium (yield of 96.2 % and 96.3 %, respectively). Lipase from A. grandidieri seed is able to transesterify its oils without an extraction thereof into ethyl ester. Performance and combustion characteristics of biodiesel and its mixtures are similar to that of diesel fuel. A significant reduction in CO, NOX, CO2 and SO2 emissions during the combustion of biodiesel and its mixtures is observed. These results show that plant lipases exploited in their crude form can be an alternative to microbial lipases and chemical catalysts.

As características nutricionais de produtos alimentícios têm sido cada vez mais valorizadas pelo mercado consumidor em função da constante preocupação com os efeitos dos alimentos na saúde. O sucesso de um produto alimentício no mercado, entretanto, depende também de suas características físicas, tais como espalhabilidade, aparência e impressões sensoriais dos alimentos na boca. Estas características desempenham papel fundamental no processamento industrial e na aceitabilidade do produto pelo consumidor. Neste contexto, o presente projeto teve como objetivo avaliar o efeito da transesterificação enzimática da gordura de leite com óleo de soja na composição e na textura do produto alimentício obtido, empregando como catalisador lipase de Aspergillus niger previamente imobilizada em suporte híbrido polissiloxano-álcool polivinílico. Em uma primeira etapa, as matérias-primas foram caracterizadas quanto à umidade, índices de acidez, saponificação, peróxido e iodo, propriedades reológicas e térmicas. Em seguida, os materiais de partida foram misturados em diferentes proporções seguindo um planejamento experimental estatístico de misturas, e empregados nas reações de transesterificação. As variáveis resposta foram os índices de acidez, saponificação, iodo e peróxido, as propriedades de textura consistência e dureza, e o grau de transesterificação. Os resultados obtidos mostraram que os índices de acidez, saponificação, peróxido e iodo apresentaram uma dependência linear em função da fração mássica de gordura de leite tanto para a mistura das matérias-primas como para os produtos transesterificados. Com relação às propriedades de textura, modelos estatísticos foram compostos e permitiram verificar que a transesterificação promoveu uma diminuição tanto da consistência quanto da dureza nas misturas contendo proporções superiores a 50% de gordura de leite. As misturas preparadas com frações de gordura inferiores a este valor praticamente não sofreram alterações de textura após a reação. A avaliação da composição dos produtos obtidos demonstrou que as amostras correspondentes à gordura de leite pura e às misturas contendo 67 e 75% desta, apresentaram modificações no perfil de triacilgliceróis (TAGs), mostrando de forma geral diminuição na concentração dos TAGs C34-C38 e C54 e aumento dos TAGs C24-C30 e C44-C50. A reação empregando gordura de leite pura resultou em um grau de transesterificação (5,20%) mais elevado em relação ao óleo puro (3,33%), o que pode estar diretamente relacionado à especificidade da enzima pelos ácidos graxos presentes nesta gordura. O grau de transesterificação mais elevado (10,75%) foi obtido para a mistura contendo 67% de gordura de leite. Os resultados indicaram que a transesterificação de misturas contendo 50-67% de gordura de leite resultou em produtos com melhores propriedades de textura (espalhabilidade) sob temperatura de refrigeração em comparação à manteiga, à gordura de leite ou à simples mistura física com igual composição, sendo os valores de consistência e dureza pelo menos 32% menores em comparação à manteiga. Portanto, o desenvolvimento experimental deste trabalho permitiu demonstrar a viabilidade da transesterificação enzimática de uma mistura contendo elevados teores de gordura de leite, o que resultou na preservação de parte de suas características organolépticas, obtendo-se como resultado um produto com melhores características de textura (maior espalhabilidade) e nutricionais (maior teor de ácidos graxos insaturados). / The nutritional food characteristics have been highly considered by consumers due to the constant worries with their effects on health. However, the success of a new food product in the market depends also on its physical characteristics, such as spreadability, appearance and mouthfeel. These properties are very important for industrial processing and acceptability of the product by the consumer. In this context, the objective of present project was to assess the effect of the enzymatic transesterification of milkfat with soybean oil on the composition and texture of the obtained product. For this reaction Aspergillus niger lipase previously immobilized in polysiloxane-polyvinyl alcohol hybrid matrix was chosen as catalyst. The work was carried in three steps. Firstly, raw materials were characterized regarding their humidity, acid, saponification, peroxide and iodine indexes, rheological and thermal properties. Following these, raw materials were mixed at different proportions according to a statistical mixture factorial design and used as substrate in the transesterification reactions. The response variables were acid, saponification, iodine and peroxide indexes, texture properties (consistency and hardness) and transesterification yields. Acid, saponification, peroxide and iodine indexes showed a linear dependence on the milkfat mass fraction for both physical raw materials blends and transesterified products. Regarding the texture properties, statistical models were composed and allowed verifying that the transesterification reaction decreased both consistency and hardness for substrates containing more than 50% of milkfat. No alteration in these properties was noted for substrates containing lower milkfat fraction. The evaluation of the product compositions showed that samples containing pure milkfat and mixtures containing 67 to 75% of milkfat yielded modifications in the triacylglycerols profile (TAGs) such as decreased on the C34-C38 and C54 TAGs concentrations and increased on the C24-C30 and C44-C50 TAGs concentrations. Reactions carried with pure milkfat resulted in higher transesterification yield (5.20%) as compared with pure soybean oil (3.33%), and this was related to the specificity of the enzyme for fatty acids presented in the milkfat. The highest transesterification yield (10.75%) was obtained for mixtures containing 67% of milkfat. Theses results indicated that the transesterification reactions of substrates containing 50-67% of milkfat gave products having better properties regarding the texture (more spreadability) under cooling temperature than butter, milkfat or simple physical blend at the same composition. Consistency and hardness values were at least 32% lower than data for butterfat. Therefore, the proposed experimental work allowed demonstrating that the enzymatic transesterification of a mixture containing high milkfat levels besides partially preserving the milkfat organoleptic characteristics also rendering a product with better texture (higher spreadability) and nutritional properties (higher amount of unsaturated fatty acids).

Produção de biodiesel a partir do óleo de palmiste: aplicação de combi-lipases como biocatalisadores em reatores descontínuo e contínuo / Production of biodiesel from palm kernel oil: application of combi-lipases as biocatalysts in batch and continuous reactors

O presente trabalho teve como objetivo o aprimoramento da transesterificação do óleo de palmiste por rota etílica, por meio da atuação simultânea de lipases de diferentes fontes microbianas, visando a produção de biodiesel com teores residuais de monoacilglicerol (MAG) e diacilglicerol (DAG) dentro dos limites exigidos pela legislação ANP no 45/2014 para biocombustíveis. Para tal propósito, as lipases foram imobilizadas em suporte híbrido sílica-hidroxietilcelulose (SiO2- HEC) e os ensaios foram iniciados em regime descontínuo em reator de tanque agitado, temperatura fixa de 45 °C e razão molar óleo:álcool de 1:8. As lipases de Burkholderia cepacia, Pseudomonas fluorescens e Thermomyces lanuginosus, foram utilizadas de maneira isolada e em associação, com base no conhecimento prévio do bom desempenho destas enzimas para a produção de biodiesel. Conforme a capacidade de atuar em diferentes posições da molécula de triacilglicerol, as associações formaram combi-lipases de mesma especificidade (B. cepacia e P. fluorescens) e de diferente especificidade (B. cepacia e T. lanuginosus), com proporções de cada lipase variando de 75, 50 e 25% em relação ao seu par enzimático. Os melhores resultados foram observados quando utilizadas ambas as combinações em proporções equivalentes. Nestas condições, foram obtidas conversões similares (?98%) e ausência de DAG, com redução da viscosidade cinemática do óleo de palmiste de 30,35 para aproximadamente 4,00 mm2/s. Embora as conversões tenham sido próximas, a combi-lipase de diferente especificidade apresentou menor teor de MAG (1,68%, m/m), sendo escolhida como biocatalisador para o estudo em reatores de leito fixo em regime contínuo. Nesta etapa, foram realizados ensaios para testar os efeitos da razão molar óleo:álcool (1:12, 1:10 e 1:8) e do tempo espacial (16 e 14 h) na conversão, qualidade e produtividade do biodiesel. O melhor desempenho foi apresentado pela razão molar de 1:8 com tempo espacial de 16 h, proporcionando teor em MAG residual de 0,62% (m/m), conversão de 98% e produtividade de 30,9 mmolésteres.gcatalisador -1.h-1. Com relação à qualidade do biodiesel formado em termos de viscosidade cinemática (4,22 mm2/s), massa específica (870 kg/m3) e teor residual de MAG (0,62%, m/m), foi verificada adequação aos parâmetros exigidos pela ANP (3-6 mm2/s, 850-900 kg/m3 e <= 0,7%, m/m, respectivamente). / The objective of the present work was to improve the transesterification of palm kernel oil by ethanolic route through simultaneous action of lipases from different microbial sources, aiming to produce biodiesel with residual levels of monoacylglycerol (MAG) and diacylglycerol (DAG) within the limits required by ANP regulation 45/2014 for biofuels. For this purpose, lipases were immobilized on silica-hydroxyethylcellulose hybrid support (SiO2-HEC) and the assays were first carried out in a stirred tank batch reactor, at fixed temperature of 45 oC and oil -to-alcohol molar ratio of 1:8. The lipases from Burkholderia cepacia, Pseudomonas fluorescens and Thermomyces lanuginosus isolated and in association were used, based on the previous knowledge of the good performance of these enzymes for the biodiesel production. According to the ability to act at different positions of the triacylglycerol molecule, the associations formed combilipases of the same specificity (B. cepacia and P. fluorescens) and different specificities (B. cepacia and T. lanuginosus) at proportion of each lipase ranging from 75, 50 and 25% relative to their enzymatic pair. The best results were attained when both combinations were used at equivalent proportions. Under these conditions, similar conversions (?98%) and absence of DAG were obtained, which was consistent with the kinematic viscosity reduction of the palm kernel oil from 30.35 to 4.00 mm2/ s, approximately. Although similar conversions were attained, the combi-lipase of different specificities provided lower MAG contentes (1.68 wt%), being chosen as a biocatalyst for performing continuous runs in packed-bed reactor. In this step, the effects of the molar ratios oil-to-alcohol (1:12, 1:10 and 1:8) and space-times (16 and 14 h) on biodiesel production, quality and productivity were evaluated. The best performance was achieved by using molar ratio of 1:8 and space-time of 16 h, yielding residual MAG contents of 0.62 wt%, conversion of 98% and productivity of 30.9 mmolester.gcatalyst-1.h-1. The quality of biodiesel produced in terms of kinematic viscosity (4.22 mm2/s), specific mass (870 kg/m3) and residual MAG content (0.62 wt%) was found in accordance of ANP resolution (3-6 mm/ s, 850-900 kg/m3 and <= 0.7 wt%, respectively).

Os biolubrificantes têm incrementado progressivamente a sua importância sobre os lubrificantes derivados do petróleo devido às rigorosas regulamentações governamentais, o esgotamento das reservas de petróleo e especialmente pelo alto impacto e efeito tóxico dos óleos petroquímicos no ambiente. Os ésteres de óleos naturais podem ser usados como biolubrificantes e são obtidos por transesterificação dos triacilgliceróis com álcoois superiores. A transesterificação enzimática de óleos com álcoois metílico e etílico é um processo muito estudado para a produção de biodiesel, porém outros ésteres podem ser obtidos a partir de aceptores de acila de tamanho de cadeias carbônicas maiores, tais como os álcoois butílico e isoamílico ou misturas de diversos álcoois como o subproduto resultante destilação do bioetanol (óleo fúsel). Neste contexto, o presente trabalho de tese teve como objetivo a obtenção de ésteres com potencial lubrificante por transesterificação enzimática de óleo de palmiste com álcoois superiores. Os ensaios foram efetuados em regime descontínuo (reator de tanque agitado) e em regime contínuo (reator de leito empacotado) utilizando como biocatalisadores as lipases de Burkholderia cepacia e Pseudomonas fluorescens imobilizadas em suportes híbridos de SiO2-?CD e SiO2-HEC numa temperatura fixa de 45°C. Foram testados os efeitos da razão molar óleo: álcool etílico, butílico, isoamílico e óleo fúsel simulado (mistura de álcoois) (1:4 a 1:12), tempos de reação em batelada (24h a 72h) e tempos espaciais em fluxo contínuo (4 a 14h) na conversão, produtividade e a qualidade dos ésteres. Os resultados mostraram bom desempenho das lipases e conversões totais foram alcançadas. No entanto, foi possível observar o efeito do tamanho da cadeia do aceptor de acila, no excesso requerido e no tempo de reação, obtendo-se os melhores resultados para o processo descontínuo com razões molares de 1:6 e 72h para álcool etílico, 1:6 e 48h para ésteres butílicos, 1:4 e 48h para álcool isomílico e óleo fúsel simulado. No processo contínuo, o melhor desempenho foi determinado para razões molares de 1:8 e tempo espacial de 10h para etanol, 1:6 e 8h para butanol, 1:4 com 6h e 8h para isoamílico e óleo fúsel simulado, respectivamente. Em termos gerais, produtividades mais elevadas foram atingidas em regime de fluxo contínuo e especificamente com álcool de isoamila e óleo fúsel (111,10 e 130,18 mg de éster.gcatalisador-1.h-1). Com relação à qualidade dos produtos obtidos nas reações que forneceram elevadas conversões foi constatado baixos teores de monoacilgliceróis (<=1%) e ausência de diacilgliceróis. As viscosidades cinemáticas a 40 °C, confirmaram o elevado grau de transesterificação do óleo de palmiste modificando a viscosidade inicial do óleo de 30,13 mm2.s-1 para valores na faixa entre 3 a 6 mm2.s-1. O índice de viscosidade variou entre 114 e 150 e a estabilidade oxidativa (período de indução) entre 22 e 37 min (9 e 20 h, método Rancimat). Os resultados obtidos foram bastante satisfatórios e contribuíram para o estabelecimento de condições reacionais e operacionais para a produção enzimática de ésteres com potencial uso como lubrificantes, tanto em regime descontínuo como contínuo utilizando óleo de palmiste e álcoois e misturas de álcoois superiores. / Biolubricants have progressively increased their importance on oil-based lubricants due to stringent government regulations, the depletion of oil reserves and especially the high impact and toxic effect of petrochemical oils on the environment. Natural fatty acid alkyl esters can be used as biolubricants and are obtained by transesterification of triacylglycerols with higher alcohols. The enzymatic transesterification of oils with methyl and ethyl alcohols is a much-studied process for the production of biodiesel. However, other esters may be obtained using higher chain acyl acceptors such as butyl and isoamyl alcohols or mixtures thereof as the fusel oil, resulting from the production of bioethanol. In this context, the present thesis aimed to obtain esters with lubricating potential by enzymatic transesterification of palm kernel oil with higher alcohols. Transesterification runs were performed under batch mode (stirred reactor) and continuous flow (packed bed reactor) using as biocatalysts the lipases from Burkholderia cepacia and Pseudomonas fluorescens immobilized on hybrid matrixes SiO2-?CD and SiO2-HEC at 45 °C. The effects of oil to ethyl, butyl, isoamyl alcohol and synthetic fusel oil (1: 4 to 1:12) molar ratios, batch reaction times (24h to 72h) and special times (4 to 14 h) were tested in the conversion, productivity and quality of final esters. The results showed good lipase performance and 100% bulk conversions were achieved. However, it was possible to observe the effect of the acyl acceptor chain on the required excess and on the reaction time, obtaining the best results for batch process with molar ratios of 1: 6 and 72 h for ethyl alcohol, 1: 6 and 48 h for butyl, 1: 4 and 48 h for isoamyl alcohol and synthetic fusel oil. In general terms, better productivities were obtained under continuous flow, particularly with isoamyl alcohol and fusel oil (111.10 and 130.18 mg of ester.g catalyst-1.h-1). Regarding the product quality, low monoacylglycerol contents (<=1% ) and absence of diacylglycerols were found in products obtained under the best reaction conditions. The kinematic viscosities at 40 °C confirmed the high degree of transesterification of palm kernel oil by modifying its initial viscosity from 30.13 mm2 s -1 to a value in the range of 3 to 6 mm2 s -1, the viscosity index between 114 and 150 and an oxidative stability (induction period) between 22 and 37 min (9 and 20 h, Rancimat method). The results obtained were very satisfactory and contributed to the establishment of reactional and operational conditions for the enzymatic production of esters with potential use as lubricants both in discontinuous and continuous flow mode using palm kernel oil and alcohols and mixtures of higher alcohols.
