An evaluation of the homoeopathic drug proving of Suiherlandia fruiescens in the light of a doctrine of signatures analysis

The second objective was to analyze the symptoms obtained from this proving according to the doctrine of signatures. It was hypothesized that Sutherlandia frutescens 30CH would produce observable signs and symptoms in healthy volunteers which would correlate to a doctrine of signatures analysis of the plant. A double blind proving of Sutherlandia frutescens 30CH was conducted. 24 provers were selected and randomly divided into two groups, the first consisted of 18 provers who received medicated powders and the second consisted of the remaining six who received placebo. The subjects were unaware of the nature of the substance that they took nor the potency thereof as an added control measure. Provers were examined and kept journals before, during and after administration of the remedy so as to serve as their own control. This information, along with data collected by the researchers from case histories and physical examination, was then assessed on completion of the proving. The researchers then translated the symptoms elicited into Materia Medica and repertory language and formulated a homoeopathic picture of this remedy. A further aim of the investigation was to compare those remedies yielding the highest numerical value and total number of rubrics on repertorisation of the proving symptoms to Sutherlandia frutescens. It was hypothesised that the 30CH potency of Sutherlandia frutescens would produce clearly observable signs and symptoms in healthy provers, and that the comparison of Sutherlandia frutescens to those remedies yielding the highest numerical value and total number of rubrics on repertorisation of the proving symptoms would highlight differences and similarities between the remedy symptoms so that confusion as to indication is erased. It was hypothesised that a fuller understanding of Sutherlandia frutescens and its relationship to other remedies would be gained following this comparison. III A double blind, placebo controlled proving of Sutherlandia frutescens 30CH was conducted on twenty-four healthy volunteers who met the inclusion criteria. Six of the twenty-four provers randomly received placebo, with neither prover nor researcher knowing who was on placebo. Provers had a homoeopathic case history taken and physical examination performed on them before commencement of the proving. The provers recorded their signs and symptoms before, during and after administration of the remedy, by means of a journal. On completion of the proving, the information was correlated and assessed by the four researchers, the symptoms elicited translated into Materia Medica and Repertory language, and a homoeopathic picture of the remedy formulated. The second objective was to analyze the symptoms obtained from this proving in a direct comparison to the effects of the major pharmacologically active compounds present in Sutherlandia frutescens. A double blind proving of Sutherlandia frutescens 30CH was conducted. Twenty-four provers were selected and randomly divided into two groups, those receiving medicated powders (18 subjects) and those receiving the placebo powders (6 subjects). As an added control measure, the subjects were also ignorant to both the nature of the proving substance and the administered potency. Prior to taking the remedy, each patient provided their own case history and received a physical examination so as to establish each individual's baseline. Each prover was then required to keep a daily journal, in which all symptoms were recorded in accordance with a suggested guide. The researchers then collated the data and translated the symptoms produced into Materia Medica and repertory language. Finally a homoeopathic picture of the remedy emerged in which marked themes exist. It was also the purpose of this study to compare all existing indications of use of Sutherlandia frutescens to the proving symptomatology. This would allow a greater understanding of the plants spheres of action and would contribute to the formation of a comprehensive remedy picture by highlighting similarities and differences between the homoeopathic and traditional indications of use. The comparison also served to investigate whether Sutherlandia frutescens acts, in accordance with the fundamental law of homoeopathy, the law of similars, in traditional application. It was the intention of the study to shed light on the possible 'homoeopathicity', or not, of current traditional use of Sutherlandia frutescens, as opposed to overt pharmacology of phytochemicals. It was hypothesized that Sutherlandiafrutescens 30CH would produce clearly observable signs and symptoms in healthy provers. The second hypothesis was that the proving symptomatology of Sutherlandia frutescens 30CH would be similar to the existing indications of use. The homoeopathic proving was a double blind placebo controlled study conducted by four Master's in Technology: Homoeopathy students. A total of 24 subjects formed the proving group, 25% of whom (6 of the 24) were randomly assigned to the placebo group. The subjects were unaware of the substance they were proving and the potency of the substance to be proven. The provers also served as intra-individual controls by recording their state prior to the administration of the remedy, to provide a baseline for comparison after the administration of the remedy. Immune modulatory effect of Dichrostachys cinerea, Carpobrotus dimidiatus, Capparis tomentosa and Leonotis leonurus

Submitted in fulfillment of the requirements for the Degree of Master of Technology: Biotechnology, Durban University of Technology, 2009. / Dichrostachys cinerea, Carpobrotus dimidiatus, Capparis tomentosa and Leonotis leonurus are all plants that are indigenous to South Africa. These plants are used in traditional medicine to treat various ailments. However, there is little or no scientific data to justify these traditional uses. Furthermore, it is difficult to reconcile traditional knowledge with scientific evidence because of the overwhelming targeting of signal-responsive systems by plant defensive compounds, multiple sites of action and the connectedness of the signaling pathways, which provide many cures and have pleiotropic effects. In order to evaluate the action spectrum of these plants, and validate its widespread use, this research evaluated the antibacterial, antioxidant, anti-inflammatory, anti-mosquito and immunomodulatory properties of these plants. Antimicrobial activity of the extract was determined by evaluating the bactericidal and fungicidal action using the agar disc diffusion assay. Anti-oxidative properties of the extracts were tested using the DPPH photometric assay. Anti-inflammatory properties were carried out using the 5-lipoxygenase assay. The larvicidal, repellency and insecticidal assay was determined against A.arabiensis. The safe use of these plant extracts was determined by evaluating toxicity, a brine shrimp lethality assay and an in vitro cell culture system using human myelogenous leukemia cell line. Potential carcinogenic activity was evaluated using the Ames Salmonella Mutagenecity assay. The immunomodulatory activity of the extracts on human peripheral blood mononuclear cells 6 was evaluated on freshly harvested lymphocytes using the MTT assay. Cytokine response was evaluated by measuring the secretion of interferon-gamma and interleukin-10. Elucidation of the B cells, T cells, activated T cells, CD 4+, CD 8+ and NK cells was performed by flow cytometry. The extracts showed anti-microbial activity against Escherichia coli, Pseudomonas aeruginosa, Klebsiella oxytoca, Salmonella typhimurium, Serratia marcescens, Bacillus cereus and Tricoderm sp. The highest activity was shown by methanolic and aqueous extracts of L. leonurus leaves followed by methanolic and aqueous extracts of D. cinerea. Extracts of C. tomentosa and D.cinerea demonstrated a higher degree of free radical scavenging than rutin, which was used as a standard indicating that these plants have strong antioxidant properties. None of the plants showed significant anti-inflammatory activity when compared to NDGA. In the anti-mosquito assays, the extracts showed strong repellency and insecticidal activity. L. leonurus extracts demonstrated the highest insecticidal and repellency activity against the mosquito, and was also found to cause ‗knockdown‘ and mortality. The extracts display no toxicity, cytotoxicity and mutagenicity. The immunological studies for immune modulation showed that the methanol extracts of these plants induce a Th1- predominant immune response because they significantly suppressed the secretion of IL-10 and augment IFN-γ production, which are hallmarks used to indicate a stimulation of the innate immune response. The disc diffusion method was employed. W somnifera is indigenous to southern Africa and its use is well established amongst the traditional healers for many varied complaints. Infusions, decoctions and tinctures of the fresh and dry whole root are used. (Gericke and Van Wyk, 2000:150.) For this study 20 plates of Mueller-Hinton agar were inoculated with each bacteria, resulting in a total of 100 plates. Four dry discs previously impregnated with the test substances and two antibiotic discs were equidistantly placed on each plate and incubated at 3rC. The vancomycin and gentamycin discs were included to account for plate-to-plate variations in the sensitivity of the bacteria to the antimicrobial substances. The plates were observed at 18, 24 and 48-hour intervals. ' Statistical analysis was performed using the Friedman test to compare test and control substances at each observation interval. Mini-dissertation submitted in partial compliance with the requirements for the Master's Degree in Technology: Homoeopathy, Durban Institute of Technology, 2003. / The purpose of this study was to determine the efficacy of Alepidea amatymbica tincture and homoeopathic dilutions to the 1st and 6th decimal potency as compared to ethanol (negative control) in the in vitro inhibition of Escherichea coli, Pseudomonas aeruginosa, Klebsiella pneumoniae, Staphylococcus aureus and Bacillus cereus in terms of the disc diffusion test. Vancomycin and gentamicin were included in the study as positive controls in order to account for plate-ta-plate variations in the sensitivity of the bacterial strains to antimicrobial substances. Antimicrobial activity was expressed as the ratio of the inhibition zone (mm) produced by the test substance and the inhibition zone (mm) produced by the two combined antibiotic discs. For this study 20 Mueller-Hinton agar plates were assigned to each bacterial species and were inoculated with their respective bacteria. Four dry filter paper discs and two antibiotic discs were placed equidistantly on each agar plate. The filter paper discs had been previously impregnated with one of the test substances or the negative control using a triple impregnation technique utilizing a micro-pipette. The plates were then incubated at 37\xB0 C. The diameters of the zones of inhibition were measured at 18 hours, 24 hours and 48 hours. Data was analysed by means of the Statistical Package for Social Sciences (SPSS). Statistical methods utilized were Friedmans' test, Mann-Whitney U test, and Kruskall - Wallis Non-Parametric Analysis of Variance by Rank test. / M

The phytogeography of the Sneeuberg, Nuweveldberge and Roggeveldberge (Great Escarpment): assessing migration routes and endemism

The Great Escarpment forms a semi-continuous mountain system 5 000 km long, stretching from Angola in the north-west, south through Namibia, and into western, southern and eastern South Africa, including Lesotho and Swaziland. It is composed of a wide variety of geological suites but is unified in representing the edge of the African plateau and the passive Gondwanan continental margin. The Great Escarpment falls into all major climatic zones on the subcontinent, is a repository of palaeo- and neo-endemics, hosts more than half of southern Africa‟s centres of plant endemism, and has a rich suite of endemic fauna. In addition, the Great Escarpment is believed to be both a refugium and corridor for biological diversity. Despite the biological richness of the Great Escarpment, research to date has been fragmented and many sections of the Great Escarpment have not been studied. The aim of this study is to contribute to research on the Great Escarpment by undertaking a detailed floristic study of the southern Great Escarpment (the Sneeuberg, Nuweveldberge and Roggeveldberge). Together these mountains comprise approximately 1 000 km (one fifth) of the Great Escarpment, and occupy a transition zone between the summer rainfall zone in the east and the winter rainfall zone in the west. They are also the sections of Great Escarpment most closely situated to the Cape Floristic Region (CFR) and would thus be involved in hypothesised migration routes for lineages that also occur further north through the Drakensberg Alpine Centre (DAC) to the East African mountain chain. Detailed fieldwork of the southern Great Escarpment was undertaken over a period of four years in all seasons. Approximately 8 000 specimens were collected. Particular emphasis was placed on areas that may represent refugia, i.e. the highest plateaux and peaks, mesic areas and cliff-lines. An overview of each mountain range, together with their endemic plant species and phytogeography, is provided. Approximately ten new species have been discovered during this study, two of which have been described to date. Numerous endemics only known from their types have also been rediscovered. The Sneeuberg is defined as a new centre of plant endemism on the Great Escarpment (endemism of 2.3%), and the role of the Boschberg and Groot-Bruintjieshoogde (part of the Sneeuberg) as a nexus for floristic migration routes is discussed. The Nuweveldberge is shown to have low endemism despite a floristic tally similar to the Sneeuberg, while the Roggeveldberge are confirmed to be the most endemic-rich section of the southern Great Escarpment. The field data collected was augmented by available data in taxonomic revisions, and floras for the Sneeuberg, Nuweveldberge and Roggeveldberge were compiled. In order to floristically compare the southern Great Escarpment with other sections of the Great Escarpment and the CFR, a database of some 12 000 taxa was created using available floristic data for the CFR, DAC and Great WinterbergAmatolas, together with the data collated for the Sneeuberg, Nuweveldberge and Roggeveldberge. These data were analysed using phenetic methods and Parsimony Analysis of Endemicity (PAE). The results indicate stronger linkages in the east, particularly between the Sneeuberg and Nuweveldberge, and between the Sneeuberg and the Great WinterbergAmatolas. The relationship of the Roggeveldberge with the rest of the southern Great Escarpment remains ambiguous. In order to refine notions of connectivity and migration routes, 19 well-sampled phylogenies were assessed for sister-taxon disjunctions to explore CFRGreat Escarpment connections. Palaeo-connectivity between the CFR and southern Great Escarpment is most strongly supported for the south-eastern (SE) connection, and less so for the north-western (NW) and Matjiesfontein connections. There is support for the current (or recent) use of these three connections from numerous species that occur on both sides of the connections. Results of these analyses indicate that the southern Great Escarpment is a palaeo-corridor, the functioning of which has been broken by the aridification of the Nuweveldberge since the Last Glacial Maximum (LGM). Floristic connectivity is strongest in the east, from the Nuweveldberge to the DAC, and is less so in the west between the Nuweveldberge and the Roggeveldberge a finding attributed to the transition from a reliable winter rainfall regime on the Roggeveldberge to an unpredictable moisture regime on the Nuweveldberge. The mountains of the southern Great Escarpment are thus a series of refugia from a previous moister, cooler climate and are a corridor between the eastern and western components of the Great Escarpment. The SE connection is the primary link between the CFR and the eastern Great Escarpment Afromontane region in southern Africa. The implications of this research are that accurate conservation assessments and Red Data listings for many of the previously poorly-known endemics can now be made, and appropriate conservation measures implemented. Climate change remains the primary threat to these endemics and montane taxa in general, while degradation of wetlands is the primary threat to the water catchment service provided by the southern Great Escarpment. Future detailed research on the Great WinterbergAmatolas and Stormberg and a comprehensive flora of the HantamRoggeveldberge will further enhance our understanding of the floristics of the southern Great Escarpment, and provide the necessary data for comprehensive GIS-based models of proposed climate change scenarios for local, regional and national conservation planning.

A taxonomic revision of the southern African endemic genus Gazania (Asteraceae) based on morphometric, genetic and phylogeographic data

Gazania is a small genus of the subtribe Gorteriinae, tribe Arctoteae, that is endemic to southern Africa. The genus was last revised in 1959 by Roessler, who noted that delimitation of the species of Gazania can be “extraordinarily difficult”. Morphometric data was collected to test the reality of the 16 species as delimited by Roessler, who based species boundaries on morphological characters. Only six taxa were found to be morphologically distinct, while the remaining samples showed no species cohesion. DNA sequence data from two nuclear spacer regions (ITS and ETS) and four chloroplast noncoding regions (the trnL and rpS16 introns, and the psbA-trnH and trnL-F spacers) of 43 samples were utilised to create a species level phylogeny and to investigate correlations between genetically delimited units and morphologically defined taxa. DNA sequence data reveal that seven species (as delimited by Roessler) are morphologically and genetically distinct. The remaining nine of Roessler’s species fall into a morphologically and genetically overlapping continuum that forms an ochlospecies. Phylogeographic methods (based on an expanded ITS and ETS DNA sequence data set from 169 samples) were employed to further resolve the limits of species, with special focus on the clades within the ochlospecies. These genetically defined clades were correlated with their geographical distributions, and in combination with molecular dating techniques, used to elucidate the recent climatic or environmental factors that may have shaped the phylogeographic structure of the genus. Phylogeographic patterns and molecular dating reveals that the genus Gazania is an example of a South African endemic clade that has undergone episodic cladogenesis in response to fluctuating climatic conditions over the last seven million years. The ochlospecies within Gazania is a result of repeated cycles of climate driven isolation in refugia and subsequent expansion and hybridization events during the Pliocene and Pleistocene. Comparisons with phylogeographic studies on other organisms reveal a common pattern indicative of the presence and evolutionary importance of an ancestral refugium in the arid Richtersveld / Namib region of southern Africa.
