The periodicity of formation of growth increments in the otoliths of South African sparids was validated by the oxytetracycline labelling technique. Intramuscular injections of oxytetracycline at a dosage of 250mg/kg marked the otoliths of laboratory held juvenile sparids, while a dosage range of 50 - 100mg/kg oxytetracycline, injected intramuscularly, marked adult sparids in the field. Laboratory held, larval sparid otoliths were marked by immersion for 24hrs a solution of 100-150mg/l alizarin complexone. Both daily (microstructural) and annual (macrostructural) growth increments were identified in the otoliths. The micro incremental pattern of deposition in the sparid otoliths conformed to the general pattern of otolith structure. Sparid otoliths consisted of a central opaque nucleus composed of multiple primordia. surrounding this nucleus were daily increments which decreased in width as the distance from the nucleus increased. Both check rings and subdaily increments were visible throughout the otolith. Minor environmental changes did not affect micro incremental deposition. Narrow opaque and wide hyaline annual growth zones were identified in sectioned sparid otoliths. Scanning electron microscope analysis of the annual growth zones revealed that microincrements within the opaque zone were narrowly spaced with prominent discontinuous phases. This resulted in the greater optical density and higher protein content of the zone. The hyaline zone was composed of widely spaced daily increments with prominent incremental phases accounting for the translucent nature of this zone. Opaque zone formation in the otoliths of many South African sparids was found to occur primarily during periods of reproductive activity and was shown to be indicative of slow otolith growth. The hyaline zone was formed after the spawning season, representative of fast otolith growth. The results of this study have resolved much of the controversy surrounding the rate of growth and time of formation of the opaque and hyaline growth zones in South African sparid otoliths.

Settlement, distribution, growth, and mortality of juvenile dover sole (Microstomus pacificus) off northern Oregon

Demographic characteristics of juvenile Dover sole (Microstomus pacificus) were examined in relation to physical and biotic factors to develop a hypothesis explaining annual variations in recruitment. Surveys were conducted along the Oregon continental shelf at depths between 50-400 m bimonthly during 1989 and annually (in March) between 1990 and 1993. To determine settlement timing, growth, and mortality, daily growth increments were validated, as was position of the first post-settlement annulus. A method of estimating precision of otolith elemental composition with the electron microprobe was developed; otolith microstructure and microchemistry were used to identify a landmark associated with settlement. Mean density of 0-group settlers varied annually, with 1989 and 1990 high, 1991 and 1992 low, and 1993 intermediate. 0-group abundance was correlated with fall offshore divergence one year before settlement and with southward geostrophic velocity during the summer before settlement. These conditions may be related to production of prey available to pelagic larvae. Density of 0-group Dover sole was highest between 100-119 m, but deeper areas were also utilized in years of highest abundance. Density was not correlated with spawning stock biomass or density of older juveniles. It was correlated with rex sole (Glyptocephalus [Errex] zachirus) density. Growth rate was positively correlated with mean April-October upwelling and was not related to density or mortality of Dover sole. First-year mortality was positively correlated with early settlement of 0-group larvae, possibly due to an advantage of settling and migrating to the primary nursery area well in advance of the spring transition. A multiple regression model predicting density of 1-group Dover sole from three physical environmental factors was applied to an independent time series of Dover sole "age-5" recruitment estimates (Turnock and Methot 1992). The model was a poor predictor of annual recruitment to fisheries but a good predictor of trends based on five-year running averages. Results suggest that multi-year trends in Dover sole recruitment to fisheries can be explained by a small set of density-independent physical factors. / Graduation date: 1995

Age, growth and yield-per-recruit analysis of ndunduma Diplotaxodon limnothrissa (Teleostei: Cichlidae), in the southeastern arm of Lake Malawi

Diplotaxodon limnothrissa Turner (1995) is a widely distributed species occurring throughout Lake Malawi, extending from the surface to a depth of at least 220m. It is probably the most abundant cichlid in the lake with biomass estimates of around 87 000 tonnes in the pelagic zone alone. The species is exploited commercially in the southern part of the lake but since its inception the fishery has never been assessed. As such this study investigates some aspects of age and growth of the species besides applying a yield-per-recruit analysis to assess the status of the fishery. Analysis of sectioned sagittal otoliths revealed that D. limnothrissa is fast growing and relatively long-lived species, attaining ages in excess of 10 years. Growth in length was rapid in immature fish, with fish attaining almost half of their maximum size within their first year. Le ngth-at-age was described by the von Bertalanffy growth model with combined-sex growth described as Lt = 211.21(1-exp(- 0.24(t+1.36))) mm TL. Total, natural and fishing mortalities were estimated at 0.76 yr⁻¹, 0.31 yr⁻¹ and 0.45 yr⁻¹ respectively. Per-recruit analysis indicated that the D. limnothrissa stock in the southeast arm of the lake is fully exploited as indicated by the current spawner biomass-per-recruit ratios of 31-55% (SB/R)F=0. Modelling indicated that the current age-at-capture (2.67 years) is lower than the age at which yield is optimised (> 5 years) based on the F₀·₁ harvesting strategy. It is, therefore, recommended that the age-at-capture should be increased from 2.67 to 5 years to optimise yield.

Lead-radium dating of two deep-water fishes from the southern hemisphere, Patagonian toothfish (Dissostichus eleginoides) and Orange Roughy (Hoplostethus atlanticus)

Patagonian toothfish (Dissostichus eleginoides) or "Chilean sea bass" support a valuable and controversial fishery, but the life history is little known and longevity estimates range from ~20 to more than 40 or 50 yr. In this study, lead-radium dating provided validated age estimates from juveniles to older adults, supporting the use of otoliths as accurate indicators of age. The oldest age groups were near 30 yr, which provided support for age estimates exceeding 40 or 50 yr from grow zone counts in otolith sections. Hence, scale reading, which rarely exceeds 20 years, has the potential for age underestimation. Lead-radium dating revealed what may be minor differences in age interpretation between two facilities and findings may provide an age-validated opportunity for the CCAMLR Otolith Network to reassess otolith interpretations. Orange roughy (Hoplostethus atlanticus) support a major deep-sea fishery and stock assessments often depend on age analyses, but lifespan estimates range from ~20 to over 100 yr and validation of growth zone counts remained unresolved. An early application of lead-radium dating supported centenarian ages, but the findings were met with disbelief and some studies have attempted to discredit the technique and the long lifespan. In this study, an improved lead-radium dating technique used smaller samples than previously possible and circumvented assumptions that were previously necessary. Lead-radium dating of otolith cores, the first few years of growth, provided ratios that correlated well with the ingrowth curve. This provided robust support for age estimates from otolith thin sections. Use of radiometric ages as independent age estimates indicated the fish in the oldest group were at least 93 yr. Lead-radium dating has validated a centenarian lifespan for orange roughy. To date, radium-226 has been measured in otoliths of 39 fish species ranging from the northern Pacific and Atlantic Oceans to the Southern Ocean. In total, 367 reliable radium-226 measurements were made in 36 studies since the first lead-radium dating study on fish in 1982. The activity of radium-226 measurements ranged over 3 orders of magnitude (<0.001 to >1.0 dpm.g⁻¹). An analysis revealed ontogenetic differences in radium-226 uptake that may be attributed to changes in habitat or diet. Radiometric age from otolith core studies was used to describe a radium-226 uptake time-series for some species, which revealed interesting patterns over long periods. This synopsis provides information on the uptake of radium-226 to otoliths from an environmental perspective, which can be used as a basis for future studies.
