Studies on Secondary Metabolites from Soft Coral Sinularia gyrosaChuang, Cheng-ta 26 August 2009 (has links)
Numerous bioactive secondary metabolites have been isolated from the
soft coral in recent years. In order to search for more bioactive secondary
metabolites, we have investigated the chemical constituents of the organic
extracts soft coral Sinularia gyrosa, collected at Dongsha island.
Chromatography separation have led to the isolation of six new
norcembranoid compounds, gyrosolides A¡VF (1¡V6), and two new
diterpenoids, gyrosanols A and B (7 and 8), as well as eleven known
norcembranoids (9¡V19). The chemical structures and configurations of
pure compounds were determined by NMR spectroscopic (1H NMR, 13C
NMR, 1H¡V1H COSY, HMQC, HMBC, NOESY, UV, IR, CD, and mass
spectra) and physical data (optical rotations), 3D Chem Draw MM2 force
field calculations, as well as comparison with the spectroscopic data
reported by literature. The structure of known compound 14 was revised
by NOESY and 3D Chem Draw MM2 force field calculations. The
anti-inflammatory effects, cytotoxicity aganist of P-388 (mouse
lymphocytic leukemia), HT-29 (human colon adenocarcinoma), and
A-549 (human lung epithelial carcinoma) cells, and anti-HCMV (human
cytomegalovirus) activity of these secondary metabolites were evaluated.
Metabolite 9 exhibited significant activity against HCMV and
cytotoxicity against A-549 cell lines, and significantly exhibited the
COX-2 protein expression by LPS stimulation.
