Moench : phytochemical exploration, biotechnological approach and dermocosmetic valorization.

L'étude phytochimique d'Armeria maritima et d'Helichrysum stoechas a permis d'isoler pour la première fois 31 molécules dans le genre Armeria dont 4 nouveaux flavonols diglycosylés, ainsi que le développement d'une stratégie de déréplication pour l'étude d'H. stoechas. Dans les deux espèces, nous avons relevé une richesse en polyphénols, qui pourraient être extraits par des techniques respectueuses de l'environnement comme la SFE. En parallèle, ces deux halophytes ont montré un fort potentiel biologique avec des extraits et des molécules dotés d'activités anti-oxydante, anti-collagénase, anti-inflammatoire ou encore cicatrisante. De plus, nous avons initié pour la première fois des suspensions cellulaires d'A. maritima et identifié des éliciteurs comme le méthyl jasmonate permettant d’augmenter dans les cellules d'H. stoechas la teneur en acide 3,5-dicaféoylquinique, un bio-marqueur de l'activité anti-inflammatoire. La production de molécules bioactives dans des cultures végétales in vitro pourrait par la suite être transposée à plus grande échelle, afin d’amplifier le potentiel de valorisation de ces deux halophytes en dermo-cosmétique. / The phytochemical study of Armeria maritima and Helichrysum stoechas led to the isolation of 31 molecules never reported before in the genus Armeria, 4 of which being new flavonol diglycosides, and to the development of a dereplication strategy for the study of H. stoechas. In both species, an abundance of polyphenols was observed, which could be extracted with eco-friendly methods like SFE. Both halophytes showed a strong biological potential as their extracts and molecules demonstrated antioxidant, anti-collagenase, anti-inflammatory and wound healing activities. Moreover, we initiated for the first time cell suspensions of A. maritima, and identified elicitors, such as methyl-jasmonate, which led to H. stoechas cell suspensions with an increased content in 3,5-dicaffeoylquinic acid, a bio-marker of anti-inflammatory activity. The production of bioactive molecules in "plant cell factories" could be scaled-up to enhance the valorization potential of both halophytes in dermocosmetics.

The Development of Marine Aquaponics

Integrated aquaponic food production systems are capable of producing more food on less land using less water than conventional food systems, and marine systems offer the potential of conserving freshwater resources. However, critical factors such as suitable species combinations, environmental conditions (salinity and pH), and nutrient management (animal to plant ratio, C/N ratio, and dietary crude protein) have not been fully understood for marine systems. There were four objectives in this project. The first objective was to evaluate the growth performance of potential comparable combination (whiteleg shrimp with three halophytic plants) for the development of marine aquaponics with BFT under different salinities. The second objective was to evaluate stocking densities and the C/N ratio on growth and production of whiteleg shrimp and three halophytes. The third objective was evaluation of varying concentrations of dietary crude protein in practical diets fed to shrimp raised in biofloc aquaponic saltwater systems. The fourth objective was to evaluate effects of pH levels and additional C on the growth and production of whiteleg shrimp and five plant species in marine aquaponics. Four conclusions were determined: 1) Regarding marine aquaponics, whiteleg shrimp and the three halophytes (Atriplex hortensis, Salsola komarovii, and Plantago coronopus) are suitable combinations for future development. According to the research results, shrimp performed better in a salinity of 15 and 20 ppt; yet, plants performed better in a salinity of 10 and 15 ppt. Therefore, a salinity of 15 ppt is suggested as the optimal saline condition for shrimp and the three halophytes in an indoor marine aquaponics system. In addition, inoculating probiotics do have the efficiency of stabilizing water quality, cultivating microbial community, and enhancing the health of shrimp and plants in the operation of aquaponics. 2) The stocking density ratio and C/N ratio exerted significant impacts on the performance of shrimp and plants in marine aquaponics. Shrimp performed better with the stocking density of 2:1 and 3:1, with no impact from the C/N ratio. Conversely, plants performed better with the stocking density of 3:1 and 5:1 with the C/N ratio at 15. Therefore, a stocking density ratio of 3:1 with a C/N ratio at 15 is suggested as the optimal condition for shrimp and the three halophytes in an indoor marine aquaponic food production system. Inoculating the water with biofloc and applying probiotics regularly can enhance the management of water quality and the health of shrimp and plants in aquaponics. 3) Among the findings of the study, shrimp growth was not affected by the protein content of the feed, suggesting that it is possible to use feeds with lower protein concentration when culturing shrimp in biofloc-based marine aquaponics. However, plants grew better in the treatments with higher protein content feed in the early and middle stages of production. Hence, for maximum production, providing a higher protein concentration feed (35 %) in the early stages of system start-up, and switching to a lower protein concentration feed (30 %) in the later stages of cultivation might be feasible. 4) The current study found no significant effects of pH or additional C on shrimp performance. In contrast, plants grew better in lower pH treatments, while additional C supplements improved the performance of plants grown in higher pH treatments and had similar results to the lower pH treatments. We suggest that RO water is not suitable source of water for shrimp-based marine aquaponics if ionic composition is not managed. The addition of C, however, led to improved growth and yields of most plants. Hence, adding C can be a promising approach in marine aquaponics to enhance the resistance to the abiotic stress of plants and improve their growth.<div> <br>The present study on marine aquaponics has produced important findings that will fill some knowledge gaps, provide management guidelines for production, and facilitate its development. <br></div>

Studium autekologie vybraných taxonů křídových rostlin pomocí izotopů uhlíku / Autecological study of selected Cretaceous plants using stable Carbon isotopes

1 Abstract This thesis presents an analysis of fossil plants from the Cenomanian Peruc-Korycany Formation of the Bohemian Cretaceous Basin and from the Bückeberg Formation of the Lower Saxony Basin in Germany. Based on earlier studies, both areas provide sediments that are considered to have developed in tidally influenced fluvial systems. Studied fossil plants are represented by ginkgoalean plant leaves (Ertemophyllum, Tritaenia), branches of conifers (Frenelopsis) and lauroid angiosperms (Eucalyptolaurus). Frenelopsis, Eretmophyllum and Tritaenia are considered to be halophytic plants, while Lauroid angiosperms were considered to grow in fresh water conditions. The fossil plants were studied using cuticle analysis and two methods of stable carbon isotope analysis: Bulk carbon isotope analysis and Compound Specific Isotope analysis. For cuticle analysis samples were observed and documented macroscopically and microscopically. To specify the environmental conditions, recent samples from three salt marshes in Great Britain were studied and analysed using the same methods as the fossil samples. The data from all observations and measurements were processed and their interpretation supported the modelled environment based on the sedimentological data. Traitement du ruissellement routier chargé en sels de déglaçage et en métaux lourds par un marais épurateur construit et adapté

Depuis la construction de l’autouroute 40 dans les années 70 (tronçon Félix-Leclerc), le lac Saint-Augustin s’est grandement enrichi en sels de déglaçage et en métaux lourds. Un marais épurateur construit et adapté (MECA), dans lequel ont été intégrées des plantes halophytes, a donc été implanté afin traiter une partie des eaux de ruissellement provenant de l’autouroute. Le comportement et l’efficacité de ce dernier ont été évalués afin de déterminer si cette technologie était en mesure de pallier à la problématique de contamination. Les observations et les analyses ont permis d’établir que les plantes halophytes sont mal adaptées aux conditions environnementales du MECA et que seulement environ 1% des sels sont emmagasinés dans les plantes. Les concentrations mesurées dans les eaux ont montré que le MECA est efficace pour emmagasiner du sel de déglaçage, principalement en période de temps sec, mais que les quantités accumulées sont peu significatives par rapport aux quantités introduites. / Following the construction of Highway 40 in the 1970s (Félix-Leclerc section), concentrations of de-icing salts and heavy metals in Saint-Augustin Lake have greatly increased. An adapted constructed wetland (MECA) in which halophyte plants were integrated was installed in order to treat part of the highway runoff. The behavior and the effectiveness of the MECA were evaluated to determine if it is an appropriate technology to mitigate the problem of de-icing salts and heavy metals contamination. Observations and analyses have allowed us to establish that halophyte plants are generally not adapted to the environmental conditions of the MECA and that approximately only 1% of the de-icing salts get stored in the plants. Concentrations measured in the water indicate that the MECA is effective at storing de-icing salts, especially during dry weather periods, but that the accumulated amounts of salts are not very significant compared to the introduced amounts.
