Aspectos estruturais e ultraestruturais das espermátides de percevejos fitófagos (Heteroptera : Pentatomidae) /

Orientador: Mary Massumi Itoyama / Resumo: Dentro da subordem Heteroptera algumas espécies pertencentes a família Pentatomidae apresentam espermátides polimórficas em seus lobos testiculares que são oriundas de dois fenômenos: a polimegalia e o lobo harlequin. Para a investigação dessas espermátides polimórficas e de seu desenvolvimento comparadas às espermátides não polimórficas, foram coletadas em São José do Rio Preto as espécies Antiteuchus tripterus, Edessa meditabunda, Loxa deducta e Proxys albopunctulatus (Pentatomidae, Heteroptera). Os machos tiveram seus testículos extraídos, fixados e incluídos em parafina ou resina. O material incluído em parafina foi cortado e corado com hematoxilina/eosina (HE) e analisados em microscopia de luz, outros testículos, destas quatro espécies, foram incluídos em resina e analisados em microscópio eletrônico de transmissão (MET). Os testículos A. tripterus são alongados e divididos em 6 lobos, sendo 1 a 3 sem alterações, 4 e 6 com polimegalia e o 5 o lobo harlequin. Foi possível observar, também, que os lobos 4 e 6 possuem espermátides grandes enquanto o 5 apresenta espermátides pequenas. Em MET foi possível observar em todos os lobos a formação da cauda da espermátide onde ocorre a formação dos derivados mitocondriais e a posterior formação do axonema entre os derivados. Além disso, foi detectada a presença de três tipos de espermatozoides no ducto eferente, sendo estes oriundos dos diferentes lobos. Há o espermatozoide maior com morfologia alongada com pequenas protuberâncias... (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: Within the suborder Heteroptera, some species belonging to the Pentatomidae family show alterations in some of their testicular lobes, where we can show the polymegaly and the harlequin lobe. For the investigation of these polymorphic cells and their development compared to non-polymorphic cells, the species Antiteuchus tripterus, Edessa meditabunda, Loxa deducta and Proxys albopunctulatus (Pentatomidae, Heteroptera) were collected in São José do Rio Preto. The males had their testicles extracted, fixed and included in paraffin or resin. The material included in paraffin was cut and stained with hematoxylin / eosin (HE) and analyzed under light microscopy, other testicles, of these four species, were included in resin and analyzed using a transmission electron microscope (TEM). The testicles A. tripterus are elongated and divided into 6 lobes, 1 to 3 without changes, 4 and 6 with polymegaly and the 5 the harlequin lobe. It was also possible to observe that lobes 4 and 6 have large cells while 5 has small cells. In TEM, it was possible to observe the formation of the sperm tail in all lobes where the formation of the mitochondrial complex occurs, which divides into two mitochondrial derivatives and the subsequent formation of the axoneme between the derivatives. Arlequín y Polichinela, Valle Inclán y García Lorca : estudio comparativo, histórico y literario sobre la commedia dell’arte y los títeres

The popularity of commercial theatre such as the "melodrama" and the "género chico" presented a barrier to new and experimental theatre at the turn of the twentieth century in Spain. Foremost among the dramatists who attempted to overcome such obstacle were Ramón del Valle Inclán and Federico García Lorca whose early productions were based on commedia dell' arte, puppetry, and generally in popular traditional theatre. The first part of this study (written in Spanish) explores the origins and development of Harlequin in the Italian commedia and traces his presence in the Spanish Renaissance using the work of Cervantes as an example. The historical background of Harlequin is indispensable in understanding Valle Inclán's attitudes towards the commedia. Harlequin is the central figure in his play La Marquesa Rosalinda first performed in 1912. Valle Inclán uses names found in Cervantes's works and through metaphores, sets out to explain the canons of commedia dell' arte that was the heart and pulse of popular theatre in the Renaissance. This thesis examines the height and decline of the commedia in the eighteenth century and includes a study of La Marquesa Rosalinda in terms of its indebitness to the commedia on one hand and Valle Inclán's original development on the other. The second half of this study deals with two works of García Lorca that involve his character Don Cristóbal Polichinela. Don Cristóbal, based on the popular Spanish puppet of the same name, has been inspired by the masked Punchinello of the seventeenth century commedia. A historical and literary appraisal is used to trace the most important characteristics of Punchinello including his transformation from a man into a puppet. Particular emphasis is placed upon the presence of this puppet in Spain, its connection to Harlequin, and the utilization of Punchinello by García Lorca. To conclude this thesis, a comparative study based on the two zanies, Harlequin and Punchinello, enlightens the reasons and motives for the use of puppets and puppets-like characters in the works of Valle Inclán and García Lorca. / Arts, Faculty of / French, Hispanic, and Italian Studies, Department of / Graduate

Irony, Ideology, and Resistance: The Amazing Double Life of Harlequin Presents

In Harlequin Presents, the recurrence of particular moments of resistance suggests that these structures of events have meaning apart from -and perhaps even antithetical to - the ideological outcome of any specific text. The ideological structures presented in the romance novel are not passively accepted, nor do they simply fulfill a single pre-existing need or desire in their reading public. Romance novels utilize ideologies in a self-consciously playful and ironic way. These texts offer multiple ways of understanding the worlds they depict, the structures of understanding contained within being posited and discarded. This thesis proposes a means of interpreting the romance novel captures the ambivalence of the reading experience. I will show how the paradox of the romance novel - the seemingly limitless potential of a feminine discourse of the private sphere that is set within the conservative confines of repetitive narratives of social integration - is incorporated into the structure of the texts themselves. While the texts in the series manifest standardized outcomes, they also exhibit recurrent patterns of resistance. The serial form of Harlequin Presents dictates that this tension between separate value sets and ideologies is never fully resolved. The appeal of the romance novel must then lie between these competing demands. Each of my chapters examines the ways that this ironic tension functions within a different intellectual space. In considering how the domestic sphere, the body and the nation are overlayed with multiple, contested meanings, this thesis maps out the scope of ideological resistance and adherence throughout Harlequin Presents. / Thesis / Doctor of Philosophy (PhD)

Breeding habitat of harlequin ducks in Prince William Sound, Alaska

Breeding habitat of Harlequin ducks (Histrionicus histrionicus) was studied in eastern Prince William Sound, Alaska, during 1991 - 1993. Streams in Prince William Sound were surveyed for Harlequin ducks and monitored with mist nets. Physical characteristics of 24 Harlequin breeding streams were compared to those of 24 streams not used for breeding using 2 sample, principal components and logistic regression analyses. Nests were located using radio-telemetry of marked females. Harlequin ducks resident in eastern Prince William Sound selected the largest anadromous salmon streams available for nesting. Volume discharge of breeding streams averaged 3.2 m��/s and was the most important factor in habitat variation between streams used and not used by breeding Harlequins. Expansive estuaries and intertidal deltas present at the outflow of large streams were important foraging and loafing areas of Harlequin ducks. Although nesting females generally avoided smaller salmon streams their intertidal estuaries were often used for foraging by females and molting males. The largest streams in Prince William Sound, glacially fed rivers, were not used by breeding Harlequins. Ten nest sites of Harlequin ducks in eastern Prince William Sound were located on southwest facing, steeply sloping banks of small, first order tributaries near timberline elevation. Nests were associated with woody debris and shrubs, in shallow depressions or cavities, and were beneath the canopy of old growth forest. Microhabitat produced by a southwest aspect, snow shadow provided by the forest canopy, and sloping stream bank may provide nesting sites earlier in the spring compared to surrounding areas. / Graduation date: 1994

Organotypic human skin disease models for the assessment of novel therapeutic approaches

Comprehensive in vitro modelling of inflammatory human skin conditions is an essential first step in the development and assessment of potential therapeutic approaches. Mouse models or monolayer keratinocyte cultures come with distinct limitations which might be complimented or overcome by the use of human-specific organotypic 3D culture models. Over the course of this thesis, an organotypic culture system, based on patientderived immortalised keratinocyte cell lines on a dermal equivalent collagen 1 gel, was established and used to recapitulate phenotypical features for two hereditary skin diseases, Harlequin ichthyosis and Tylosis with oesophageal cancer. Small molecular compounds, supplied via the medium, or RNA interference were used to modulate disease-specific changes in histology and marker expression of the skin equivalent. Since hyperproliferative skin conditions can be associated with an aberrant wound healing phenotype, the organotypic system was manipulated to obtain a basic in vitro wound healing model. This model displays typical features of re-epithelialisation over time (both normal and disease-specific) which can further be manipulated via shRNAmediated knockdown or the exogenous supply of compounds. In parallel, a non-disease model was used to assess the topical application of novel nanopolymeric drug delivery systems in regard to their ability to penetrate across the permeability barrier. Penetrance profiles for the organotypic model (in dependence of co-application with chemical enhancers) showed a similar pattern as for topical applications performed in parallel on explant skin. In conclusion, a highly adaptable human organotypic keratinocyte culture model was developed and used to recapitulate (and manipulate) skin disease phenotypes and epidermal wound healing in vitro, as well as perform first essential assessments of novel drug delivery systems.
