Flavonoid compounds are substances widely extended in nature and many of them have a positive influence on human health, primarily for their vasodilatory, antioxidant and anti- inflammatory effects. Three substances were selected for this diploma theses: two substances of isoflavonoid group, genistin and genistein, and the end product of genistein metabolism, 4-ethylphenol. The aim of this work is examination of vasorelaxant effects of this substances in vitro. Vasorelaxing potential of tested substances was tested in vitro in isolated aortic rings of Wistar rat. The effect of increasing doses of individual substances in precontracted aortic rings with intact endothelium was measured. From the obtained values of vessel tension, the DRC curves and EC50 values were created. The results were evaluated. Evaluating the effects of essential oils and condensed tannin on fermentation and methane production under in vitro conditions

The effects of essential oils (EO) and condensed tannin (CT) on methane production and fermentation were evaluated under in vitro conditions. Experiment one screened the effects of ten different EO at 500 mg/L of culture fluid on methane production in a 24 hr rumen batch culture. Based on the results from experiment one, four EO (clove oil (CLO), cinnamon oil (CNO), white thyme oil (WTO), and anise oil (ANO)) were selected and tested at three different dose levels (125, 250, and 500 mg/L of culture fluid) for their effects on methane production and fermentation in a 24 hr batch culture (experiment two). Methane production was significantly reduced with the four EO at the 500 mg/L dose but at 250 mg/L; only CLO and CNO reduced methane production. The reduction in methane production with the 500 mg/L dose was also associated with reductions in dry matter (DM) digestibility and total volatile fatty acids (VFA). In the third experiment, the effect of CT at 2.5, 5 and 7.5% of diet DM on methane production was also tested in a 24 hr batch culture. Results from this experiment showed no effect of CT on methane production although methane production tended to be lower at 7.5% CT. In conclusion, the reduction in methane production with EO most likely resulted from EO negative effects on DM digestibility and fermentation.

The air quality in sterile environment is an important aspect to cell culture. It is known the culture conditions are essential for in vitro human embryo development; however there is no standardization for incubator filters in in vitro fertilization (IVF) laboratories. Recently, the incubator air quality has been receiving attention, and effect of volatile organic compounds (VOC) has being highlighted. The aim of this study was to evaluate the laboratorial outcomes of IVF cycles with embryo culture using CO2 incubators with the HEPA filter for air particles and a high efficiency VOC filter with activated coal. Material and methods. This is an observational prospective controlled study which included 160 IVF cycles at Human Reproduction Clinic CRIAR between 2008 January and 2009 September. The cycles were matched to patient age, infertility etiology, pituitary desensitization protocol, and semen origin and quality. Thirty five cycles were compared in which the embryos were cultured in CO2 incubator with HEPA filter (HEPA group) and 35 cycles where a CO2 incubator with VOC filter was used for embryos cultures (VOC group). Results. The groups were similar for general characteristics of IVF cycles. We did not observed differences about normal fertilization and embryo cleavage rates. On the other side, the proportion of high quality embryos (grade A) was higher on VOC group (50.8%) than HEPA group (35.7%; p=0.05). Conclusions. IVF cycles using CO2 incubator with VOC filter for embryo culture results in a increased high quality embryos rate, those with high implantation potential. / Introdução e objetivos. A qualidade do ar em um ambiente estéril é um aspecto de grande importância para a cultura celular. Apesar de a condição da cultura ser um fator preponderante para o desenvolvimento de embriões in vitro, não há uma padronização dos filtros utilizados nas incubadoras dos laboratórios de fertilização in vitro (FIV). Nos últimos anos, a qualidade do ar nas incubadoras de cultura embrionária vem recebendo maior atenção, e o papel dos compostos orgânicos voláteis (VOC) é um item em destaque. O objetivo deste estudo foi avaliar os resultados laboratoriais de ciclos de FIV, utilizando incubadoras de CO2 para cultura de embriões com o filtro HEPA para partículas de ar e filtro VOC de alta eficiência com carvão ativado. Materiais e métodos. Este trabalho consiste de um estudo observacional prospectivo, incluindo 160 ciclos de FIV realizados na Clínica de Reprodução Humana CRIAR, de janeiro de 2008 a setembro de 2009. Os ciclos foram pareados de acordo com a idade da paciente, fator de infertilidade, protocolo de bloqueio hipofisário e origem e qualidade do sêmen utilizado. Trinta e cinco (35) ciclos nos quais os embriões foram cultivados em incubadora de CO2 equipada com filtro HEPA (grupo HEPA) e 35 ciclos cujos embriões foram cultivados em incubadora de CO2 equipada com filtro VOC (grupo VOC) foram comparados. Resultados. Os grupos foram semelhantes em relação aos aspectos gerais dos ciclos de FIV. Não foram observadas diferenças entre os grupos quando avaliadas as taxas de fertilização normal e clivagem embrionária. À proporção de embriões de alta qualidade (grau A), entretanto, foi maior no grupo VOC (50,8%) comparado ao grupo HEPA (35,7%; p=0,05). Conclusões. Ciclos de FIV utilizando incubadoras de CO2 equipadas com sistema de filtragem de ar do tipo VOC para cultivo embrionário resultam, em maior taxa de embriões grau A, ou seja, aqueles com maior potencial de implantação.
