The Effect of Lipid/Hydrocolloid Coatings on the Postharvest Storage Quality of 'Golden Delicious' Apples

The performance of three different lipid/hydrocolloid coatings was tested on Golden Delicious apples. The coatings consisted of a mixture of lipid, wax, and various gum blends. The three treatments were compared to an uncoated group (control). Both objective and sensory tests were performed to determine the shelf-life stability of the apples. Objective results indicated that all three treatments significantly decreased (P<0.0083) the respiration rates of the apples when compared to the control. The treatment groups also significantly (P<0.0083) maintained the texture of the apples along with a lower (P<0.0083) starch degradation rate. The treatment groups also significantly (P<0.0083) maintained the malic acid level in the apple. There were no significant differences (P>0.0083) among the groups with regard to soluble solids and fresh weight loss. Externally, the treated apples had a significantly greener hue (Hunter color "a") than the control apples, however, there were no significant differences (P>0.0083) in yellow color (Hunter color "b") between the groups. Sensory results indicated that the treated apples were firmer, crisper and juicier (P<0.0083) than the control apples. However there were no significant differences (P>0.0083) in the sweetness, tartness, and appearance of all of the groups. Overall, the derived coatings could serve as a resource for extending and maintaining the shelf life of perishable fruit. / Master of Science

Development of Mass Spectrometric Methods for the Analysis of Components and Complex Interactions in Biological Systems

The anti-cancer agent 4, 4-dihydroxybenzophenone-2, 4- dinitrophenylhydrazone (A007) forms complexes with pdelocalized lymphangitic dyes that allow its penetration through the skin effectively delivering it to a meta-stable type cancerous site. Previous in vitro studies, combined with gas phase mass spectrometry studies, have shown that a stronger binding affinity equates to a greater efficacy of the drug. For the determination of drug:dye complex binding strength coefficients in solution, two methods have been developed by affinity capillary electrophoresis (ACE), and cation exchange liquid chromatography (CELC). The methods demonstrated that A007 non-covalent binding strength was greatest for methylene green, followed by methylene blue, and lastly toluidine blue. Bond dissociation energies and apparent reaction enthalpies for the fragmentation pathways of lithiated acylglycerols were experimentally determined by collision activation in a triple quadrupole mass spectrometer. A developed novel derived effective path length approach for predicting bond dissociation energies (BDE) for electrostatic complex's alkali metal adducts (Li+), and halide adducts (Cl-) of acylglycerols was applied to the major fragmentation product ions of a lithiated mono-acylglycerol, a 1, 2-diacylglycerol, and a 1, 3-diacylglycerol, to predict the covalent bond dissociation energies involved in fragmentation pathways. The model's calculated apparent reaction enthalpies are used in conjunction with the energy-resolved mass spectrometry method of breakdown graphs to give a more complete quantitative aspect to the interpretation of the fragmentation processes. The dry eye condition affects millions of individuals world wide. The symptoms can be a result of simple irritation to the eye or a serious disease state. A dry eye model was developed using rabbits in order to study the compositional makeup of the tear components in hopes of identifying an underlying cause, or expressed effect of the dry eye condition. The major non-polar lipids of the tear were identified by mass spectrometry as mono and diacylglycerols, with a smaller extent of triacylglycerols. The major polar phosphorylated lipids were identified in the tear extract revealing that sphingomyelin based species were being expressed in the dry eye condition. The major proteins were determined to be the lower molecular weight lipid binding proteins where two specific species were found to increase in expression for the dry eye condition.

Dietary polyunsaturated fatty acids modify plasma lipids and red blood cell membrane composition but do not induce &#8710;6 desaturase mediated conversions in the domestic feline

This study explored the effects of dietary unsaturated fatty acids on feline lipid metabolism. It was hypothesized that high dietary linoleic acid (18:2n-6, LA) would enhance conversion to arachidonic acid (20:4n-6), enrichment of dietary long chain n-3 FA (LCn-3FA) would affect lipid parameters, and n-3 FAs incorporation may blunt n-6 FA incorporation. Twenty-nine cats were randomized into groups (n = 9, 10, 10), and fed for 28 days with blood collections on days 0, 14, and 28. Experimental diets consisted of a commercial diet, supplemented with 8g oil/100g kibble. Oil supplements and subsequent diets were: high-oleic sunflower (H diet) with 82% oleic acid (18:1n-9), Menhaden fish (M diet) with LCn-3FA, and safflower (S diet) with 75% 18:2n-6. Dietary 20:4n-6 content was: 0.03 for H and S, and 0.09 for M (g FA/kg diet). Nonesterified fatty acid (NEFA), triacylglycerol (TG), total cholesterol (TC), lipoproteincholesterol (LP-C), plasma phospholipid (PL) FAs, red blood cell membrane (RBC) FAs, and ∆5 and ∆6 desaturase indices were measured. Statistical analyses were performed with SAS PROC MIXED with p < 0.05 determining significance. Neither TC nor NEFA showed significant effects. Diet M resulted in significant TG lowering, despite typically low feline TGs. Similarly, pre-β LP-C (i.e. TG-rich VLDL) was decreased in diet M. Plasma PL FAs revealed significant accumulations of the following: 18:1n-9 in diet H, 18:2n-6 in diet S, and LCn-3FA in diet M. Despite high dietary 18:2n-6, plasma PL 20:4n-6 was not increased in diet S over diets H or M. Increased docosadienoic acid (20:2n-6) in diet S demonstrated that 18:2n-6 chain elongation occurred in deference to its ∆6 desaturation further substantiating low feline ∆6 desaturase activity. Interestingly, no diet M blunting of 20:4n-6 incorporation occurred because fish oil supplementation provided additional 20:4n-6. Tissue 20:4n-6 content appears to be diet-dependent. Accumulation of eicosapentaenoic acid (20:5n-3), but low affinity for docosahexaenoic acid (22:6n-3) occurred in diet M RBC membranes. After 28 days, plasma PLs reflect dietary intake more readily than RBC membranes. Fish oil supplementation resulted in plasma PL LCn-3FA enrichment and lowered plasma TG concentrations, both of which may have physiological significance in cats.

Dyslipidaemic pancreatitis : clinical assessment and analysis of disease severity and outcomes.

Introduction: The relationship between pancreatitis and dyslipidaemia is unclear and has never been studied in a South African context. Patients and methods: A prospective evaluation of all admissions with acute pancreatitis to a regional hospital general surgical service was performed to ascertain its relationship to dyslipidaemia. Aetiology was determined by history and ultrasound assessment. Disease severity was assessed using a modified Imrie score and an organ failure score. Body mass index was calculated. A lipid profile was obtained. Abnormal profiles were repeated. Secondary causes of dyslipidaemia were noted. A comparison of the demographic profile, aetiology, disease severity scores, complications and deaths were made in relationship to the lipid profiles. Results: From June 2001 to May 2005, there were 230 admissions, of whom 31% were women and 69% men. The median age was 38 years(range 13- 73). The pancreatitis was associated with alcohol in 146(63%), gallstones in 42(19%) and idiopathic in 27(12%). The amylase was significantly higher with a gallstone aetiology (p / Thesis (MMedSc)-University of KwaZulu-Natal, 2006.
