Histological and ultrastructural study of mice with hereditary muscular dystrophy

The history of human muscular dystrophy with reference to clinical, histological and biochemical studies is reviewed. The value of the recent discovery of an experimental mouse with hereditary muscular dystrophy with clinical, histological, and biochemical similarities to human muscular dystrophy is discussed. The histology of normal muscle, with special reference to ultrastructure, is also reviewed. For this investigation mouse muscle samples for the electron microscope were fixed in Palades osmic acid solution, embedded in methacrylate or epon and stained with either lead hydroxide or phosphotungstic acid. A method for obtaining one day old muscle samples, while keeping the animals alive until the dystrophic symptoms are noted clinically, is outlined. Light microscope sections were obtained from electron microscope blocks and examined after staining with toluidine blue. Experimental results by light and electron microscope observations showed there to be no histological differences between one day old dystrophic and normal muscle. However one day old muscle showed histological differences compared to older muscle. The chief differences were the smaller size of fibers, random distribution of mitochondria and enlarged nuclei, and the presence of abundant interfibrillar sarcoplasm with a conspicuous granular components. The regular repeating pattern of the endoplasmic reticulum of adult muscle fibers was hot seen. Some "atypical" fibers showed similarities to altered fibers seen in older dystrophic mice. The mitochondria were swollen and vacuolated with few cristae and pale matrix. Endoplasmic reticular components were vacuolated and adjacent myofibrils were disorganized. Atrophy of fibers was first noted at 14 days of age and were conspicuous at 63 days. By 63 days alterations were also noted in mitochondria and endoplasmic reticulum and these changes became more prominent with the progress of the disease. Atrophy of the myofibrils was evident and the Z-bands were often irregular and out of register. Connective tissue also increased greatly. Several miscellaneous structures are also discussed. The histological findings are compared to those found by other workers. Several suggestions put forward by others as to the possible cause of the disease are summarized. On the basis of morphological findings it is suggested that ribonucleo-protein synthesis in the nuclei of dystrophic muscle fibers is increased but that there may be some intermediate stage at fault which prevents the conversion of amino acids to myofilaments. / Medicine, Faculty of / Graduate

Foram coletadas amostras indeformadas e deformadas através da abertura de um poço de inspeção no Campo Experimental para estudos de Mecânica dos Solos e Fundações da UNICAMP (Campus Campinas), localizado na FEC - Faculdade de Engenharia Civil, Arquitetura e Urbanismo. Procurou-se atingir tais objetivos através da realização de ensaios laboratoriais (caracterização, resistência, deformabilidade, permeabilidade e sucção) e de correlações entre os parâmetros obtidos através dos ensaios, visando classificar o subsolo, dando enfoque a sua condição laterítica e não saturada. Ensaios edométricos simples foram realizados com o solo na condição natural e com inundação nas tensões de 100kPa, 200kPa e 400kPa para analise da colapsibilidade. Dos resultados obtidos, verificou-se que o solo se apresenta colapsível para todas as tensões de inundação. A partir dos resultados experimentais, fez-se a correlação de alguns parâmetros utilizando-se o programa de computador Wolfram Mathematica 7.0, verificando-se qual função melhor se ajustava para a correlação desejada / Abstract: This research aimed to study and evaluate some geotechnical properties of the northern region of Campinas soil profile, which have great importance for practice and development of geotechnical engineering in the Sao Paulo State. Disturbed and undisturbed samples from the subsoil were collected through the opening of inspection shafts at the experimental site for studies of Soil Mechanics and Foundations, UNICAMP (Campus Campinas), located on the FEC - Faculty of Civil Engineering, Architecture and Urbanism. In order to achieve these goals, laboratory tests have been performed, such as: characterization, resistance, compressibility, permeability and suction as well as correlation between parameters obtained from the tests to classify subsoil, giving emphasis to its unsaturated and lateritic condition. Oedometer tests were performed with the soil in natural and flooded condition with 100 kPa, 400 kPa and 200 kPa for analysis of collapsibility. From the results, it was verified that the soil is collapsible for all flooding stresses. High values of collapse potential were found and the results allowed to do correlations of some parameters using the computer program Wolfram Mathematica 7.0, verifying which function was the most appropriated to the desired correlation / Mestrado / Geotecnia / Mestre em Engenharia Civil

A Virtualized Scada Laboratory for Research and Teaching

This thesis describes a virtual Supervisory Control and Data Acquisition (SCADA) laboratory. This virtual laboratory was built using virtual devices that simulate industrial processes, emulate control system ladder logic functionality, and utilize control system communication protocols. Human machine interfaces (HMIs) were developed for distribution and testing, and commercially available HMIs were implemented. In addition to virtual devices and HMIs, attacks were implemented against this virtual system. Uses for this laboratory include both SCADA security research and pedagogy. The laboratory serves research purposes, as it utilizes industry standard SCADA communication protocols as well as commercial HMIs, and is capable of interfacing with physical SCADA equipment, and is also capable of producing volumes of industrial control system traffic. It also serves pedagogical purposes as several laboratory exercises were developed in conjunction with the simulators to demonstrate the workings of cyber-physical security in a classroom environment.
