Natural products from tropical and temperate marine algae

Following surveys of seaweeds in Oregon and the Caribbean, four algal species were selected for study on the basis of their crude extract biological activity or thin-layer chromatographic characteristics. Extracts were fractionated by normal phase chromatography, including high performance liquid chromatography (HPLC). Derivatives (methyl esters, acetates, and benzoates) of natural products were produced to facilitate isolation or structure elucidation efforts. Structure proofs utilized high-field nuclear magnetic resonance (NMR) spectroscopy, including hetero- and homo-nuclear two-dimensional experiments. Infrared and ultraviolet spectra, optical rotations, mass spectra, and circular dichroic spectroscopy were also employed. The Puerto Rican red alga Murrayella periclados was found to contain (12S)-hydroxyeicosapentaenoic and (12S)-hydroxyeicosatetraenoic acids, two mammalian autacoids for which new biological activities are reported. Further eicosanoid metabolism was demonstrated by the isolation of (6E)-leukotriene B₄ and two diastereomers each of the insulin-release mediators hepoxilin B₃ and B₄. The Oregon green alga Acrosiphonia coalita also was found to contain novel fatty acid-derived substances. Some of these contain an unprecedented structural feature, a conjugated trienal in which the aldehyde moiety branches off from the fatty acid chain. Exciton chirality studies of benzoate derivatives indicate that the alga introduces oxygenation in the 9R, 13S, and 16S positions on precursor polyunsaturated fatty acid chains. The Oregon red alga Laurencia spectabilis was found to produce an equilibrium mixture of (±)-2-hydroxy-2-methyldihydrofuran-3-one and 5-hydroxy- 2,3-pentanedione, which dimerizes on silica gel to produce spiro-bis-pinnaketal, a compound which was previously reported from L. pinnatifida and is probably an artifact of isolation. The Oregon red alga Prionitis lanceolata displays high levels of apparent tryptophan metabolism. Never before isolated from a plant or marine source, 3- (hydroxyacetyl)indole, active at 10⁻¹⁰ M in the lettuce-seedling root elongation assay, was isolated in 0.07% yield along with indole-3-carboxaldehyde (0.5%) and indole-3-carboxylate (0.04% ). / Graduation date: 1992

Light intensity influences on algal pigments, proteins and carbohydrates: implications for pigment-based chemotaxonomy

Phytoplankton Chlorophyll a (CHLa), total protein, colloidal carbohydrates, storage carbohydrates and taxonomic pigment relationships were studied in two cyanophytes (Microcystis aeruginosa and Synnechococcus elongatus), two chlorophytes (Dunaliella tertiolecta and Scenedesmus quadricauda), one cryptophyte (Rhodomonas salina), two diatoms (Cyclotella meneghiniana and Thalassiosira weissflogii) and one dinophyte (Amphidinium carterae) to assess if algal biomass could be expressed in other indices than just chlorophyll a alone. Protein and carbohydrates are more useful currencies for expressing algal biomass, with respect to energy flow amongst trophic levels. These phytoplankton were grown at low light (LL = 37 (So(Bmol photons m-2 s-1), medium light (ML = 70-75 (So(Bmol photons m-2 s-1), and high light (HL= 200 (So(Bmol photons m-2 s-1). / Even though pigment per cell increased with increasing light intensity, statistically light had very little effect on the CHL a : taxonomic marker pigment ratios, as they covaried in the same way. Protein, colloidal carbohydrates and storage carbohydrates per cell all increased with increasing light intensity, but they did not covary with CHLa. Statistical data showed that light intensity had a more noticeable effect on protein: CHL a, colloidal carbohydrate: CHLa, storage CHO: CHLa, therefore a general mathematical expression for these relationships cannot be generated. This study showed that light intensity does have an influence on these biomass indices, therefore, seasonal and latitudinal formulas may be required for meaningful algal biomass estimation. However, more studies are needed if that goal is to be realized. / While studying the effects of light intensity on algal pigment content and concentration, a new pigment was isolated from a cyanophyte (Scytonema hofmanii) growing between 300-1800 (So(Bmol photons¨m-2¨s-1 and from samples collected in areas of the Florida Everglades. This pigment was characterized and structurally determined to possess indolic and phenolic subunits that are characteristic of scytonemin and its derivatives. In addition, the pigment has a ketamine functionality which gives it its unique polarity and spectral properties. Spatial and temporal distributions of heavy metals in Hong Kong seaweeds with an analysis on the effects of heavy metals on the reproduction of the green alga ulva lactuca. / CUHK electronic theses & dissertations collection

No periodic patterns of temporal variations in the metal levels in U. lactuca or in other seven common seaweed species from Ping Chau were observed from 1999 to 2000. Cu levels were generally negatively correlated with other metals in seaweeds. / Spore production of U. lactuca was significantly reduced by the elevation of copper and nickel levels in the seaweed samples. The reproductive frequency of U. lactuca generally increased from January and February to the maxima in March and April. Copper, nickel and nitrate levels showed significant negative correlations with these reproductive frequencies. / The metal abundance in 24 seaweeds showed the following trend: Fe > Mn, Zn > Cu, Ni, Pb, Cr > Cd. U. lactuca and Padina australis showed relatively high mean and large range values of metal levels. Principal component analysis summarized the overall metal loadings in these 24 seaweed species. The variations in Pb, Fe, Mn and Cr levels in the seaweeds varied greatly. / There were significant spatial variations of different metal levels in the extensive study of U. lactuca from various intertidal waters in Hong Kong from 1999 to 2001. In general, metal levels in U. lactuca increased from January to March or April and then dropped in the following months. No periodic patterns or temporal trends of variations of metal levels in U. lactuca were found. Different metal levels in U. lactuca were comparatively lower than those in other studies in other countries and in past studies in Hong Kong. / There were significantly differences in various metal levels in different structures of Sargassum hemiphyllum, generally decreased in the following order: receptacles > vesicles > leaves > branches. / This thesis research involves biomonitoring levels of eight metal species (Cd, Cr, Cu, Fe, Mn, Ni, Pb and Zn) in seaweed and the effects of these metals on the reproduction of Ulva lactuca. The study started from September 1999 and ended in June 2001, covering 40 intertidal sites in Hong Kong and 24 seaweed species.
