Charles University Faculty of Pharmacy in Hradec Králové Department of Biological and Medical Sciences Student: Zuzana Drobná Supervisor: PharmDr. Miroslav Kovařík, Ph.D. Title: Dynamometry as a prognostic and prediction marker Dynamometry is a method for measuring muscle strength. It is one of the methods of biomechanical motion analysis. There are two types of dynamometry. Isokinetic dynamometry is based on isokinetic muscle contraction. In this contraction, the muscles change their length. Isometric dynamometry is based on isometric muscle contraction. The length of the muscle does not change during the contraction. In my master thesis, 22 studies were included. Of this, 18 were engaged in a dynamometer, which measured muscle strength. Studies have been focused on prediction of short and long-term mortality, prognosis of hospitalized and severely ill patients, prediction of disease symptoms (cancer, chronic obstructive pulmonary disease, liver disease). I searched for studies according to the PubMed database. In 16 studies, it was confirmed that dynamometry can be used in the present time as a reliable diagnostic, predictive and prognostic method. Human carbonic anhydrases are metalloenzymes that are involved in many physiological processes in the body, but also play an important role in the pathogenesis of numerous diseases. Under regular conditions, expression of carbonic anhydrase IX (CAIX) is very limited, unlike that of other 14 human carbonic anhydrase isozymes. But in hypoxic tumors this enzyme is highly overexpressed on the cell surface. For this reason, this enzyme represents a good target for therapy and diagnosis of tumors and thus various anti-CAIX monoclonal antibodies and specific inhibitors are being developed. In this work we investigated the possibility to use fluorescent polymer conjugate carrying a CAIX specific inhibitor for florescent labeling of tumor cells. Specific binding of polymer conjugate to different cell lines was investigated by flow cytometry and confocal microscopy. Binding properties of the polymer conjugate was compared to CAIX specific monoclonal antibody M75 and its single-chain fragment scFv M75. Ability of the polymer conjugate to inhibit CAIX enzyme activity was also investigated. For these experiments, recombinant protein CAII was prepared and purified, which was also used for protein crystallization. This thesis presents a comprehensive study of the Japanese double-ga construction and offers an explanation as to why the Japanese double-ga construction does not allow scrambling. In chapter 2, the particle-ga and the particle-wa are defined as the focus marker and the topic marker respectively. The different shades of meaning that both particles have are also explained. Chapter 3 illustrates the Japanese double -ga construction. Chapter 4 deals with the impossibility of scrambling in the double-particle constructions. A strong parallelism is shown between the double-ga construction and the double-wa construction. The claim is that there are three "pragmatic slots" that the particle-ga and -wa can occupy in the sentence. The rigid-fixed-order of these three slots contributes to the prohibition of scrambling.
