For the purpose of numerically simulating continuum mechanical structures, different types of material may be represented by the extreme values {<img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Cepsilon" />,1}, where 0<<img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Cepsilon" /><img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Cll" />1, of a varying coefficient <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Calpha" /> in the governing equations. The paramter <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Cepsilon" /> is not allowed to vanish in order for the equations to be solvable, which means that the exact conditions are approximated. For example, for linear elasticity problems, presence of material is represented by the value <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Calpha" /> = 1, while <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Calpha" /> = <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Cepsilon" /> provides an approximation of void, meaning that material-free regions are approximated with a weak material. For acoustics applications, the value <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Calpha" /> = 1 corresponds to air and <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Calpha" /> = <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Cepsilon" /> to an approximation of sound-hard material using a dense fluid. Here we analyze the convergence properties of such material approximations as <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Cepsilon" />!0, and we employ this type of approximations to perform design optimization. In Paper I, we carry out boundary shape optimization of an acoustic horn. We suggest a shape parameterization based on a local, discrete curvature combined with a fixed mesh that does not conform to the generated shapes. The values of the coefficient <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Calpha" />, which enters in the governing equation, are obtained by projecting the generated shapes onto the underlying computational mesh. The optimized horns are smooth and exhibit good transmission properties. Due to the choice of parameterization, the smoothness of the designs is achieved without imposing severe restrictions on the design variables. In Paper II, we analyze the convergence properties of a linear elasticity problem in which void is approximated by a weak material. We show that the error introduced by the weak material approximation, after a finite element discretization, is bounded by terms that scale as <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Cepsilon" /> and <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Cepsilon" />1/2hs, where h is the mesh size and s depends on the order of the finite element basis functions. In addition, we show that the condition number of the system matrix scales inversely proportional to <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Cepsilon" />, and we also construct a left preconditioner that yields a system matrix with a condition number independent of <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Cepsilon" />. In Paper III, we observe that the standard sound-hard material approximation with <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Calpha" /> = <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Cepsilon" /> gives rise to ill-conditioned system matrices at certain wavenumbers due to resonances within the approximated sound-hard material. To cure this defect, we propose a stabilization scheme that makes the condition number of the system matrix independent of the wavenumber. In addition, we demonstrate that the stabilized formulation performs well in the context of design optimization of an acoustic waveguide transmission device. In Paper IV, we analyze the convergence properties of a wave propagation problem in which sound-hard material is approximated by a dense fluid. To avoid the occurrence of internal resonances, we generalize the stabilization scheme presented in Paper III. We show that the error between the solution obtained using the stabilized soundhard material approximation and the solution to the problem with exactly modeled sound-hard material is bounded proportionally to <img src="http://www.diva-portal.org/cgi-bin/mimetex.cgi?%5Cepsilon" />.

Die vorliegende Arbeit hat die Entwicklung und Validierung eines Analysealgorithmus zur objektiven Auswertung der Materialverteilung entlang der Zarge an gezogenen Karton-formziehteilen zum Ziel. Die dafür verwendeten Messdaten werden durch Oberflächenab-tastung mit konfokaler Sensorik erzeugt. Unter Anwendung des Analysealgorithmus wer-den die Einflüsse der Prozessparameter auf die Materialverteilung ermittelt. Abschließend wird aus den gewonnenen Daten ein Ansatz zur Berechnung der Werkzeuggeometrie ent-wickelt.
Zur Bestimmung der Materialverteilung wird die Zarge beidseitig abgetastet. Aus der Diffe-renz der Radien von Außen- und Innenkontur wird der Dickenverlauf entlang der Zarge bestimmt, die die Materialverteilung beschreibt. Die Messung mit konfokalen Sensoren wird mit mikroskopischen Aufnahmen von Mikrotomschnitten und röntgentomographi-schen Aufnahmen geprüft.
Der Mittelwert und die Standardabweichung des Dickenverlaufs charakterisieren die Mate-rialverteilung. Der Analysealgorithmus ermittelt aus den durch Sensoren aufgenommenen Daten die charakteristischen Werte. In der Auswertung zeigt sich, dass eine Höhere Zieh-stempeltemperatur als Ziehbüchsentemperatur sowie eine hohe Temperatursumme die mittlere Dicke verringern. Eine hohe Faltenhalterkraft und eine hohe Stempeltemperatur bewirken eine gleichmäßigere Materialverteilung.
Der ermittelte Zusammenhang von der Beeinflussung des Mittelwertes der Dicke mit den Temperaturunterschieden von Stempel und Büchse wird durch ein Modell der thermischen Ausdehnung von Stempel und Büchse belegt. / The present thesis comprises the development and validation of an algorithm for the objec-tive evaluation of data on material distribution alongside the architrave of drawn cardboard shells. The necessary data are obtained through detection of the surface with confocal sensors. The influences of the process parameters on material distribution are investigated with algorithm. Resulting from this data, the basis for the calculation of the tools geometry is developed.
The determination of material distribution is accomplished by double-sided optical scanning of the architraves inner and outer surface. The trend of cardboard thickness alongside the architrave is calculated by the subtracion of the inside radius from the outside radius of the cardboard shell. The cardboard thickness describes material distribution. The measures with confocal sensors are approved of examined microtome cuts under microscope and x-ray analyses.
Material distribution is characterised by average of the thickness and standard deviation of thickness. The analysis algorithm calculates with sensor data the characteristic values. A higher temprature of the drawing die as the temprature of the drawing box and the sum of temperatures lead to a decrease of the average thickness. The combination of a high die temperature and a high force applied by the blankholder result in a more even material distribution.
The identified correlation of average thickness and temperature gradients between the drawing die and the box is discribed by thermal expansion of die and box.

Surface Modified Capillaries in Capillary Electrophoresis Coupled to Mass Spectrometry : Method Development and Exploration of the Potential of Capillary Electrophoresis as a Proteomic ToolZuberovic, Aida January 2009 (has links)
The increased knowledge about the complexity of the physiological processes increases the demand on the analytical techniques employed to explore them. A comprehensive analysis of the entire sample content is today the most common approach to investigate the molecular interplay behind a physiological deviation. For this purpose a method that offers a number of important properties, such as speed and simplicity, high resolution and sensitivity, minimal sample volume requirements, cost efficiency and robustness, possibility of automation, high-throughput and wide application range of analysis is requested. Capillary electrophoresis (CE) coupled to mass spectrometry (MS) has a great potential and fulfils many of these criteria. However, further developments and improvements of these techniques and their combination are required to meet the challenges of complex biological samples. Protein analysis using CE is a challenging task due to protein adsorption to the negatively charged fused-silica capillary wall. This is especially emphasised with increased basicity and size of proteins and peptides. In this thesis, the adsorption problem was addressed by using an in-house developed physically adsorbed polyamine coating, named PolyE-323. The coating procedure is fast and simple that generates a coating stable over a wide pH range, 2-11. By coupling PolyE-323 modified capillaries to MS, either using electrospray ionisation (ESI) or matrix-assisted laser desorption/ionisation (MALDI), successful analysis of peptides, proteins and complex samples, such as protein digests and crude human body fluids were obtained. The possibilities of using CE-MALDI-MS/MS as a proteomic tool, combined with a proper sample preparation, are further demonstrated by applying high-abundant protein depletion in combination with a peptide derivatisation step or isoelectric focusing (IEF). These approaches were applied in profiling of the proteomes of human cerebrospinal fluid (CSF) and human follicular fluid (hFF), respectively. Finally, a multiplexed quantitative proteomic analysis was performed on a set of ventricular cerebrospinal fluid (vCSF) samples from a patient with traumatic brain injury (TBI) to follow relative changes in protein patterns during the recovery process. The results presented in this thesis confirm the potential of CE, in combination with MS, as a valuable choice in the analysis of complex biological samples and clinical applications.

In this work, the material distribution, structure of fiber orientation and fiber attrition in
sandwich and push-pull injection molded short fiber composites are investigated, regarding the
effect of fiber content and processing parameters, given its direct relevance to mechanical
properties. The prediction of the tensile strength of conventional, sandwich and push-pull
injection molded short fiber composites are derived by an analytical method of modified rule of
mixtures as a function of the area fraction between skin and core layers. The effects of fiber
length and fiber orientation on the tensile strength are studied in detail. Modeling of the
specialized injection molding processes have been developed and performed with the simulation
program in order to predict the material distribution and the fiber orientation state. The secondorder
orientation tensor (a11) approach is used to describe and calculate the local fiber
orientation state. The accuracy of the model prediction is verified by comparing with
corresponding experimental measurements to gain a further basic understanding of the melt flow
induced fiber orientation during sandwich and push-pull injection molding processes.

The description of the main processes in the factory from a purchase, a production and a distribution of milk and milk products are a part of this thesis. Description of the milk collection to the dairies , its processing and storage up to expedition. The relationships of a delivery , the process of order, the distribution and supply are mentioned. I have pointed on a cohesion of all partnerships of Madeta Group, Inc. and I have described the organisational structure in the factory together the responsibilities of a leadership workers. The work is specialised on current situation in a factory as well- the milk prices, an export of material to Germany, a limitation of a production and a closing of a branches, growth of a prices of the butter and evaluation of the situation. The evaluation of a whole company, economic situation and valuation of logistic mentioned company are a conclusion of this thesis.
