Dentre os fitoplasmas causadores de doenças na cultura da mandioca (Manihot esculenta Crantz), se destaca o causador da doença do “Couro de Sapo da mandioca” (DCSM), o qual é responsável por grandes perdas na cultura em alguns países da América do Sul. Com o objetivo de detectar e caracterizar o fitoplasma causador da DCSM no Brasil, 30 plantas de mandioca apresentando sintomas típicos e 20 plantas de mandioca sem sintomas típicos de DCSM foram coletadas em Mocambinho (Jaíba), no estado de Minas Gerais, Brasil. O DNA total das raízes, caules e folhas foi extraído e utilizado como molde para a reação de PCR duplo. Foram desenhados iniciadores específicos para facilitar a detecção do fitoplasma na presença de Bacillus sp. (endofítico). Utilizando estes iniciadores foi possível detectar o fitoplasma em 16,7% das raízes, 66,7% dos caules e 73,3% das folhas em plantas sintomáticas, e em nenhuma das raízes, 45% dos caules e 30% das folhas em plantas assintomáticas. Um total de 37 plantas coletadas apresentou resultado positivo para a presença de fitoplasmas. O fitoplasma detectado foi identificado como pertencente ao grupo 16SrIII, subgrupo A, através de RFLP, sequenciamento e RFLP in silico. Análises filogenéticas, baseadas no 16S rDNA deram suporte à caracterização realizada através do RFLP. / Diseases caused by phytoplasmas in different crops have emerged in the past decades being responsible for significant economic losses worldwide. Of particular note among the disease-causing phytoplasmas in cassava (Manihot esculenta Crantz) is that responsible for cassava frog skin disease (CFSD), which causes great losses in some South American countries. In order to detect and characterize the phytoplasma that causes CFSD in Brazil, 30 cassava plants showing typical disease symptoms and 20 cassava plants CFSD symptomless were collected in Mocambinho (Jaíba) in the state of Minas Gerais, Brazil. Total DNA of tubers, stems and leaves was extracted and used as a template for nested-PCR reacions. Specific primers were designed to facilitate the detection of the phytoplasma in the presence of endophytic Bacillus sp. Using these primers it was possible to detect the phytoplasma in 16.7% of the tubers, 66.7% of the stems and 73.3% of the leaves of symptomatic plants, but in none of the tubers, 45% of the stems and 30% of the leaves of asymptomatic plants. A total of 37 cassava plants showed positive results for presence of phytoplasma. This phytoplasma was identified as belonging to the group 16SrIII, subgroup A, by in vitro and in silico RFLP analyses and sequencing. Phylogenetic analysis, based on 16S rDNA, supports the characterization performed by RFLP.
