Bioprospecção de leveduras killer com potencial para aplicação em biotipagem de microorganismos patogênicos humanos

Uma das formas de se realizar estudos epidemiológicos de dispersão de patógenos é pela discriminação intra-específica (biotipagem) desses microrganismos em subgrupos (biotipos). Métodos fenotípicos que permitem a discriminação de linhagens clínicas são almejados principalmente pela facilidade de execução e por terem menor custo, quando comparados com os métodos moleculares. O sistema killer, baseado em padrões de sensibilidade à toxina, pode representar uma ferramenta simples, barata, sensível e reprodutível para biotipagem de microrganismos de importância clínica. Neste estudo, foi pesquisada a presença deste fenótipo em 595 isolados de alimentos e de fontes ambientais, de onde foram selecionadas 32 cepas para testes de identificação molecular, caracterização genética do fenótipo killer, caracterização morfológica do mecanismo de ação da toxina e aplicação em painéis de biotipagem.A linhagem KYQU89 (CBS10423), proveniente de queijo, foi detectada como uma espécie nova, com seqüência idêntica a dois isolados killer oriundos de insetos. As três linhagens demonstraram ser relacionadas ao clado Ovoides no gênero Trichosporon, sendo caracterizadas por sequenciamento da região D1/D2 do rDNA , da região interespaçadora ITS e perfil bioquímico. A espécie nova foi denominada Trichosporon insectorum. Vinte linhagens killer foram selecionadas para compor um painel de biotipagem sobre cem isolados clínicos e ambientais de Cryptococcus neoformans e Cryptococcus gattii. A partir dos padrões de sensibilidade à toxina, foi gerado um dendrogramademonstrando a total discriminação das linhagens. A ação letal da toxina sobre as células destas duas espécies de Cryptococcus foi observada por microscopia eletrônica de varredura e óptica, evidenciando que não houve a formação de poros na parede celular, mas sim uma provável interferência no ciclo celular da célula. Da mesma forma, um painel de 11 linhagens killer selecionadas foi utilizado para a biotipagem de cepas multi-resistentes de Staphylococcus epidermidis, provenientes de dois hospitais em Porto Alegre, RS, também sendo capaz de discriminá-las totalmente. / One of the forms of performing epidemiological studies of dispersion of pathogenic microorganisms is by means of intra-specific discrimination (biotyping) of these microorganisms in sub-groups (biotype). Phenotypic methods that allow the clinical discrimination of strains are needed, especially because they are easy to perform and have low cost, when compared to molecular methods. The killer system, based on patterns of sensitivity towards the toxin, can represent a simple, cheap, sensible and reproductive tool for biotyping of microorganisms of clinical importance. In our study, the presence of this phenotype was evaluated in 595 isolates from natural environments and food and 32 killer strains were selected for molecular identification, genetic characterization of killer phenotype, morphologic characterization of the mechanism of action, and application in biotyping panels.The strain KYQU89 (CBS10423), isolated from cheese, was found to be a new species, with D1/D2 sequence identical to two killer isolates from insects. The three strains were shown to be related to the Ovoides clade of the genus Trichosporon, and were characterized bysequencing of the D1/D2 region of the LSU rDNA and physiological profiles the new species was called Trichosporon insectorum. The lethal action of the toxin on the cells of Cryptococcus was observed by means of optical and scanning electron microscopy, showing that there was no formation of pores on the cell surface of the sensitive cells in contact with the toxins, but a probable interference in the cell cycle. In the same way, a panel of 11 selected killer strains was used for the biotyping of multi-resistant Staphylococcus epidermidis strains, provenient from two hospitals in Porto Alegre, RS, being also capable of discriminating all the strains.
