This thesis work is aimed at the meaning of excursion in teaching biology at secondary vocational school for thematic complex of microbiology. The topic of microbiology is not attractive for students of secondary vocational school and it is very important to find out what the impact of the excursion will be on the knowledge of students who will participate on the excursion Part of this work is evaluation of excursion as an alternative method of teaching and its overall importance in the teaching of biology. Diploma thesis work brings an overview how the individual stages of the excursion should theoretically look like. It provides list of institutions and factories which may be asked for microbiology excursion in practice. Next part includes completely described excursion in brewery U Fleků including preparation realization and evaluation. Excursion was taken by two classes of different fields with identical pre concept of their studies. Part of the school phase of the excursion is testing students in the form of pre test with the knowledge of microbiology theme, particularly beer production and yeasting process. Předložená dizertační práce s názvem “Rastrovací elektronová mikroskopie a její aplikace pro senzitivní vzorky” pojednává o problematice rastrovací elektronové mikroskopie v kontextu instrumentálního a metodologického vývoje vedoucího k inovativnímu řešení, které je dobře aplikovatelné zejména v mikrobiologickém výzkumu. Součástí práce je rozprava o historii a současném stavu elektronové mikroskopie (EM) jakožto vědecké zobrazovací a analytické techniky, tato část se nachází v úvodních kapitolách. Nepopiratelný přínos EM v biologických a lékařských oborech je dokazován mnoha citovanými vědeckými publikacemi. Předložená dizertační práce přináší novinky z oblasti přípravy preparátů a kryogenní rastrovací elektronové mikroskopie (cryo-SEM) vyvinuté na pracovišti Ústavu přístrojové techniky AV ČR, v.v.i. v Brně. Jedná se především o návrhy a výrobu speciálních držáků vzorků a vývoj nových metodik v oblasti přípravy mikrobiologických preparátů vedoucích k nalezení optimálních parametrů jednotlivých procesů. V experimentální části se nachází ověření metodologických postupů při studiu hydratovaných a na elektronový svazek senzitivních preparátů. Následné srovnání různých přístupů na definovaném biologickém systému z oblasti mikrobiologie přispívá k rozšíření interpretace doposud známých výsledků. Mezi zkoumanými mikrobiologickými kmeny byly biofilm-pozitivní bakterie Staphylococcus epidermidis a kvasinky jako Candida albicans a Candida parapsilosis, jež jsou považovány za klinicky významné, protože se podílejí na vzniku závažných infekcí zejména u imunokompromitovaných pacientů. Dále byl studován vliv růstu biofilmu bakterie Bacillus subtilis na biodeteriorizaci a biodegradaci poly--kaprolaktonových fólií. Vývoj v oblasti cryo-SEM byl aplikován ve výzkumu mikrobů s biotechnologickým potenciálem, jako jsou např. Cupriavidus necator a Sporobolomyces shibatanus.

The thesis contains studying material – textbook for the practice in food microbiology for the students of the secondary technical schools of chemistry and food science. Verification of practical utility and didactical advisability of submited exercices was realized at The Secondary Technical School of Chemistry in Brno.

The diploma thesis evaluates the microbiological contamination of meat and background in the company of Jatky Bučovice. Numbers of the choosen microorganisms are weighted against the change of the season.

The aim of a presented diploma thesis was to evaluate the level of contamination in fish by nontuberculous mycobacteria (NTM). Methods of molecular biology were used to monitor the incidence and distribution of NTM in tissues of freshwater fish (gills, pharyngeal teeth, muscles, intestine etc.). NTM frequently occur in aquacultures for commercial use. With this in mind the occurrence of NTM was monitored in fish meat (Ctenopharyngodon idella, Cyprinus carpio, Tinca tinca, Sander lucioperca, Oreochromis niloticus etc.) from the market, because processing and consumption of insufficiently heat treated meat contaminated with NTM may pose health risk to humans. The theoretical part deals with the description of mycobacteria, their clinical significance and responsibilities in food safety. There are summarised methods for detection and identification of individual mycobacterial species. The experimental part is devoted to the description of used methods followed by evaluation of experimentally obtained data. The plasmid DNA was prepared as positive controls for PCR reaction. Furthermore, the analytical sensitivity of PCR reaction and efficiency of isolation of mycobacterial DNA isolation from fish meat were determined. Lastly, the level of NTM contamination in fish was evaluated.
