Investigating reorganization of the motor cortices following stem cell therapy in a non-human primate model of cortical ischemiaArndt, Kevin Richard 22 January 2016 (has links)
Prior work demonstrated that impairments in fine motor function produced by a controlled ischemic lesion in monkey primary motor cortex were ameliorated by treatment with a cell drug product, CNTO 0007. This drug contains human umbilical tissue-derived cells, in a proprietary thaw and inject formulation. Brain tissue sections from subjects with and without CNTO 0007 therapy were processed immunohistochemically to identify neurons expressing c-Fos as a marker for neuronal activity. Neurons expressing c-Fos were quantified using unbiased stereology. The number of c-Fos positive neurons in dorsal pre-motor cortex ipsilateral to the lesion were greater in treated animals but only approached statistical significance. These findings suggest that cortical reorganization in the dorsal pre-motor cortex may underlie the observed functional recovery. However, c-Fos expressing neurons in other motor areas, such as the ventral pre-motor cortex, remain to be studied.

Perineuronal nets in the cortical white matter – visualized with WFA (Wisteria floribunda agglutinin) in adult macaque monkeysZhang, Amy 20 June 2016 (has links)
PURPOSE: To characterize the distribution of white matter neurons (WMNs) positive for perineuronal nets (PNNs) in the adult monkey. WMNs are a mixed population of excitatory and inhibitory neurons. They have an important role in axon guidance during cortical development, but their role in the adult brain is less understood. In vitro and in vivo experiments provide evidence that WMNs are incorporated into cortical circuitry. The majority of investigations in the adult, however, have focused on regional variations in overall density, or on characterization of morphological and neurochemical subtypes. The present study was motivated by the observation that some WMNs exhibit PNNs in adult monkey. Since PNNs are associated with plasticity in younger animals, their occurrence with some WMNs might be functionally significant.
METHODS: PNNs were visualized, at the light microscopic level, by WFA staining in three adult macaque brains. Density of WFA positive WMNs was scored at three anterior-posterior levels (frontal, mid-hemispheric, and occipital), and compared with overall density of WMNs, as visualized by immunocytochemistry for NeuN. Quantitation of WFA+ neurons and neuron morphology were analyzed via light microscopy. Soma size and appearance, and dendritic length were recorded and measured.
RESULTS: On the basis of soma size and proximal dendritic shape, several types of WFA+ WMNs were provisionally identified, consistent with previous reports in the literature. Subpopulation densities were of highest density in mid-cortical areas and lowest quantities at occipital, matching previous studies. Morphological measurements suggested a heterogeneous neuron population through soma measurements and dendrite orientation. Soma sizes exhibited a range of circularity and size (10 µm – 30 µm). Dendrites were stained beyond the “proximal” area, including intermediate areas beyond the first branch, and up to 500 µm.
CONCLUSIONS: A small population of WMNs are coated by PNNs in adult monkey. On the basis of morphology, these might be further subdivided, but combined studies with other markers would be needed. Future studies might investigate age- or pathology-related changes in the density and subtypes of WMNs that express PNNs in human or nonhuman primates. We speculate that these WMNs might have functional specializations, perhaps similar to the plasticity effects documented for PNNs in early development.

Este fato, somado à expansão de áreas urbanas não consolidadas, muitas vezes para dentro dos limites de Unidades de Conservação Florestal, permitem um contato permanente e crescente entre os seres humanos e a fauna selvagem, como os macacos-prego. O comportamento imperativo destes animais e a escassez de alimentos em seu habitat peculiar fazem com que estes sejam induzidos pela busca de alimentos, que pode resultar em agressão direta aos visitantes de Parques, tais como mordeduras e arranhões em humanos, tornando-os potenciais disseminadores de micro-organismos patogênicos. O objetivo deste estudo foi caracterizar fenotipicamente micro-organismos isolados da cavidade bucal de 10 primatas Sapajus libidinosus capturados no Parque Ambiental Onofre Quinan, Anápolis-GO. Foram isoladas 111 amostras bacterianas e 12 fúngicas, sendo duas bactérias anaeróbias estritas do gênero Peptostreptococcus, 109 bactérias anaeróbias facultativas e 12 leveduras. Dentre as facultativas, destacaram-se as enterobactérias e os Staphylococcus. Quanto ao teste de antibiograma, foi detectada resistência de enterobactérias aos antimicrobianos tetraciclina e ampicilina, e de Staphylococcus à tetraciclina, eritromicina e clindamicina. As demais cepas apresentaram-se sensíveis aos antimicrobianos testados. A cefoxitina apresentou 100% de eficácia para todas as bactérias.Em casos de mordeduras por macacos-pregos recomenda-se realizar completa higienização nas lesões, as quais devem permanecer abertas, para que possam cicatrizar por segunda intenção. A antibioticoterapia deve ser realizada de acordo com recomendações médicas. / The State of Goiás has several areas of urban forest fragments. A total of 111 bacterial and 12 fungal samples were isolated, two strict anaerobic bacteria of the genus Peptostreptococcus, 109 facultative anaerobic bacteria and 12 yeasts. Among the facultative ones, enterobacteria and Staphylococcus were highlighted. As for the antibiogram test, resistance of enterobacteria to tetracycline and ampicillin antibiotics was detected, and from Staphylococcus to tetracycline, erythromycin and clindamycin. 5-hydroxytryptamine and sexual behaviour in rhesus monkeysGradwell, Peter Bertram January 1976 (has links)
Selective inhibition of 5-hydroxytryptamine by parachlorophenylalanine (PCPA) is able to restore sexual receptivity in female rhesus monkeys made unreceptive by bilateral adrenalectomy. PCPA in the doses used reduces the levels of 5-hydroxyindoleacetic acid (5HIAA) in the cerebrospinal fluid to 40 per cent of the normal oestradioltreated condition. Both the increased sexual receptivity and the lowered 5HIAA levels "in the CSF are in turn reversed by 5-hydroxytryptophal (5HTP), the irrmediate precursor of 5HT and the substance whose synthesis is inhibited by PCPA. 5HTP on its own reduces sexual receptivity and increases 5HIAA levels in the CSF of ovariectomised, oestradiol-treated (but otherwise intact) female rhesus monkeys. A substance other than an adrenal androgen has therefore been shown to restore sexual receptivity in adrenalectomised female monkeys . Testosterone propionate and oestradiol benzoate both lower the turnover rates of 5HT in the brains of ovariectomised female monkeys, as measured by the 2 hour probenecid test. Taken together, these findings suggest that adrenal androgens could act on specific sites in the female monkey brain via 5HT-containing neural systems, to control (or at least influence) sexual receptivity. All the results of administering oestradiol to ovariectomised monkeys in these experiments are consistent with the established roles of this hormone in female sexual attractiveness and in the gonadotrophin- controlling systems of the hypothalamo-hypophyseal axis. In contrast to these findings on 5HT and sexual receptivity in female monkeys, no clear role for 5HT- containing neural systems could be demonstrated in the grooming, aggressive or social behaviours of female monkeys. No clear role for 5HT could be demonstrated in the refractory period following ejaculation in male monkeys , or when testosterone replacement is given to castrated male monkeys.
